Abstract

Die Trinkkur bezweckt den Aufbau, die Förderung und den Erhalt des Darm Mikrobioms (Eubiose) und sie besteht aus einem Getränk mit mindestens den folgenden Komponenten: • Einem probiotischen Kräuterferment-Getränk (Probiotikum), • Einem Präbiotikum kombiniert mit essenziellen Vitaminen, Mineralien und Spurenelementen, • Einem natürlichen Speiseöl, das reich an Omega 3 Fettsäuren sowie Tocopherol haltig ist. Diese Komponenten sind zu einer Emulsion aufbereitet und das so erhaltene Getränk kann im Rahmen einer Trinkkur regelmässig, vorzugsweise täglich eingenommen werden.

Trinkkur-Getränk aus drei Komponenten für den Aufbau, die Förderung und den Erhalt der Darmqesundheit
[0001 ] Die Erfindung betrifft ein besonderes Getränk aus drei Komponenten, einzunehmen in einer Trinkkur, die vorzugweise täglich angewendet werden soll. Für das Verständnis der Motivation für diese besondere Komposition dieses Getränks und der biologischen und medizinischen Hintergründe zur Wahl der Komponenten, und für die Erläuterung dieser T rinkkur und ihrer Wirksamkeit werden zunächst einige wichtige Definitionen angegeben, wobei teilweise in eckigen Klammern auf wissenschaftliche Literatur verwiesen wird:
Mikrobiom: Im engeren Sinne bezeichnet das die Gesamtheit der mikrobiellen DNA im menschlichen Körper. Im weiteren und umgangssprachlichen Sinne bezeichnet der Begriff Mikrobiom die Gesamtheit aller Mikroorganismen im menschlichen Körper. Beim Menschen werden Mikrobiome je nach Körpernische (Darm, Haut, Mundhöhle, Vagina, usw.) unterschieden. Das Darm-Mikrobiom befindet sich im Lumen in der äusseren Mukusschicht des Darms.
Kommensale: Bezeichnet den Organismus, der zusammen mit einem anderen Organismus von der gleichen Nahrung lebt, ohne diesen zu schädigen.
Eubiose: Bezeichnet das Gleichgewicht der Bakterienkulturen im Mikrobiom des gesunden Menschen.
Dysbiose: Bezeichnet ein gestörtes Gleichgewicht der Bakterienkulturen im Mikrobiom, das Gegenteil der Eubiose.
Vitalität: Bezeichnet die körpereigene mentale, physische, emotionale und soziale Leistungsfähigkeit.
Probiotika: Bezeichnet lebende, nichtpathogene Mikroorganismen, die dem Wirt (hier konkret der Darm) einen gesundheitlichen Nutzen bringen [1 ]. Präbiotika: Bezeichnet Substrate, die selektiv von Wirtsmikroorganismen (hier konkret das Mikrobiom) genutzt werden und einen gesundheitlichen Nutzen bringen [2].
Synbiotika: Bezeichnet eine Mischung aus Probiotika und Präbiotika mit einem gesundheitlichen Nutzen für den Wirt (hier konkret für den Darm) [3].
[0002] Im Mikrobiom des menschlichen Darms sind rund 100 Billionen (1014) Bakterien angesiedelt. Kein anderer Lebensraum auf der Erde ist dichter besiedelt. Im Dickdarm sind es 1010 - 1012 Bakterien pro Gramm Biomasse. Die Bakterien setzen sich zu >90% aus den beiden anaeroben Phyla der Bacillota (>75%) und Bacteriodota (>15%) zusammen und bilden etwa 10’000 verschiedene kommensale Arten. Im Dünndarm sind hauptsächlich die Arten Lactobacillus und Enterococcus, und im Dickdarm ist hauptsächlich die Art Bifidobacterium neben E. Coli., Enterobacterium und Clostridium, angesiedelt. Diese komplexe Bakterienvielfalt macht den Menschen zu einem Superorganismus, welcher rund 300 mal mehr Gene als eine menschliche Zelle besitzt, und ein rund 100 mal höheres metabolisches Potential als die menschliche Leber hervorbringt [4].
[0003] Das Darm-Mikrobiom gilt als (endokrines) ‘Superorgan’ [5] oder ‘zweites Gehirn’ [6]. Es ist ein komplexes mikrobielles Ökosystem, in dem die Bakterien miteinander leben, kommunizieren, Stoffwechselprodukte austauschen und rekombinieren. Und diese komplexe Bakterien-Darm Wechselwirkung wird stark von der Ernährung beeinflusst. Je nach Zusammensetzung der Nahrung und der bakteriellen Zusammensetzung werden im Mikrobiom eine Vielzahl verschiedener physiologisch aktiver Metaboliten geformt.
[0004] Voraussetzung für eine gesunde Vitalität ist Gleichgewicht (Eubiose) der Bakterienkulturen im Mikrobiom. Dazu braucht es eine Versorgung des Mikrobioms mit einer ausgewogenen Mischung der Makronährstoffe Kohlenhydrate, Proteine und Fette [7]. Im Mikrobiom werden die unverdaulichen Nährstoffe wie komplexe Glucane und Glucokonjugate der pflanzlichen Zellwände (umgangssprachlich Ballaststoffe oder Fasern genannt), Proteine sowie Glycin, Taurin, Gallensäure und andere Konjugate, das heisst Produkte, die durch die kovalente Kopplung von mindestens 2 Molekülen entstanden sind, metabolisiert. Die anaerobe saccharolytische Fermentation der Ballaststoffe führt über die Brenztraubensäure zu ‘kurzkettigen Fettsäuren’ (engl. short chain fatty acid (SCFA)), das heisst Essigsäure, Propionsäure und Buttersäure.
[0005] Die Änderung der bakteriellen Zusammensetzung des Mikrobioms durch Störungen beeinträchtigt wesentlich die Vitalität. Über 90% aller Krankheiten und Beschwerden sind auf eine Dysbiose zurückzuführen. Prominente Beispiele sind chronische Darmentzündungen, Fettleibigkeit, Diabetes, Akne, Atherosklerose, Asthma, neurodegenerative Erkrankungen und bestimmte Arten von Krebs [8]. Eine Dysbiose entsteht durch mangelhafte Zufuhr von Nährstoffen, bedingt durch eine fehlerhafte Ernährung, durch die Einnahme von Medikamenten (vor allem Antibiotika) und durch chronischen Stress.
[0006] Bekannte Metaboliten des Darm-Mikrobioms sind beispielsweise Serotonin, kurzkettige Fettsäuren, diverse B- Vitamine (B1 , B2, B5, B7, B9, B12), Vitamin K, sekundäre Gallensäuren und die verzweigten Aminosäuren. Funktional bewirken diese Metaboliten eine Verbesserung des Nährstoffabbaus und regulieren Herz- Kreislauf-, Immun- und Nervensystem über die Achsen von Darm-Hirn [9], Darm- Lunge [10], Darm-Herz [1 1 ] und Darm-Haut [12].
[0007] Kohlenhydrate, Proteine und Fette machen etwa 90 % des Trockengewichtes der Nahrung und 100 % ihrer Energie aus.
[0008] Verschiedene Milchsäurebakterienstämme der Gattung Bifidobacterium haben auch eine entgiftende Wirkung. Milchsäurebaktieren aus dem Stamm Enterococcus faecalis können sich auch entgiftend und gesundheitsfördernd auswirken. Sie bewirken den Oxalat-Abbau und beugen damit der Bildung von Nierensteinen vor [13].
[0009] Bakterien der Gattung Lactobacillus sind heterofermentative Milchsäurebakterien und fermentieren Ballaststoffe über den Phosphoketolaseweg zu Essigsäure und Milchsäure. Sie sind die im Mikrobiom am häufigsten anzutreffende Bakteriengattung [14].
[0010] Es ist bekannt [15], dass die im Mikrobiom vorhandenen Bakterien der Gattung Bifidobacterium die grösste Vielfalt an Ballaststoffen fermentieren. Aus der Fermentierung der Ballaststoffe werden die Hexosen zu Essigsäure und Milchsäure im Verhältnis 3:2 synthetisiert. Die Essigsäure ist eine der ‘kurzkettigen Fettsäuren’ und an der Regulierung des Stoffwechsels sowie des Immun- und Nervensystems beteiligt.
[0011 ] Milchsäure dient zur Energiegewinnung (Gluconeogenese) und hat zahlreiche vaskuläre Wirkungen. Die kurzkettigen Fettsäuren dienen den Epithelzellen des Darms als Energiequelle und sind wichtige Signalmoleküle. Sie binden an die Rezeptoren GPR41 und GPR43 auf den Epithel- und Immunzellen. Die Rezeptoren regulieren die Ausschüttung pro-inflammatorischer Cytokine, welche über GLP1 (Glukagon-Iike Peptide) und PYY (Peptide-Tyrosin-Tyrosin) direkt auf das Zentralnervensystem wirken. Desweiteren inhibieren kurzkettige Fettsäuren die Histon Deacetylase (HDAC) in Immunzellen und Adipozyten und wirken so auf die Transkription dieser Zellen. Auf diese Weise wird der Stoffwechsel sowie das Immun- und Nervensystem reguliert.
[0012] Die kurzkettigen Fettsäuren verteilen sich mit Hilfe von Transporterproteinen im gesamten menschlichen Gewebe inklusive im Gehirns [16]. Dort regulieren diese den Stoffwechsel sowie das Immun- und Nervensystem. Zusätzlich wird der pH-Wertes auf 5.5 abgesenkt und verhindert so die Vermehrung pathogener Darmbakterien.
[0013] Aus den Gallensäure Konjugaten metabolisieren Enzyme der Darmbakterien die sekundären Gallensäuren. Diese emulgieren die Fette aus der Nahrung und ermöglichen so den enzymatischen Fettabbau durch Lipasen.
[0014] Aus den Proteinen hydrolysieren Enzyme der Darmbakterien kurzkettige Peptide und einzelne Aminosäuren. Diese werden zum Aufbau und Regeneration der Darmschleimhäute genutzt und gelangen über die Blutbahn zu den Zellen, wo sie der Proteinbiosynthese zugeführt werden.
[0015] Die Häufigkeit von Krankheiten, die auf Beeinträchtigung des Herz-Kreislauf, Immun- und Nervensystem beruhen, hat in der westlichen Zivilisation zugenommen. Der Anstieg korreliert mit dem zurückgehenden Verbrauch an Ballaststoffen. Es besteht ein kausaler Zusammenhang zwischen der Ernährung, der Aufnahme von Ballaststoffen und den daraus durch Fermentation gebildeten Stoffwechselprodukten, wie beispielsweise den kurzkettigen Fettsäuren.
[0016] Die Ernährung und die daraus resultierende Änderung der Zusammensetzung des Mikrobioms hat damit einen direkten Zusammenhang mit westlichen Zivilisationskrankheiten wie Verdauungsproblemen, Übergewicht, Aufmerksamkeitsdefizite, Konzentrations- und Schlafstörungen. Eine Vorbeugung oder Korrektur durch spezifisch abgestimmte probiotische Nahrungszusätze ist möglich.
[0017] Die Einnahme von Präbiotika beeinflusst das relative Wachstum der Bakterienstämme und ändert damit die bakterielle Zusammensetzung des Mikrobioms und damit die Zusammensetzung der physiologisch wirksamen Stoffwechselprodukte.
[0018] Eingedenk all dieser wissenschaftlichen Erkenntnisse ist es die Aufgabe der vorliegenden Erfindung, ein Getränk für den Aufbau, die Förderung und den Erhalt der Darmgesundheit anzugeben, das im Rahmen einer Trinkkur täglich einnehmbar ist und besonders wirksam und einfach in der Anwendung ist. Dabei sind gewählten Inhaltsstoffe des Getränks von entscheidender Bedeutung.
[0019] Die Lösung dieser Aufgabe besteht in einem Getränk für den Aufbau, die Förderung und den Erhalt des Darm Mikrobioms (Eubiose), bestehend aus mindestens den folgenden Komponenten:
• Einem Probiotikum, mit in der Grössenordnung 30 Milliarden lebenden Milchsäurebakterien pro 10 mL, und mindestens 10 natürliche Kräuter und Gewürze, die durch Fermentation konserviert wurden, indem die Kräuter und Bakterien mit Wasser und Zuckerrohrmelasse gemischt wurden, wodurch die Bakterien den Zucker in der Melasse verbrauchten und ihn in Milch- und Essigsäure umwandelten,
• Einem Präbiotikum kombiniert mit essenziellen Vitaminen, Mineralien und Spurenelementen,
• Einem natürlichen Speiseöl, enthaltend mehr als 50 Gew-% Omega 3 Fettsäuren sowie Tocopherol, indem diese Komponenten zu einer Emulsion als Getränk aufbereitet vorliegt und im Rahmen einer Trinkkur vorzugsweise täglich verabreichbar ist.
[0020] Die tägliche Einnahme dieses Getränks in Rahmen einer Trinkkur stärkt den Aufbau, die Förderung und den Erhalt der Darmgesundheit. Die Inhaltsstoffe der ersten Komponente enthalten rund 30 Milliarden lebende Milchsäurebakterien pro 10 mL und über 10 verschiedene natürliche Kräuter und Gewürze. Je nach Bedarf kann das Probiotikum auch Bakterien vom Stamm Enterococcus faecium enthalten, sowie einen oder mehrere Stämme der Gattungen Lactobacillus und Bifidobacterium, zum Beispiel diese, welche alle aus Dänemark stammen:

Das Produkt wird auf natürliche Weise durch Fermentation konserviert, bei dem die Kräuter und Bakterien mit Wasser und Zuckerrohrmelasse gemischt werden. Die Bakterien verbrauchen den Zucker in der Melasse und wandeln ihn in Milch- und kurzkettige Fettsäuren um.
[0021] Die Inhaltsstoffe der zweiten Komponente umfassen >10 pflanzliche Ballaststoffe als präbiotische Substrate und >30 verschiedene Mineralien, Vitamine und Spurenelemente. Die Hauptkomponente mit >25 Gew.-% enthält pflanzliche Ballaststoffe bestehend aus komplexen Glycanen, wie Glucomannanen, Arabinogalactanen, Xyloglucanen, Galacturonanen, Fructooligosacchariden oder Galactomannanen. Galactomannane bestehen aus ß-1 ,4 verknüpfter Mannose als Hauptkette, die partiell mit a-1 ,6 Galactoseresten verknüpft ist. Beispielsweise enthält Guarkernmehl dieses Glycan. [0022] Klinische Studien haben gezeigt, dass mit teilweise hydrolysiertem Guarkernmehl angereicherte Nahrungsergänzungsmittel Vorteile und klinische Effekte bei der Reduktion von Diarrhö, bei der Therapie des Reizdarmsyndroms und bei Obstipation (Verstopfung) erreicht werden [17]. Ausserdem wird die Verfügbarkeit von Fett und Cholesterin für die Resorption deutlich vermindert und das Risiko von Gefässerkrankungen reduziert [18].
[0023] Die in der zweiten Komponente enthaltenen präbiotischen Substrate werden von den saccharolytisch fermentierenden Bakterien im Mikrobiom zu Milchsäure und den kurzkettigen Fettsäuren Essigsäure, Propionsäure und Buttersäure metabolisiert.
[0024] Zweck der zweiten Komponente ist es, dem Mikrobiom Substrate zuzuführen, welche als Vorläufer für die bakterielle Synthese kurzkettiger Fettsäuren dienen. Substrate, die durch die bakterielle Synthese zu giftigen oder schädlichen Metaboliten führen, werden in der zweiten Komponente bewusst weggelassen.
[0025] Die dritte Komponente ist ein an Omega 3 hochangereichertes natürliches Speiseöl, beispielsweise Leinöl oder das Öl der Sacha-Inchi-Nuss. Es enthält zu >50% Omega 3 Fettsäuren sowie bis zu 50 mg/100 mL an Tocopherol. Omega 3 Fettsäuren fördern das Wachstum der Buttersäure, eines der kurzkettigen Fettsäuren produzierenden Bakterien im Mikrobiom. Auf diese Weise beugen sie neurodegenerativer Erkrankungen vor und schützen vor Entzündungen, Gefässverkalkung und einem Herzinfarkt [19].
[0026] Studien zeigen, dass Änderungen in der Mikrobiom Zusammensetzung mit dem Auftreten von sogenannten Life-Style Erkrankungen kausal korreliert. Life-Style Erkrankungen sind Krankheiten, die mit der Art und Weise der Lebensführung Zusammenhängen. Dazu gehören vorher nicht gekannte Inzidenzen von Stoffwechselerkrankungen wie Adipositas und Diabetes, entzündliche Darmerkrankungen, Asthma, Allergien und Immundefekte und neurogenerative Erkrankungen [20].
[0027] Bei diesen Erkrankungen wird eine deutliche Abnahme der bakteriellen Diversität und der Verlust bestimmter metabolischer Funktionen festgestellt. Die Abwesenheit bestimmter Bakterien oder die Dysbiose ist Ursache für das Auftreten dieser komplexen Erkrankungen [21].
[0028] Die täglich immer zeitgleiche, ritualisierte Einnahme der Trinkkur auf der Basis des vorgestellten Getränks mit seinen drei kardinalen Komponenten hat den Zweck, dass die zugeführten Stoffe mit den täglichen zirkadianischen Schwankungen in Zusammensetzung und Funktion des Mikrobioms zusammenpasst [22].
[0029] Die gleichzeitige Einnahme der drei Komponenten in einer täglichen, ritualisierten Trinkkur kombiniert pro- und präbiotische Nahrungs- und Nahrungsergänzungsmittel und führt damit zu einem einzigartigen synbiotischen Präparat.

Claims

Patentansprüche
1. Getränk für den Aufbau, die Förderung und den Erhalt des Darm Mikrobioms (Eubiose), bestehend aus mindestens den folgenden Komponenten:
• Einem Probiotikum, das in der Grössenordnung 30 Milliarden lebende Milchsäurebakterien pro 10 mL enthält, und mindestens 10 natürliche Kräuter und Gewürze, die durch Fermentation konserviert wurden, indem die Kräuter und Bakterien mit Wasser und Zuckerrohrmelasse gemischt wurden, wodurch die Bakterien den Zucker in der Melasse verbrauchten und ihn in Milch- und Essigsäure umwandelten,
• Einem Präbiotikum kombiniert mit essenziellen Vitaminen, Mineralien und Spurenelementen,
• Einem natürlichen Speiseöl, enthaltend mehr als 50 Gew-% Omega 3 Fettsäuren sowie Tocopherol, indem diese Komponenten zu einer Emulsion als Getränk aufbereitet vorliegt und im Rahmen einer Trinkkur vorzugsweise täglich verabreichbar ist.
2. Getränk nach Anspruch 1 , dadurch gekennzeichnet, dass die Inhaltsstoffe der ersten Komponente, des Probiotikums, zusätzlich lebende Bakterien vom Stamm Enterococcus faecium enthalten, sowie einen oder mehrere Stämme der Gattungen Lactobacillus und Bifidobacterium.
3. Getränk nach einem der Ansprüche 1 bis 2, dadurch gekennzeichnet, dass die Inhaltsstoffe der zweiten Komponente, des Präbiotikums, mehr als 10 pflanzliche Ballaststoffe als präbiotische Substrate und mehr als 30 verschiedene Mineralien, Vitamine und Spurenelemente umfassen, wobei die Hauptkomponente mit mehr als 25 Gew.-% pflanzliche Ballaststoffe aus komplexen Glycanen, wie Glucomannane, Arabinogalactane, Xyloglucane, Galacturonane, Fructooligosaccharide oder Galactomannane enthält, welches aus ß-1 ,4 verknüpfter Mannose als Hauptkette mit partiell verknüpften a-1 ,6 Galactoseresten besteht.

Getränk nach einem der Ansprüche 1 bis 2, dadurch gekennzeichnet, dass die Inhaltsstoffe der zweiten Komponente, des Präbiotikums, mehr als 10 pflanzliche Ballaststoffe als präbiotische Substrate und mehr als 30 verschiedene Mineralien, Vitamine und Spurenelemente umfassen, wobei die Hauptkomponente mit mehr als 25 Gew.-% Ballaststoffe aus der Familie der Glycane, nämlich Guarkernmehl, das solche Ballaststoffe enthält.
5. Getränk nach einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass die dritte Komponente ein an Omega 3 Fettsäuren hochangereichertes natürliches Speiseöl ist, ein Leinöl oder das Öl der Sacha-Inchi-Nuss, das zu mehr als 50% Omega 3 Fettsäuren sowie bis zu 50 mg/100 mL an Tocopherol enthält.
