- La micro-déposition consiste à fixer des sondes longues de quelques dizaines à plusieurs centaines de bases à la surface d'une lame de verre. Ces sondes sont généralement extraites de bases de données et se présentent sous forme de produits amplifiés et purifiés. Cette technique permet de réaliser des puces dénommées microarrays portant environ dix mille spots, dit zones de reconnaissance, d'ADN sur une surface d'un peu moins de 4 cm2. Il ne faut toutefois pas oublier l'emploi de membranes de Nylon, dites « macroarrays », qui portent des produits amplifiés, généralement par PCR, avec un diamètre de 0,5 à 1 mm et dont la densité maximale est de 25 spots/cm2. Cette technique très flexible est utilisée par de nombreux laboratoires. Dans la présente invention, cette dernière technique est considérée comme faisant partie des biopuces. En protégeant cette surface par des masques de formes définies, on peut illuminer et donc activer sélectivement des zones de la puce où l'on souhaite fixer l'un ou l'autre des quatre nucléotides. L'utilisation successive de masques différents permet d'alterner des cycles de protection/réaction et donc de réaliser les sondes d'oligonucléotides sur des spots d'environ quelques dizaines de micromètre carré (µm2). Cette résolution permet de créer jusqu'à plusieurs centaines de milliers de spots sur une surface de quelques centimètres carré (cm2). - la recherche de la présence ou de l'absence de mutations. Connaissant la structure moléculaire du gène, on fabrique des oligonucléotides qui représentent toute ou partie complémentaire de ce gène. En présence de l'échantillon biologique, il y aura hybridation entre toute ou partie dudit gène, généralement marqué, par exemple en fluorescence, et les oligonucléotides, et l'image obtenue par fluorescence permet de savoir s'il y a une mutation et en quelle position elle est située. Dans cette application, l'utilisation des puces à ADN est l'équivalent d'un séquençage pour un diagnostic de mutation, avec un énorme avantage en terme de rapidité, et
- la mesure du niveau d'expression de gènes dans un tissu. Le réseau de la puce porte de très nombreuses sondes qui correspondent à l'ensemble des gènes de l'espèce à étudier. On hybride un échantillon, par exemple d'ARNm préalablement amplifiés, qui représente les gènes actifs du tissu. L'analyse par fluorescence permet de connaître le niveau d'expression de chaque gène.

- Method according to Claim 1 which consists in:- bringing at least one second liquid sample, containing at least one biological target molecule (3), into contact with at least one third liquid sample, containing at least one biological detection molecule (4), in order to bind this (these) target molecule(s) (3) to the detection molecule (s) (4),- bringing at least one first liquid sample, containing at least one group of mutually identical biological recognition molecules (2), into contact with the mixture of the at least two second and third liquid samples, containing at least one target molecule (3) optionally complexed with a detection molecule (4), in order to bind this (these) target molecule(s) (3) to said recognition molecule(s) (2),- bringing the support (1) into contact with the mixture of the at least three first, second and third liquid samples, containing at least one group of mutually identical biological recognition molecules (2), said recognition molecule(s) (2) optionally being complexed with at least one target molecule (3) which itself (3) is optionally complexed with a detection molecule (4), in order to bind these identical recognition molecules (2) to the support (1), preferentially in a recognition zone,- making a first image (10) of said support (1) after this binding of the labelled detection molecule(s) (4 and/or 9),- treating the support under physicochemical conditions that allow the separation of each detection molecule (4) from a target molecule (3) to which it (4) is specifically bound,- making a second image (11) of said support (1) after this separation, allowing the detection of the labelled detection molecule(s) (9) which is (are) not specifically bound to the target molecule(s) (3), and- analysing the two images in order to determine the specific bindings between the target molecule(s) and detection molecules.
- Method according to any one of Claims 5 to 10, characterized in that it comprises the following additional subsequent steps:- bringing the treated, functionalized support, after specific separation, into contact with the third liquid sample or with another liquid sample in order to again bind at least one labelled detection molecule (4) to:- the target molecule(s) (3) from which it (they) had been separated, and/or- any other target molecule(s) (3) bound to one or more recognition molecules (2) originating from the same recognition zone,- making a third image (12) of said support (1) after this new binding of the detection molecule(s) (4 and/or 9), and- analysing the third image by comparison with the previous analysis of the first two images in order to confirm the specific bindings between the target molecule(s) and detection molecules.
- Method according to any one of Claims 2 to 11, characterized in that at least one wash is carried out after the binding of:- the recognition molecules (2) to the support (1), the recognition molecules (2) optionally being bound to labelled target molecules and/or unlabelled target molecules (3), the latter (3) optionally being bound to detection molecules (4), and/or- the labelled target molecules and/or unlabelled target molecules (3) to the recognition molecules (2), the latter (2) previously being bound to said support (1), the target molecules (3) optionally being bound to detection molecules (4), and/or- the detection molecules (4) to unlabelled target molecules (3), the latter (3) being bound to recognition molecules (2) which themselves (2) are bound to the support (1).
- Method according to any one of Claims 2 to 12, characterized in that the support (1) consists of magnetic particles, and in that the method comprises steps for the magnetization of said magnetic particles, said particles preferably being put in place:- during any physicochemical treatment step and/or image making step according to any one of Claims 1 to 10, and/or- before any washing step according to Claim 11.
- Method according to any one of Claims 2 to 13, characterized in that the bringing of the support (1) into contact with at least two different first liquid samples, optionally mixed beforehand, allows the binding of at least two different recognition molecules (2) to said support (1) in at least two distinct recognition zones.
- Method according to any one of Claims 2 to 14 for quantifying at least two optionally different target molecules (3), characterized in that the detection molecule (4) consists of a molecule directly or indirectly associated with at least one marker, such as a nanoparticle, for which the means of making the images (10, 11 and/or 12) allow individual visualization despite the presence of other neighbouring and similar detection molecules (4 and/or 9).
- Method according to any one of Claims 2 to 15, characterized in that the physicochemical conditions that allow the separation of the labelled target molecules from the recognition molecules (2) or the detection molecules (4) from the target molecules (3) are obtained by heating to a melting point.
- Method according to any one of Claims 2 to 16, characterized in that the labelled target molecules, the recognition molecules (2) or the target molecules (3) consist of, or the detection molecules (4) comprise, nucleic acids.
- Method according to any one of Claims 2 to 16, characterized in that the labelled target molecules, the recognition molecules (2) or the target molecules (3) consist of, or the detection molecules (4) comprise, antibodies and/or antigens.
- Method according to any one of Claims 2 to 18, characterized in that the labelling of the target molecules or the detection molecules (4) is effected by:- particles visualizable by conventional optical microscopy, fluorescence microscopy, dark-field microscopy or atomic force microscopy,- molecules visualizable by atomic force microscopy,- precipitating-product enzymes which produce a signal detectable e.g. by fluorescence or luminescence, such as horseradish peroxidase, alkaline phosphatase, β-galactosidase or glucose-6-phosphate dehydrogenase,- chromophores such as fluorescent or luminescent compounds,- electronic markers detectable by electron microscopy, or- absorbent molecules visualizable by thermal lens microscopy.
- Method according to a combination of Claim 4 or 7, on the one hand, and Claim 18, on the other, characterized in that the other liquid sample, taking the place of either the second or the third liquid sample, to be brought into contact with the treated, functionalized support, after specific separation, comprises a different marker from that present in said second or third liquid sample.
