Abstract

A method of preparing a dna transfer vector with a nucleotide sequence for a specific protein, which method comprises: a) isolating cells containing mrnas that encode the protein from an organism that produces the protein. B) extracting the mrna from the cells in the presence of an rnase inhibitor composition, thereby preventing degradation of the rnase mrna; c) separating the mrna essentially free of protein, dna and other rna; d) synthesizing a double helix cdna, in which a strand contains a nucleotide sequence complementary to the mrna, thus producing a cdna with a nucleotide sequence encoding the desired protein; e) providing a dna transfer vector with reactive ends capable of being linked together or of being joined with double-stranded cdna, f) attaching the transfer vector of the adb with the double-stranded cdna containing a nucleotide sequence that it encodes insulin, thereby producing a dna transfer vector that contains a nucleotide sequence that encodes the desired protein.
