Abstract
Description
Nanopartículas de ácido colomínico y derivados.Nanoparticles of colominic acid and derivatives.
The present invention relates to systems nanoparticles that can be used for the administration of active ingredients, the compositions of said systems and a procedure for its elaboration.
Food and Drug Administration. Nanotechnology, A Report of the U.S. Food and Drug Administration Nanotechnology Task Forcé, FDA Ed., Rockville, MD, July 2007), (OMS, Initiative for Vaccine Research of the Department of Immunization, Vaccines and Biologicals, WHO/IVB/06.03, WHO Ed., Geneva, Switzerland, April 2006). Aguado, Advanced Drug Delivery Reviews, 57, 2005, 325-31); (T.G. Park, J.H. Jeong, S.W. Kim, Advanced Drug Delivery Reviews, 58, 2006, 467-486).The nanoparticular systems have enormous potential clearly recognized in numerous fields, having aroused great interest especially in the biomedical field (US Food and Drug Administration. Nanotechnology, A Report of the US Food and Drug Administration Nanotechnology Task Forcé, FDA Ed. , Rockville, MD, July 2007), (WHO, Initiative for Vaccine Research of the Department of Immunization, Vaccines and Biologicals, WHO / IVB / 06.03 , WHO Ed., Geneva, Switzerland, April 2006 ). In spite of this, the nanoparticular systems developed to date have a series of limitations that lead to the general idea that the development of new systems that are capable of meeting the challenge of taking advantage of their recognized potential is necessary (M. Friede and MT Aguado, Advanced Drug Delivery Reviews, 57, 2005, 325-31); (TG Park, JH Jeong, SW Kim, Advanced Drug Delivery Reviews, 58, 2006, 467-486).
Las causas de dichas limitaciones son diversas. The causes of these limitations are diverse.
Food and Drug Administration. Nanotechnology, A Report of the U.S. Food and Drug Administration Nanotechnology Task Forcé, FDA Ed., Rockville, MD, July 2007).Considering the specific case of Chitosan-based nanoparticles, polymer widely cited in literature as indispensable for their formation by ionic crosslinking, cytotoxicity has recently been reported associated with said chitosan nanoparticles, which has been related directly with the surface electric charge of these systems (B. Loretz and A. Bernkop-Schnürch, Nanotoxicology, 1, 2007, 139-148). This type of results of toxicity of particular concern to regulatory agencies such as FDA, which believes it is important not to lose sight of aspects such as significant positive charge associated with some systems nanoparticles (U.S. Food and Drug Administration. Nanotechnology, A Report of the U.S. Food and Drug Administration Nanotechnology Task Forcé, FDA Ed., Rockville, MD, July 2007).
On the other hand, in the case of nanosystems destined for systemic administration, the toxicity problems and / or adverse or unwanted effects associated with the surface load or the characteristics of the materials used so far in its development, take on special importance. In fact, although a system with a positive net charge may be of great interest as a topical administration vehicle, that positive charge can also be a problem when these systems arrive at a systemic level or if they are directly administered systemically. which will undoubtedly lead to hemagglutination and other adverse effects related to its interaction with natural components of the organism (Kainthan et al ., Biomaterials 27, 2006, 5377-5390).
Food and Drug Administration, FDA Consumer magazine, FDA Ed., November-December 2005 Issue, 2005).Considerations such as those described above have recently led to suggest the interest of investigating the application of nanotechnologies to new materials and develop, of this way, new nanoparticular systems (U.S. Food and Drug Administration, FDA Consumer magazine, FDA Ed., November-December 2005 Issue, 2005).

Mol. Life Sci. 2000, 57, 1964-1969). The material used in the nanoparticular systems of the present invention is colominic acid and its derivatives. Colominic acid is a polymer belonging to the family of polysalic acids, natural polymers of bacterial origin. It is a linear polymer consisting of residues of N-acetylneuraminic acid (Neu5Ac; also known as sialic acid), a natural constituent of cells and tissues, linked by α-glycosidic bonds (2 → 8). Each N-acetylneuraminic acid residue has a carboxyl group, responsible for the negative charge of colominic acid. It is a material of undoubted interest in the pharmaceutical and cosmetic field, as it is biocompatible and biodegradable, non-immunogenic, whose degradation products are not toxic (Gregoriadis G et al . Cell. Mol. Life Sci. 2000, 57, 1964- 1969). On the other hand, polysalic acids are characterized by having, among other properties, a very long plasma half-life, so they have been proposed as an alternative to polyethylene glycol derivatives to prolong the residence time in the plasma of drugs and release systems of bioactive molecules, such as liposomes. In fact, in the patent "WO / 2008/033253 - Liposome complexes containing pharmaceutical agents and methods" its use is used to modify preformed liposomes on the surface. Finally, taking into account its structural characteristics, this material offers the possibility of its modification, for example the introduction of amino groups and consequent cationization.
For the preparation of nanoparticles of Colominic acid has used one of the techniques used in the preparation of nanoparticles, called ionic crosslinking, or also ionic or ionotropic gelation. In a traditional way this technique has been developed for charged molecules electrical which, in the presence of ionic crosslinking agents, they give rise to the formation of systems that, unlike the complex, are characterized by being matrix structures in the that the associated bioactive molecule is totally or partially trapped within the polymer matrix constituting the themselves and generated in the process of ionotropic crosslinking. This polymeric matrix is obtained as a result of ionic bonds inter- and intra-molecular between the crosslinker ionic and macromolecule with electric charge, which gels spontaneously in nanoparticular form. This mechanism of training provides, as an added value with respect to complexes, a protection of the bioactive molecule against the external environment that those cannot contribute equally. Consequently, it It is a smooth, fast, economical technique, easily reproducible and scalable and that requires a very technology simple, all aspects of undoubted interest to the industry.
In one mode, the present invention provides a optimized and adapted procedure that allows to prepare nanoparticles based on colominic acid or derivatives that form by incorporating a polyanionic molecule or polycationic or a mixture of these molecules, low weight molecular, capable of acting as crosslinkers.

The composition, size and surface load of The nanoparticles can be modulated by proper selection of the components that constitute them. Such modulation allows to develop very versatile systems, since those that have a positive charge have a favorable interaction with the biological surfaces of the organism, because these are found Last negatively charged. On the other hand, the nanoparticles of negative surface charge are especially suitable when they are intended to reach systemic level, since they will not give place to hemagglutination phenomena or other adverse effects related to its interaction with natural components of the organism but, on the contrary, benefit from the prolonged residence time in biological fluids such as the plasma described for the colominic

For this, aqueous solutions of colominic acid (0.1-1 mg / mL) and polyarginine (1-2 mg / mL) in milli-Q water. How Crosslinking agent tripolyphosphate (TPP) was used in aqueous solution (0.0625-1 mg / mL) (milli-Q water). He corresponding genetic material was incorporated in a proportion between 2.5-5% by weight with respect to the components previous. The bioactive molecule was associated with the dissolution of the colominic acid and tripolyphosphate crosslinker and solution resulting was mixed with the polyarginine solution, under magnetic stirring, which was maintained for half an hour, allowing the complete evolution of the system towards a form stable nanoparticular. The relationships between the various System components are shown in Table 1 Also, said Table 1 shows the average diameter and electric charge surface (zeta potential) of the systems obtained. Just like it can be seen, it has been possible to modulate the composition of the nanoparticles and their physicochemical properties. In this way, although nanoparticles made using only colominic have negative surface electric charge, the use of polyarginine in various proportions with respect to colominic allows modulating the value of said load, both in sign (systems of positive or negative charge) as in absolute value.
Nanoparticles were prepared from acid colominic and polyarginine according to the procedure previously described, containing genetic material in plasmid form, pEGFP, (2.5% load). Acid nanoparticles were prepared colominic, incorporating cationized jelly with spermine, according to The procedure previously described. The opposite charge of said polymers were used to modulate the final net electric charge of the systems made from mixtures thereof. Be proceeded to the association of a bioactive hydrophilic macromolecule in its composition, selecting genetic material for that purpose, specifically a plasmid, the pEGFP. It is a macromolecule negatively charged bioactive, so it was incorporated next to the colominic acid or the tripolyphosphate crosslinking agent, fillers also negative, to avoid the appearance of previous interactions to the formation of particles.
