Furthermore, the present invention relates to a MALDI chip and a system consisting of the holder and the MALDI chip.
- mass spectrometry For the analysis of samples, for example in drug chemistry or in biological research and production, mass spectrometry has become increasingly popular. For analysis of biomolecules in the samples, mass spectrometry with ionization by matrix-assisted laser desorption and ionization (MALDI) is preferably used.
- MALDI matrix-assisted laser desorption and ionization
- MALDI chip - In the MALDI method, in some cases more than 200 MALDI matrix points are applied to a sample carrier - referred to below as a MALDI chip - which then has to be analyzed. It will be understood by those skilled in the art that both the application of the samples and the later analysis, due to the plurality of MALDI matrix points arranged in a comparatively small space, requires very high precision and very precise positioning of the MALDI chip.
- a MALDI chip can be positioned very precisely and reproducibly with the holder according to the invention, so that the metering of the substances to be analyzed and / or the subsequent analyzes can be carried out very precisely.
- the holder according to the invention is simple and inexpensive to manufacture and use. Even deposits on the holder do not affect the exact position of the MALDI chips.
- the holder is suitable for holding MALDI chips with a broken, i. flattened corner contour.
- the holder has a first and a second contact surface, which are preferably arranged at right angles to one another and against which the MALDI chip rests. Furthermore, the holder according to the invention has a fixing means with which the MALDI chip can be fixed in contact with the contact surfaces. According to the MALDI chip is biased by the fixing means against the contact surfaces. This has the advantage that even with mechanical influences such as vibrations or temperature influences, the MALDI chip is permanently in contact with the first and the second contact surface, so that its position in a plane is at least clearly defined. Furthermore, according to the invention, the fixing agent cooperates with the broken corner contour.
- the holder has a support surface on which rests the MALDI chip.
- the contact surfaces of the holder according to the invention are chamfered such that the MALDI chip, in particular, when it is under pretension, is biased against the support. This has the advantage that the spatial position of the MALDI chip on the holder according to the invention is uniquely determined and the MALDI chip is always held in a unique position.
- the fixing means also has a contact surface, which cooperates with the MALDI chip and is preferably complementary to a corner contour of the MALDI chip.
- this contact surface is chamfered, so that the MALDI chip is also pressed by this contact surface against the support surface of the holder.
- the fixative is a slider.
- this slider has a spring which biases him in the direction of the contact surfaces.
- At least one contact surface of the holder is at least partially coated with a highly electrically conductive material, which is particularly preferably not corroded.
- the material is gold.
- the support surface has a recess, in particular a bore. This hole facilitates removal of the MALDI chip after analysis.
- one or more spring means is arranged on the opposite side of the support surface of the holder, with which the holder relative to the analyzer is biased.
- the holder has a lever which is reversibly convertible from a locking to an unlocking position.
- the lever is provided detent in both positions.
- the lever In the locked position, the lever preferably conceals the recess in the support surface and continues to block prefers the fixative. Both measures prevent the MALDI chip from being unintentionally removed from the holder.
- the opening in the support surface In the unlocked position, the opening in the support surface is freely accessible and the fixing means unlocked, so that the MALDI chip is detachable from the holder.
- Another object of the present invention is a Maldi chip having a gold-plated edge region.
- the gold-plated edge region and the analysis region are connected to one another by a conductor track.
- Yet another object of the present invention is a system consisting of the holder according to the invention and the Maldi chip according to the invention.
- FIG. 1 shows a MALDI chip 6 having a plurality of points 12 to which a MALDI matrix can be dosed.
- the MALDI chip has its upper left Corner a flattened corner contour 8, wherein the skilled artisan recognizes that the corner contour can also be designed differently.
- FIG. 2a shows the holder 1 according to the invention in the MALDI chip fixing state, the MALDI chip has been omitted for illustrative purposes.
- the holder 1 has a first and a second contact surface 2, 3, on which the MALDI chip 6 (not shown in this figure) is applied. Between the contact surfaces 2, 3, a recess 10 is arranged, which is required for a processing of the contact surfaces 2, 3, but also prevents the MALDI chip is applied only at its corner.
- the holder according to the invention has a support surface 5, on which the MALDI chip 6 rests.
- the slide 4 which is movable according to the directions indicated by the double arrow 9 and which is biased by a spring (not shown) in the direction of the contact surfaces 2.3, the MALDI chip is pressed against the contact surfaces 2.3.
- the upper illustration shows the slider from above ie from the side where the MALDI chip rests.
- the slider 4 is located in its front, ie in a position fixing the MALDI chip.
- the recess 15 can be seen in the support surface 5, which is covered in the MALDI chip fixing state from the rear of the lever 14.
- the bottom view shows the holder 1 from below.
- the lever 14 abuts against the holder and conceals the recess 15.
- the lever prevents on the one hand, that an object is inserted through the recess 15, which presses the MALDI chip from its fixation. Furthermore, the slider 4 is blocked by the lever 14 so that it can not be transferred to a retracted position.
- FIG. 2b shows the holder according to FIG. 2a in the rear unlocking position of the slide, in which the MALDI chip can be removed or a new MALDI chip can be inserted into the holder.
- the lever 14 is arranged in this position substantially perpendicular to the holder and thereby releases the opening 15. Furthermore, the locking of the slider 4 is released by the lever.
- FIG. 3 shows the MALDI chip 6 in its exactly fixed position.
- the abutment surfaces 2, 3 are chamfered, so that the Spring force of the fixing means 4 to the contact surfaces 2.3 also induces a downward, symbolized by the arrow 11 movement, with which the MALDI chip 6 is permanently held in contact with the support surface 5, thereby ensuring that the MALDI chip. 6 also spatially clearly positioned on the holder.
- FIG. 4 shows the slider of the inventive holder, which has a contact surface 7, which corresponds to the corner contour 8 of the MALDI chip 6. Also, the contact surface 7 is chamfered, so that the MALDI chip is also pressed by this surface against the support surface.
- the lever 14, which is shown here in the locking position, does not touch the slider 4 in this position. The slider 4 has contact in this position with the corner contour 8 of the MALDI chip 6 and presses it against the contact surfaces 2, 3.
- FIG. 5 shows how a MALDI chip is inserted into the holder according to the invention.
- the slider 4 is first moved to the bottom right and locked in this position. Thereafter, the MALDI chip is inserted into the holder so that the flattened corner contour 8 points in the direction of the slider.
- the slider is folded over and presses due to the force of the existing spring (not shown) the MALDI chip in the upper left corner that it rests against the contact surfaces 2, 3.
- FIG. 6 shows the underside of the holder, the leaf springs 13 which bias the holder against the analyzer and ensure good electrical contact with the analyzer.

The present invention relates to a holder for a MALDI chip, which has a broken corner contour, with a first and a second respectively chamfered contact surface and a movable fixing means, which cooperates with the broken corner contour and biases the MALDI chip against the contact surfaces. Furthermore, the present invention relates to a MALDI chip and a system consisting of the holder and the MALDI chip.
For the analysis of samples, for example in drug chemistry or in biological research and production, mass spectrometry has become increasingly popular. For analysis of biomolecules in the samples, mass spectrometry with ionization by matrix-assisted laser desorption and ionization (MALDI) is preferably used.
In the MALDI method, in some cases more than 200 MALDI matrix points are applied to a sample carrier - referred to below as a MALDI chip - which then has to be analyzed. It will be understood by those skilled in the art that both the application of the samples and the later analysis, due to the plurality of MALDI matrix points arranged in a comparatively small space, requires very high precision and very precise positioning of the MALDI chip.
It was therefore the object of the present innovation to provide a holder on which a MALDI chip can be positioned very accurately and reproducibly.
The object is achieved with a holder according to the
It was extremely surprising for the person skilled in the art and not to be expected that a MALDI chip can be positioned very precisely and reproducibly with the holder according to the invention, so that the metering of the substances to be analyzed and / or the subsequent analyzes can be carried out very precisely. The holder according to the invention is simple and inexpensive to manufacture and use. Even deposits on the holder do not affect the exact position of the MALDI chips.
The holder is suitable for holding MALDI chips with a broken, i. flattened corner contour.
According to the invention, the holder has a first and a second contact surface, which are preferably arranged at right angles to one another and against which the MALDI chip rests.
The holder has a support surface on which rests the MALDI chip.
The contact surfaces of the holder according to the invention are chamfered such that the MALDI chip, in particular, when it is under pretension, is biased against the support. This has the advantage that the spatial position of the MALDI chip on the holder according to the invention is uniquely determined and the MALDI chip is always held in a unique position.
In a preferred embodiment of the holder according to the invention, the fixing means also has a contact surface, which cooperates with the MALDI chip and is preferably complementary to a corner contour of the MALDI chip. Preferably, this contact surface is chamfered, so that the MALDI chip is also pressed by this contact surface against the support surface of the holder.
Any suitable fixing agent is known as a fixing agent. Preferably, however, the fixative is a slider. Preferably, this slider has a spring which biases him in the direction of the contact surfaces.
Preferably, at least one contact surface of the holder is at least partially coated with a highly electrically conductive material, which is particularly preferably not corroded. For example, the material is gold.
In a further preferred embodiment of the present invention, the support surface has a recess, in particular a bore. This hole facilitates removal of the MALDI chip after analysis.
Further preferably, one or more spring means is arranged on the opposite side of the support surface of the holder, with which the holder relative to the analyzer is biased.
In addition, preferably, the holder has a lever which is reversibly convertible from a locking to an unlocking position. Preferably, the lever is provided detent in both positions. In the locked position, the lever preferably conceals the recess in the support surface and continues to block prefers the fixative. Both measures prevent the MALDI chip from being unintentionally removed from the holder. In the unlocked position, the opening in the support surface is freely accessible and the fixing means unlocked, so that the MALDI chip is detachable from the holder.
Another object of the present invention is a Maldi chip having a gold-plated edge region.
Preferably, the gold-plated edge region and the analysis region are connected to one another by a conductor track.
Yet another object of the present invention is a system consisting of the holder according to the invention and the Maldi chip according to the invention.
