Abstract
Procedimiento para la determinación de la actividad de las proteínas desacoplantes (UCPs) mediante la monitorización del consumo de NAD(P)H. La presente invención trata del diseño de un procedimiento para determinar la actividad de las UCPs y que puede ser utilizado para evaluar la capacidad de distintos fármacos para modificar la actividad de dichas proteínas. Estos fármacos podrán ser utilizados para tratar todas aquellas enfermedades o condiciones en las que se produzcan variaciones en su actividad termogénica. Ejemplos son la obesidad, fiebre, caquexia, etc. Debido a que la actividad de las UCPs produce variaciones en la velocidad de respiración, en el presente procedimiento se determina la actividad de las UCPs monitorizando la desaparición del NADH o NADPH según la disminución en la absorción entre 300-380 nm o la fluorescencia a 420-520 nm. En este procedimiento estos coenzimas son oxidados por las mitocondrias de la levadura Saccharomyces cerevisiae en las que se expresan distintasUCPs de modo recombinante.
