La présente invention concerne des ARN interférents, en particulier des miARN, capables de bloquer spécifiquement la réplication d'une souche de virus du VIH résistante à un composé antirétro viral, et leur utilisation pour le traitement des infections par ce type de virus.

Description

Utilisation d'ARN interférents pour le traitement d'une infection par le VIH
La présente invention relève du domaine de la médecine, et plus particulièrement de celui du traitement des infections par le virus du VIH. Elle concerne notamment des ARN interférents capables de bloquer spécifiquement la réplication d'une souche de virus du VIH résistante à un composé antirétroviral et leur utilisation pour le traitement des infections par ce type de virus.
Arrière-plan technologique de l'invention
Le virus de l'immunodéficience humaine, ou VIH, est un rétrovirus du genre des lentivirus qui infecte l'homme et est responsable du syndrome d'immunodéfîcience acquise (SIDA). Bien qu'il existe des traitements antirétroviraux permettant de retarder l'apparition du SIDA, en revanche, aucun vaccin ou traitement définitif n'est disponible à l'heure actuelle.
Le VIH dispose d'une enveloppe composée en partie de la membrane de la cellule infectée dont il est issu et de deux types de glycoprotéines, à savoir gpl20 et gp41. À l'intérieur de l'enveloppe, se trouve une matrice protéique composée de protéines pl7 et renfermant la capside constituée de protéines p24 et la nucléocapside constituée de protéines p6 et p7. Le génome du VIH est un ARN simple brin qui est contenu dans la capside en présence de deux enzymes, la transcriptase inverse p64 nécessaire à la transcription de l'ARN viral en ADN, et l'intégrase p32 nécessaire à l'intégration de l'ADN viral dans l'ADN de la cellule infectée. Ces deux enzymes ainsi que la protéase plO qui participe à l'assemblage de la particule virale, sont spécifiques des rétrovirus et sont donc les principales cibles des traitements antirétroviraux.
Dans les cinq dernières années, l'arsenal des antirétroviraux s'est enrichi de neuf nouvelles molécules et de trois nouvelles classes d'inhibiteurs. Les antirétroviraux ont pour but d'interférer avec différents mécanismes : d'une part, les enzymes du VIH nécessaires à sa réplication et d'autre part, ses mécanismes d'entrée dans la cellule. Les protéines cibles de ces molécules sont donc essentiellement la transcriptase inverse, l'intégrase, la protéase et la glycoprotéine gp41. Parmi les antirétroviraux les plus récents, le raltégravir, premier représentant de la classe des inhibiteurs de l'intégrase (INI), a récemment obtenu son Autorisation de Mise sur le Marché. L'elvitégravir, deuxième médicament de cette classe, est actuellement en essai clinique. Les résultats des différents essais cliniques ayant évalué l'utilisation de ces deux molécules sont très encourageants en ce qui concerne le succès virologique, la tolérance et la réponse immuno logique. Cependant, comme pour les autres antirétroviraux, des résistances aux INI apparaissent dans les études in vitro et in vivo. Ces résistances sont caractérisées par la sélection de mutations sur le gène de l'intégrase du VIH qui ont un impact sur la sensibilité du virus aux INI. Les mutations mises en évidence chez les patients en échec thérapeutique sont principalement localisées dans le domaine catalytique central de l'enzyme situé entre les acides aminés 50 et 212.
Le phénomène de résistance du VIH aux antirétroviraux est un problème majeur dans la thérapeutique antirétro virale car il entraîne une diminution de l'efficacité des traitements et une augmentation de la mortalité des patients.
De précédents travaux ont démontré l'interaction entre des micro ARN (miARN) et des virus. Les miARN sont des ARN simple-brin longs d'environ 21 à 24 nucléotides. Il existe plusieurs centaines de gènes de microARN dans les génomes de la plupart des organismes pluricellulaires et, à ce jour, environ 500 miARN ont été identifiés chez l'humain. Les miARN sont des répresseurs post-transcriptionnels : en s'appariant à des ARN messagers, ils guident leur dégradation, ou la répression de leur traduction en protéine. Les gènes de miARN sont transcrits sous la forme de longs précurseurs appelés « pri-miARN ». Ces précurseurs sont clivés dans le noyau en un produit intermédiaire appelé « pre-miARN » par un complexe nommé Microprocesseur qui est formé chez les animaux par les enzymes Drosha et DGCR8 (Di George Critical Région 8 ou Pasha) et chez les plantes par une enzyme de la famille Dicer. Le pre-miARN est un ARN long d'environ 70 nucléotides, replié en tige-boucle imparfaite par complémentarité de bases entre la première moitié et la deuxième moitié de sa séquence. Ce pre-miARN est transporté du noyau au cytosol par transport actif GTP- dépendant grâce à une interaction avec l'exportine 5. Le pre-miARN est ensuite clivé dans le cytoplasme par une enzyme de la famille Dicer pour libérer un petit ARN double-brin appelé « miARN». Ce miARN double brin interagit alors avec une protéine de la famille Argonaute (Agol ou Ago2) pour former le complexe RISC (RNA- induced Silencing Complex). Ce complexe d'environ 16OkDa a été décrit comme étant suffisant à l'activité des miARN de répression de la traduction. Cependant d'autres protéines telles que la geminine peuvent également s'ajouter au complexe. Au cours de la formation du complexe RISC, le miARN double brin devient simple brin et seul le brin spécifique de l'ARNm cible est conservé. L'ARNm cible est alors chargé au sein du complexe RISC. A ce stade, deux voies sont alors possibles selon la composition du complexe. Dans le cas où le complexe contient la protéine Ago2, l'ARNm cible sera dégradé. Si le complexe contient la protéine Agol, la traduction de l'ARNm cible sera alors réprimée.
Il a été précédemment démontré que certains miARN des cellules hôtes étaient capables de cibler des ARN viraux et possédaient donc un rôle antiviral ou au contraire permettaient au virus de s'accumuler dans la cellule. Cette situation a notamment été décrite pour le miARN hsa-miR-122a spécifiquement exprimé dans le foie et favorisant la réplication de IARN du virus de l'hépatite B (Chen et al, 2007).
Résumé de l'invention
L'objectif de la présente invention est de fournir de nouveaux composés pouvant être utilisés dans le traitement d'une infection par une souche du virus VIH résistante à un composé antirétroviral.
Les inventeurs ont démontré l'existence de miARN capables de s'hybrider préférentiellement aux gènes de souches de VIH résistantes aux antirétroviraux, notamment aux inhibiteurs de l'intégrase ou de la transcriptase inverse. Les études présentées dans ce document montrent que ces miARN possèdent une activité antivirale et sont notamment capables de bloquer la réplication et la production virale du VIH.
Ainsi, la présente invention concerne un ARN interfèrent qui s 'hybride préférentiellement avec l'ARNm d'un gène codant pour une protéine mutante d'une souche du virus VIH, ladite protéine mutante contenant une ou plusieurs mutations conférant à la souche virale un phénotype de résistance à un composé antirétroviral, pour une utilisation dans le traitement d'une infection par une souche du virus VIH résistante audit composé antirétroviral. De préférence, le virus VIH est le virus VIH-I.
Selon un mode de réalisation, l'ARN interfèrent est choisi dans le groupe constitué d'un ARNsi, d'un ARNsh et d'un miARN. De préférence, l'ARN interfèrent est un miARN.
Selon un mode de réalisation, la protéine mutante est choisie dans le groupe constitué de la transcriptase inverse, la protéase, la glycoprotéine gp41 et l'intégrase.
Selon un mode de réalisation particulier, la protéine mutante est l'intégrase du virus VIH-I et le composé antirétroviral est un inhibiteur d'intégrase, de préférence le raltégravir ou l'elvitégravir. Selon un mode de réalisation, l'intégrase contient une ou plusieurs mutations conférant à la souche virale un phénotype de résistance audit inhibiteur d'intégrase, le ou les résidus mutés de l'intégrase étant choisis dans le groupe constitué du résidu T66, E92, F121, E138, G140, Y143, S147, Q148, S153, N155, E157 et R263 de la SEQ ID No. 1, et une combinaison de ceux-ci. Dans un autre aspect, la présente invention concerne également un ARN interfèrent qui s'hybride préférentiellement avec l'ARNm d'un gène codant pour une protéine mutante d'une souche du virus VIH, ladite protéine mutante contenant une ou plusieurs mutations conférant à la souche virale un phénotype de résistance à un composé antirétro viral, ou un vecteur d'expression comprenant une séquence nucléotidique codant pour ledit ARN interfèrent ou pour un précurseur de celui-ci, pour une utilisation en combinaison avec ledit composé antirétroviral dans le traitement d'une infection par le virus VIH. De préférence, le composé antirétroviral est un inhibiteur d'intégrase ou de transcriptase inverse.
Dans encore un autre aspect, la présente invention concerne une composition pharmaceutique comprenant au moins un ARN interfèrent selon l'invention et/ou un vecteur d'expression selon l'invention, et un ou plusieurs excipients et/ou véhicules pharmaceutiquement acceptables. De préférence, la composition pharmaceutique comprend au moins un miARN choisi dans le groupe constitué de hsa-miR-185* (SEQ ID No :4), hsa-miR-370 (SEQ ID No :6), hsa-miR-518c* (SEQ ID No :7), hsa-miR-595 (SEQ ID No :8), hsa-miR-609 (SEQ ID No :16), hsa-miR-613 (SEQ ID No :9), hsa- miR-939 (SEQ ID No :10), hsa-miR-1228* (SEQ ID No :11), hsa-miR-1231 (SEQ ID No :12) et hsa-miR-1909 (SEQ ID No :13), et une combinaison de ceux-ci. Selon un mode de réalisation, la composition pharmaceutique comprend en outre un ou plusieurs composés antirétro viraux.
Description détaillée de l'invention
Les inventeurs ont identifié des séquences de miARN capables de se fixer spécifiquement sur les séquences nucléotidiques de souches de VIH portant des mutations de résistance à un composé antirétroviral. Ils ont également démontré que ces miARN étaient ainsi capables de bloquer de manière particulièrement efficace la réplication virale desdites souches résistantes.
Ainsi, dans un premier aspect, la présente invention concerne un ARN interfèrent qui s'hybride préférentiellement avec l'ARNm d'un gène codant pour une protéine mutante d'une souche du virus VIH, ladite protéine mutante contenant une ou plusieurs mutations conférant à la souche virale un phénotype de résistance à un composé antirétroviral, pour une utilisation dans le traitement d'une infection par une souche du virus VIH résistante audit composé antirétroviral.
L'interférence par l'ARN est un phénomène bien connu de l'homme du métier qui permet d'inhiber de manière spécifique l'expression du gène cible au niveau post- transcriptionnel. Le terme « ARN interfèrent » ou « ARNi », tel qu'utilisé dans le présent document, se réfère à tout ARN, simple ou double brin, qui interfère avec un ARN messager spécifique conduisant ainsi à sa dégradation et/ou à la diminution de sa traduction en protéine. Ce terme englobe les petits ARN interférents (ARNsi), les ARN bicaténaires (ARNdb), les ARN monocaténaires (ARNsb), les ARN courts à épingle à cheveux (ARNsh), les ARNi ADN-dirigés (ddARNi) et les microARN (miARN).
Les ARN interférents utilisés selon l'invention peuvent être sous forme simple brin ou en duplexe ou en un mélange des deux. Ils peuvent contenir des nucléotides modifiés ou des modifications chimiques leur permettant, par exemple, d'augmenter leur résistance aux nucléases et ainsi d'augmenter leur durée de vie dans la cellule. Ils peuvent notamment comprendre au moins un nucléotide modifié ou non naturel tel que, par exemple, un nucléotide comportant une base modifiée, telle que l'inosine, la méthyl- 5-désoxycytidine, la diméthylarnino-5-désoxyuridine, la désoxyuridine, la diamino-2,6- purine, la bromo-5-désoxyuridine ou toute autre base modifiée permettant l'hybridation.
Les ARN interférents utilisés selon l'invention peuvent également être modifiés au niveau de la liaison internucléotidique comme par exemple les phosphorothioates, les
H-phosphonates ou les alkyl-phosphonates, ou au niveau du squelette comme par exemple les alpha- oligonucléotides, les 2'-O-alkyl ribose ou les PNA (Peptid Nucleic
Acid) (M. Egholm et al, 1992).
Les ARN interférents peuvent être des ARN naturels, synthétiques ou produits par des techniques de recombinaison. Ces ARN interférents peuvent être préparés par toutes méthodes connues de l'homme du métier telles que, par exemple, la synthèse chimique, le criblage de banques, la transcription in vivo ou des techniques d'ADN recombinant ou d'amplification.
Selon un mode de réalisation, l'ARN interfèrent est choisi dans le groupe constitué d'un ARNsi, d'un ARNsh et d'un miARN.
Selon un mode de réalisation particulier, l'ARN interfèrent est un ARNsi sous forme double brin d'environ 15 à 50 nucléotides de longueur, de préférence d'environ 15 à 30 nucléotides. Les ARNsi sont parfaitement complémentaires de leurs ARNm cibles.
Selon un autre mode de réalisation particulier, PARN interfèrent est un ARNsh sous forme simple brin, en forme de tige-boucle, d'environ 25 à 30 nucléotides et dont la taille de la boucle varie entre 4 à 23 nucléotides.
Selon un mode de réalisation préférée, l'ARN interfèrent est un miARN. Le terme « miARN » ou « microARN » englobe les miARN simple brin et double brin. De préférence, les miARN sont sous forme double brin. Les miARN sont partiellement complémentaires de leurs ARNm cibles et ont une taille comprise entre 10 et 25 nucléotides, de préférence entre 20 et 25 nucléotides. Les miARN utilisés selon l'invention peuvent être administrés sous la forme de précurseurs. Les miARN peuvent notamment être administrés sous la forme de pré-miARN ou de pri-miARN. Les pri- miARN sont des précurseurs de miARN qui sont clivés dans le noyau des cellules pour former des pré-miARN. Un pri-miARN peut comprendre un ou plusieurs pré-miARN. Les pré-miARN sont également des précurseurs des miARN. Ils comprennent entre 60 et 80 nucléotides et sont repliés en une structure tige-boucle imparfaite. Ces pré-miARN sont clivés dans le cytoplasme pour former des miARN double-brin puis des miARN simple brin capables d'interagir avec une protéine de la famille Argonaute pour former le complexe RISC grâce auquel soit l'ARNm cible est dégradé, soit la traduction de cet ARNm est réprimée.
Le terme « qui s 'hybride » signifie, dans le présent document, que des liaisons hydrogène de type Watson-Crick peuvent s'établir entre les bases complémentaires de deux brins d'acide nucléique pour former un duplexe. Le terme « qui s'hybride préférentiellement » signifie dans le présent document que l'ARN interfèrent utilisé selon l'invention est capable de s'hybrider à un gène ou un transcrit codant pour une protéine mutante d'une souche du virus VIH dans des conditions d'hybridation de forte ou de moyenne stringence. Les conditions de forte ou de moyenne stringence sont largement décrites dans la littérature, par exemple dans Sambrook et al., (1989), Maniatis et al., (1982 ou l'une de ses récentes rééditions) et dans Ausubel et al., (1995), et ces conditions peuvent être adaptées par l'homme du métier en fonction de la taille des fragments nucléotidiques, selon les enseignements appropriés connus.
La capacité de l'ARN interfèrent utilisé selon l'invention à s'hybrider préférentiellement à un gène ou un transcrit codant pour une protéine mutante d'une souche du virus VIH peut également être évaluée au moyen d'un test de compétition. L'ARN interfèrent est mis en présence d'une quantité équivalente (i) du gène ou du transcrit d'une protéine d'une souche de VIH non résistante à un composé antirétroviral et (ii) du gène ou du transcrit de la même protéine issue d'une souche VIH résistante à un composé antirétroviral et portant une ou plusieurs mutations. Ce mélange est réalisé dans des conditions de moyenne ou de forte stringence permettant l'hybridation. L'ARN interfèrent s'hydride préférentiellement à un gène ou un transcrit codant pour une protéine mutante d'une souche du virus VIH si, dans ces conditions, au moins 60% de l'ARN interfèrent s'hybride avec le gène ou le transcrit de la protéine mutante, de préférence au moins 70%, et de manière tout particulièrement préférée au moins 80%, au moins 90% ou au moins 95%, et si moins de 40% de l'ARN interfèrent s'hybride avec le gène ou le transcrit de la protéine non mutée, de préférence moins de 20%, moins de 15%, moins de 10% ou moins de 5%.
La capacité de l'ARN interfèrent utilisé selon l'invention à s'hybrider préférentiellement à un gène ou un transcrit codant pour une protéine mutante d'une souche du virus VIH peut également être évaluée au moyen d'outils bio informatiques bien connus de l'homme du métier.
Dans le cadre de la présente invention, le terme « ARNm d'un gène codant pour une protéine » est utilisé pour désigner la séquence ribonucléotidique issue de la transcription du gène codant pour la protéine correspondante. Ce terme englobe les régions en 3' et en 5' non traduites (3'-UTR et 5'-UTR), les exons et, éventuellement, les introns non épissés.
Le virus VIH peut être du type VIH-I ou VIH-2, de préférence du type VIH-I .
Tel qu'utilisé dans le présent document, le terme « souche de virus VIH résistante à un composé antirétroviral » désigne une souche de VIH pour laquelle l'administration dudit composé antirétroviral n'a pas d'effet, ou a un effet diminué, sur la réplication ou la production virale.
La protéine mutante conférant à la souche virale un phénotype de résistance à un composé antirétroviral peut être toute protéine codée par le génome du virus du VIH telle que, par exemple, la transcriptase inverse, la protéase, la glycoprotéine gp41, l'intégrase, la protéine pi 7, la protéine p24, la protéine p7, la protéine p6 ou la glycoprotéine gpl20. De préférence, La protéine mutante est choisie dans le groupe constitué de la transcriptase inverse, la protéase, la glycoprotéine gp41 et l'intégrase.
Le terme « composé antirétroviral » tel qu'utilisé dans ce document, désigne une molécule utilisée pour le traitement des infections liées à des rétro virus, notamment au VIH. Ces molécules sont essentiellement dirigées contre les enzymes ou autre molécules spécifiques de ce type de virus. Les molécules antirétro virales peuvent être des inhibiteurs de la transcriptase inverse c'est-à-dire des inhibiteurs nucléosidiques tels que la zidovudine, la lamivudine, l'emtricitabine, la didanosine, la stavudine, l'abacavir, la zalcitabine ou le tenofovir, des inhibiteurs non nucléosidiques tels que l'efavirenz, le nevirapine, l'étravirine ou la delavirdine, ou des analogues nucléotidiques tels que le tenofovir ou la fosalvudine ; des inhibiteurs de la protéase tels que l'amprenavir, le tipranavir, l'indinavir, le saquinavir, le fosamprenavir, le ritonavir, le darunavir, l'atazanavir ou le nelfînavir ; des inhibiteurs de la fusion tels que l'enfuvirtide ; des inhitibiteurs d'entrée tels que le maraviroc, le vicriviroc ou le TNX-355 ; des inhibiteurs de maturation tels que le bevirimat ; ou des inhibiteurs d'intégrase tels que le raltégravir, l'elvitégravir ou le MK-2048.
Selon un mode de réalisation particulier, la protéine mutante est l'intégrase et le composé antirétroviral est un inhibiteur d'intégrase. Dans le cas d'un virus non résistant aux inhibiteurs d'intégrase, ces derniers bloquent l'action de l'intégrase et empêchent ainsi le génome viral de se lier à celui de la cellule cible. Selon ce mode de réalisation de l'invention, l'intégrase contient une ou plusieurs mutations conférant à la souche virale un phénotype de résistance à un inhibiteur d'intégrase, de préférence le raltégravir (Isentress ou MK-0518, numéro CAS 518048-05-0) ou l'elvitégravir (JTK-303 ou GS 9137, numéro CAS 697761-98-1). De manière particulière, le ou les résidus mutés sont choisis dans le groupe constitué du résidu T66, E92, F121, E138, G140, Y143, S147, Q148, S153, N155, E157 et R263 de la SEQ ID No. 1, et une combinaison de ceux-ci. De manière préférée, le ou les résidus mutés sont choisis dans le groupe constitué du résidu E92, G140 et Y143 de la SEQ ID No. 1, et une combinaison de ceux-ci. De manière tout particulièrement préférée, la ou les mutations sont choisies dans le groupe constitué de E92Q, G140S, G140A, Y143R et Y143C, et une combinaison de celles-ci.
Les résidus d'acides aminés sont identifiés dans le présent document par le code à une lettre de l'acide aminé et sa position au sein de la séquence protéique. Les mutations d'acides aminés sont identifiées dans le présent document par le code à une lettre de l'acide aminé muté suivi de sa position au sein de la séquence protéique et du code à une lettre de l'acide aminé se substituant à l'acide aminé initial. A titre d'exemple, la mutation E92Q correspond à la substitution de l'acide glutamique en position 92 par une glutamine.

Dans un autre aspect, la présente invention concerne un vecteur d'expression comprenant une séquence nucléotidique codant pour un ARN interfèrent qui s 'hybride préférentiellement avec l'ARNm d'un gène codant pour une protéine mutante d'une souche du virus VIH, ladite protéine mutante contenant une ou plusieurs mutations conférant à la souche virale un phénotype de résistance à un composé antirétroviral, ou codant pour un précurseur de celui-ci, pour une utilisation dans le traitement d'une infection par une souche du virus VIH résistante à un composé antirétroviral.
Le vecteur d'expression selon l'invention comprend les éléments permettant l'expression de ladite séquence nucléotidique dans les cellules eucaryotes. De tels vecteurs sont bien connus de l'homme du métier et ont notamment été décrits dans la demande de brevet WO 06/085016 ou dans les articles de Barton and Medzhitov, 2002 ; Tiscornia et al, 2004 ; Xia et al, 2002 et Shen et al, 2003.
La présente invention concerne aussi un ARN interfèrent qui s 'hybride préférentiellement avec l'ARNm d'un gène codant pour une protéine mutante d'une souche du virus VIH, ladite protéine mutante contenant une ou plusieurs mutations conférant à la souche virale un phénotype de résistance à un composé antirétroviral, ou un vecteur d'expression comprenant une séquence nucléotidique codant pour ledit ARN interfèrent ou un précurseur dudit ARN, pour une utilisation en combinaison avec ledit composé antirétroviral dans le traitement d'une infection par le virus VIH.

La composition selon l'invention peut également comprendre en outre un ou plusieurs composés antirétroviraux tels que des inhibiteurs de la transcriptase inverse, des inhibiteurs de la protéase, des inhibiteurs de la fusion, des inhibiteurs de maturation, ou des inhibiteurs d'intégrase. Dans le cas où la composition comprend plusieurs composés antirétroviraux, ces derniers peuvent être des inhibiteurs de même type (inhibition du même mécanisme viral ou de la même enzyme) ou des inhibiteurs de types différents.
Selon un mode de réalisation particulier, la composition selon l'invention est un vaccin.
De manière avantageuse, le véhicule de la présente composition facilite la pénétration de l'ARN interfèrent ou au vecteur d'expression utilisé selon l'invention dans les cellules de l'organisme à traiter, et/ou protège l'ARN interfèrent ou le vecteur d'expression d'une éventuelle dégradation pouvant nuire à son efficacité. Parmi les véhicules utilisables, on peut notamment citer les polymères cationiques naturels tels que le chitosan ou l'atelocollagène, ou synthétiques tels que le po Iy(L- lysine), le polyéthylènimine (PEI) ou les dendrimères, qui forment des complexes avec les acides nucléiques de l'invention ; les liposomes ; les liposomes cationiques ; les liposome galactosylés ; les liposomes recouverts d'un ligand leur permettant de cibler un type de cellules tels que les immuno liposomes recouvert d'un anticorps spécifique de la cellule cible (Zheng et al., 2009) ; les liposomes disposés au sein d'une nanoparticule formée par des polymères (Carmona et al., 2009) ou encore des films multicouches de polycations et polyanions.
La composition selon l'invention peut être adaptée pour une administration par voie orale, sublinguale, parentérale, topique, rectale ou transdermique selon des protocoles classiques bien connus de l'homme du métier. De manière préférée, la composition est adaptée pour une administration par voie orale, par exemple sous la forme d'un comprimé, d'une gélule, ou d'une solution buvable, ou par voie parentérale, en particulier sous la forme d'une solution injectable, notamment par voie intraveineuse, intradermique ou sous cutanée.
Il est entendu que la dose à administrer peut être adaptée par l'homme du métier en fonction de nombreux paramètres tels que la physiologie du patient, l'état pathologique ou le mode d'administration. Typiquement, les ARN interférents sont administrés à des doses pouvant varier entre 0,1 et 20 mg par kilo et par jour, préférentiellement de 1 à 10 mg par kilo et par jour. Les administrations peuvent être hebdomadaires ou quotidiennes voire répétées plusieurs fois par jour. Les ARN interférents peuvent être administrés sous forme de dose unitaire comprenant de 0,05 à 20 mg d'ARN interférents, préférentiellement de 0,1 à 5 mg.
La présente invention concerne en outre un ARN interfèrent selon l'invention en tant que médicament. Selon un mode de réalisation particulier, l'ARN interfèrent est un miARN choisi dans le groupe constitué de hsa-miR-18a (SEQ ID No :2), hsa-miR-92a- 1 (SEQ ID No :3), hsa-miR-185* (SEQ ID No A), hsa-miR-219-l-3p (SEQ ID No :5), hsa-miR-370 (SEQ ID No :6), hsa-miR-518c* (SEQ ID No :7), hsa-miR-595 (SEQ ID No :8), hsa-miR-613 (SEQ ID No :9), hsa-miR-939 (SEQ ID No :10), hsa-miR-1228* (SEQ ID No :11), hsa-miR-1231 (SEQ ID No :12), hsa-miR-1909 (SEQ ID No :13), hsa-miR-138 (SEQ ID No :15) et hsa-miR-609 (SEQ ID No :16), et une combinaison de ceux-ci. La présente invention concerne également une méthode de traitement d'une infection par une souche du virus VIH résistante à un composé antirétroviral comprenant l'administration à un sujet, notamment humain, d'une quantité thérapeutiquement efficace d'un ARN interfèrent, d'un vecteur d'expression ou d'une composition pharmaceutique tels que définis ci-dessus.
Le terme « traitement » tel qu'utilisé dans ce document, se réfère à une amélioration ou disparition des symptômes, un ralentissement de la progression de la maladie, un arrêt de l'évolution de la maladie ou une disparition de la maladie. Ce terme englobe aussi bien le traitement préventif que curatif.
Le terme « quantité thérapeutiquement efficace » tel qu'utilisé ici, se réfère à une quantité suffisante pour avoir un effet inhibiteur sur la réplication et/ou la production virale.
Les exemples qui suivent sont présentés dans un but illustratif et non limitatif. Exemples
Matériels et méthodes
Lignée cellulaire et souches virales
Les expérimentations ont été effectuées avec des cellules Hela-P4 qui sont permissives à la réplication du VIH-I de sous type B utilisé dans ces expériences et dont le génome complet est clone dans le plasmide pNL4-3 (numéro d'entrée GenBank:
AF324493.1). Les cellules ont été cultivées dans du milieu DMEM avec 5% de sérum de veau fœtal.
Le plasmide pNL4-3 contient le génome complet de la souche sauvage du virus VIH-I. Les souches virales contenant des mutations de résistance aux antirétro viraux ont été obtenues par mutagénèse dirigée en modifiant le plasmide pNL4-3. Les virus utilisés dans ces expériences ne présentent donc pas d'autres mutations. Les virus comprenant la mutation E92Q (cag), E92Q (caa), G 140S (agc), G 140S (agt), G 140A
(gca), Y143C (tgc) ou Y143R (cgc) dans le gène codant pour l'intégrase ont ainsi été obtenus. Ces mutations correspondent aux mutations les plus fréquemment observées chez les patients en échec thérapeutique. Une même mutation de résistance peut être codée par des séquences nucléotidiques différentes.
ARN interférents
Les ARN interférents utilisés lors de ces expériences sont des ARN double brin dont la séquence correspond à un miARN compris dans la liste suivante : hsa-miR-18a (SEQ
ID No :2), hsa-miR-92a-l (SEQ ID No :3), hsa-miR-185* (SEQ ID No A), hsa-miR-
219-l-3p (SEQ ID No :5), hsa-miR-370 (SEQ ID No :6), hsa-miR-518c* (SEQ ID
No :7), hsa-miR-595 (SEQ ID No :8), hsa-miR-613 (SEQ ID No :9), hsa-miR-939
(SEQ ID No :10), hsa-miR-1228* (SEQ ID No : 11), hsa-miR-1231 (SEQ ID No :12) et hsa-miR- 1909 (SEQ ID No : 13).
Transfection des cellules
24 heures avant la transfection, 4.104 cellules ont été mises en cultures dans chaque puits de plaques 24 puits. Le lendemain, les cellules Hela-P4 ont été co- transfectées avec 0, 5, 10, 20 et 50 pMol d'un miARN et avec 100 ng d'un plasmide codant pour le génome complet d'un VIH-I sauvage (pNL4-3) ou muté. Cette transfection a été réalisée à l'aide du kit Lipofectamine 2000 (Invitrogen). Une solution de transfection comprenant 100 ng de pNL4-3 ou de pNL4-3 muté, 5 à 50 pMol d'un miRNA et 1 μL de Lipofectamine2000 a été dilué dans 50 μL de milieu OptiMEM. Les cellules Hela-P4 ont été mises en contact avec cette solution de transfection avant d'être incubées à 37°C (sous 5% de CO2) durant 4 à 6 heures. Les cellules ont ensuite été lavées et remises en cultures dans du milieu DMEM avec 5 % de sérum de veau fœtal.
Détermination de la CI 50
Après la transfection, les cultures cellulaires ont été suivies pendant 6 jours afin de détecter la production de VIH dans le surnageant en mesurant la production d'antigène p24 mesurée par technique ELISA (tests ELISA réalisés sur 150 μL de surnageant de culture cellulaire) et exprimée en pg/ml. Afin d'évaluer la capacité du miARN étudié à inhiber la production virale, l'antigénémie p24 a été mesurée dans les cultures transfectées avec et sans miARN. La concentration inhibitrice 50, c'est-à-dire la concentration de miARN permettant de diminuer la production d'antigène p24 de 50%, a ainsi été calculée.
Effets des miARN sur la réplication du VIH-I résistant aux inhibiteurs d'intégrase
La mesure de la production d'antigène p24 à chaque concentration de miARN a permis de définir la CI50 des différents miARN testés. Les valeurs de ces CI50 sont présentées dans le tableau ci-dessous.
Ces résultats démontrent que les miARN qui s'hybrident préférentiellement avec l'ARNm du gène codant pour une intégrase contenant une mutation conférant au virus une résistance aux anti-rétroviraux, sont capables d'inhiber la production virale d'une telle souche avec une grande efficacité.

Claims

Revendications
1. ARN interfèrent qui s'hybride préférentiellement avec l'ARNm d'un gène codant pour une intégrase mutante d'une souche du virus VIH, ladite intégrase mutante contenant une ou plusieurs mutations conférant à la souche virale un phénotype de résistance à un inhibiteur d'intégrase, pour une utilisation dans le traitement d'une infection par une souche du virus VIH résistante audit inhibiteur d'intégrase.
2. ARN interfèrent selon la revendication 1, ledit virus VIH étant le virus VIH-I.
3. ARN interfèrent selon la revendication 1 ou 2, ledit ARN étant choisi dans le groupe constitué d'un miARN, d'un ARNsi et d'un ARNsh.
4. ARN interfèrent selon l'une quelconque des revendications 1 à 3, ledit ARN étant un miARN.
5. ARN interfèrent selon l'une quelconque des revendications 1 à 4, ledit inhibiteur d'intégrase étant le raltégravir ou l'elvitégravir.
6. ARN interfèrent selon l'une quelconque des revendications 1 à 5, ladite intégrase contenant une ou plusieurs mutations conférant à la souche virale un phénotype de résistance audit inhibiteur d'intégrase, le ou les résidus mutés de l'intégrase étant choisis dans le groupe constitué du résidu T66, E92, F121, E138, G140, Y143, S147, Q148, S153, N155, E157 et R263 de la SEQ ID No. 1, et une combinaison de ceux-ci.
7. ARN interfèrent selon la revendication 6, ladite ou lesdites mutations étant choisies dans le groupe constitué de E92Q, G140S, G140A, Y143R et Y143C, et une combinaison de celles-ci.
8. ARN interfèrent selon l'une quelconque des revendications 1 à 7, ledit ARN interfèrent étant un miARN étant choisi dans le groupe constitué de hsa-miR-18a (SEQ ID No :2), hsa-miR-92a-l (SEQ ID No :3), hsa-miR-185* (SEQ ID No A), hsa-miR- 219-l-3p (SEQ ID No :5), hsa-miR-370 (SEQ ID No :6), hsa-miR-518c* (SEQ ID No :7), hsa-miR-595 (SEQ ID No :8), hsa-miR-613 (SEQ ID No :9), hsa-miR-939 (SEQ ID No :10), hsa-miR-1228* (SEQ ID No : 11), hsa-miR-1231 (SEQ ID No :12) et hsa-miR-1909 (SEQ ID No :13), et une combinaison de ceux-ci.

ARN interfèrent qui s'hybride préférentiellement avec l'ARNm d'un gène codant pour la transcriptase inverse mutante d'une souche du virus VIH, ladite transcriptase inverse mutante contenant une ou plusieurs mutations conférant à la souche virale un phénotype de résistance à un inhibiteur de transcriptase inverse, le ou les résidus mutés 10 de la transcriptase inverse étant choisis dans le groupe constitué du résidu D67 et T215 de la SEQ ID No. 14, et une combinaison de ceux-ci, pour une utilisation dans le traitement d'une infection par une souche du virus VIH résistante audit inhibiteur de transcriptase inverse.
15 10. ARN interfèrent selon la revendication 9, ledit ARN étant choisi dans le groupe constitué d'un miARN, d'un ARNsi et d'un ARNsh.
11. ARN interfèrent selon la revendication 9 ou 10, ledit ARN étant un miARN.
20 12. ARN interfèrent selon l'une quelconque des revendications 9 à 11, ledit ARN interfèrent étant un miARN choisi dans le groupe constitué de hsa-miR-138 (SEQ ID No :15) et hsa-miR-609 (SEQ ID No :16), et une combinaison de ceux-ci.
13. Vecteur d'expression comprenant une séquence nucléotidique codant pour un ARN 25 interfèrent tel que défini dans l'une quelconque des revendications 1 à 12, ou codant pour un précurseur dudit ARN interfèrent, pour une utilisation dans le traitement d'une infection par une souche du virus VIH résistante à un composé antirétroviral.
14. ARN interfèrent tel que défini à l'une des revendications 1 à 12 ou vecteur 30 d'expression tel que défini à la revendication 13, pour une utilisation en combinaison avec un composé antirétroviral, dans le traitement d'une infection par le virus VIH.
15. Composition pharmaceutique comprenant au moins un ARN interfèrent tel que défini dans les revendications 1 à 12 et/ou au moins un vecteur d'expression tel que défini dans la revendication 13, et un ou plusieurs excipients et/ou véhicules pharmaceutiquement acceptables.
16. Composition pharmaceutique selon la revendication 15 comprenant en outre un ou plusieurs composés antirétro viraux.
