Abstract
Biosensor for the measurement of fructose, method for preparing it, and the application thereof for the analysis of fructose in food products. Carbon paste biosensor for the measurement of fructose based on amperometric electrodes for direct electronic transfer between the enzyme fructose dehydrogenase, FDH, in which inactivation of the enzyme is prevented by paraffin oil, adding an aqueous solution of polyethylene imine, PEI, to the graphite-enzyme mixture, and, once the mixture is dry, adding the paraffin to form the electrode. The enzyme FDH can be the commercial enzyme isolated from Gluconosbacter sp. or from any other natural biological source or one obtained by genetic engineering techniques. Detergents are added to the mixture of the enzyme with the graphite. The graphite used is subjected to heat treatment (which can be with air) for more than 1 minute at a temperature greater than 650 degree C. The preparation of the biosensor is carried out in the following stages: a) A solution of enzyme in a pH 5 buffer containing a surfactant is used as the starting material b) The mixture is dried at reduced pressure at 3-8 degree C c) An aqueous solution of PEI buffered at pH 5 is added to it and the mixture is dried at 3-8 degree C d) The resultant graphite powder is mixed with paraffin oil, thus obtaining the corresponding modified carbon paste.
