Beschrieben wird ein Verfahren zur Bestimmung der Aktivität von JNK/SAPKs, das auf der Durchführung einer Kinase-Reaktion mit aus einem Zellysat erhaltenen, vorzugsweise über Immunpräzipitation isolierten JNK/SAPKs in Gegenwart von ATP und GST-Transkriptionsfaktor (bevorzugt: GST-c-Jun) und anschliessender Bestimmung der Menge an phosphoryliertem GST-Transkriptionsfaktor (bevorzugt: p-GST-c-Jun) über die spezifische Bindung eines anti-p-GST-Transkriptionsfaktor-Antikörpers (bevorzugt: anti-p-GST-c-Jun-Antikörpers) basiert.

Description

Verfahren zur Messung der JNK/SAPKs-A tivität mittels der Bestimmung der Phosphorylierung von Transkriptionsfaktoren
Die vorliegende Erfindung betrifft ein Verfahren zur Bestimmung der Aktivität von JNK/SAPKs, das auf der Durchführung einer Kinase-Reaktion mit aus einem Zellysat erhaltenen JNK/SAPKs in Gegenwart von ATP und GST- Transkriptionsfaktoren und anschließender Bestimmung der Menge an phosphoryliertem GST-Transkriptionsf ktor (p-GST- Transkriptionsfaktor) über die spezifische Bindung eines anti- p-GST-Transkriptionsfaktor-Antikörpers basiert.
c-Jun-NH2-terminale Kinasen (JNK) , die auch unter der Bezeichnung SAPK (Stress-aktivierte Proteinkinasen) bekannt sind, sind Signal transduktionsproteine , die unter verschiedensten Bedingungen aktiviert werden können (Davis, Biochem. Soc . Symp. 64 (1999), 1-12; Gupta et al . , Science 267 (1995), 389-393). JNK/SAPKs regulieren die Genexpression über die Phosphorylierung und, dadurch, die Aktivierung von Transkriptionsfaktoren wie z.B. c-Jun (Davis, Biochem. Soc. Symp. 64 (1999), 1-12; Minden und Karin, Biochim. Biophy. Acta 1333 (1997), 85-104; Herr et al . , EMBO J. 16 (1997), 6200- 6208) . Somit spiegelt der Phosphorylierungsstatus der Transkriptionsfaktoren, z.B. von c-Jun, die Aktivität von JNK/SAPKs wieder und darauf beruhen auch die üblichen Assays zur Bestimmung dieser Aktivität. In diesen JNK/SAPKs-Assays werden JNKs aus Zellysaten immunpräzipitiert und durch Inkubation der gereinigten JNKs zusammen mit GST-c-Jun-Protein und 32P-γ-ATP wird ein in vitro Kinase-Assay durchgeführt. Über SDS-PAGE und Autoradi ographi e wird die Menge an phosphoryliertem GST-c-Jun-Protein bestimmt, die die Aktivität der JNK/SAPKs reflektiert (Hibi et al . , Genes Dev. 7 (1993), 2135-2148) . Allerdings weisen diese Assays folgende Nachteile auf: Sie basieren auf radioaktiven Markierungen, sind relativ unempfindlich, zeitaufwendig und weisen einen hohen Background auf.
Somit liegt der vorliegenden Erfindung das technische Problem zugrunde, einen JNK/SAPKs-Assay bereitzustellen, der die vorstehend zusammengefaßten Nachteile vermeidet, d.h. keine radioaktive Markierung erfordert und darüber hinaus trotz einem geringeren Zeitaufwand (Expositionszeit) empfindlich ist.
Die Lösung dieses technischen Problems wird durch die Bereitstellung der in den Patentansprüchen gekennzeichneten Ausführungsformen erzielt.
Es stellte sich während den zu dieser Erfindung führenden Experimenten heraus, dass die vorstehend geschilderten Probleme überwunden werden können, indem die Aktivität von JNK/SAPKs dadurch bestimmt wird, dass eine Kinase-Reaktion mit aus einem Zellysat erhaltenen JNK/SAPKs in Gegenwart von ATP und GST-Transkriptionsfaktoren durchgeführt wird, und anschließend eine Bestimmung der Menge an phosphoryliertem GST-Transkriptionsfaktor (p-GST-Transkriptionsfaktor) über die spezifische Bindung eines anti-p-GST-Transkriptionsfaktor- Antikörpers erfolgt, wobei die Menge an gebundenem Antikörper die Menge an phosphoryliertem GST-Transkriptionsfaktor (p-GST- Transkriptionsfaktor) reflektiert und somit indirekt die Aktivität der JNK/SAPKs.
Somit betrifft die vorliegende Erfindung ein Verfahren zur Bestimmung der Aktivität von JNK/SAPKs, wobei das Verfahren die folgenden Schritte umfaßt:
(a) Herstellung eines Zellysats oder Gewebelysats ;
(b) Aufreinigung von endogenen JNK/SAPKs;
(c) Durchführung einer Kinase-Reaktion mit den nach Schritt (b) erhaltenen JNK/SAPKs in Gegenwart von ATP und eine oder beide N-terminale Phosphorylierungss teilen enthaltendem GST- Transkriptionsfaktor-Protein; und (d) Nachweis der Menge an phosphoryliertem GST- Transkriptionsfaktor-Protein durch Zugabe eines spezifischen a ti-p-GST-Transkriptionsfaktor- Antikörpers zu dem Gemisch aus Schritt (c) und Bestimmung der Aktivität der JNK/SAPKs anhand der Menge an gebundenem anti-p-GST-Transkriptionsfaktor- Antikörper.
Unter dem Begriff "Transkriptionsfaktor" sollen die nachfolgenden speziellen Substrate für JNK/SAPKs verstanden werden, z.B. (TCF)/Elk-1, c-Jun, Jun B, Jun D und ATF-2 , ganz besonders c-Jun.
Die Herstellung eines Zellysats zur Gewinnung der JNK/SAPKs kann mittels dem Fachmann bekannten Routineverfahren erfolgen, z.B. mittels des in dem nachstehenden Beispiel beschriebenen Verfahrens. Der Fachmann kennt auch alle Maßnahmen, die ergriffen werden müssen, damit bei dem Aufschließen der Zellen die JNK/SAPKs vollständig und in aktiver Form zur Verfügung stehen. So kann z.B. der Abbau durch Proteasen mittels bekannten Protease-Hemmern, wie z.B. PMSF, unterdrückt werden.
Um die Aktivität der JNK/SAPKs bestimmen zu können, ist es vorteilhaft, die endogenen JNK/SAPKs aus dem Zellysat anzureichern und dadurch die endogenen Transkriptionsfaktor- Proteine abzutrennen. Dies kann durch dem Fachmann bekannte Verfahren erfolgen, z.B. mittels Immunpräzipitation oder klassischen Proteinaufreinigungsverfahren.
In einer bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens erfolgt die Anreicherung bzw. Isolierung der JNK/SAPKs aus dem Zellysat über Immunpräzipitation. Dies erfolgt über anti-JNKs/SAPK-Antikörper, die z.B. an eine Säule oder an Kügelchen, z.B. magnetische Kügelchen, gebunden sind, gemäß Standardverfahren. Dabei handelt es sich bei den für dieses Verfahren geeigneten Antikörpern um monoklonale, polyklonale oder synthetische Antikörper. Der hier verwendete Ausdruck "Antikörper" umfaßt auch Fragmente. In diesem Zusammenhang bedeutet der Begriff "Fragment" alle Teile des monoklonalen Antikörpers (z.B. Fab-, Fv- oder "Single chain Fv"-Fragmente) , welche die gleiche Epitopspezifität wie der vollständige Antikörper aufweisen. Die Herstellung solcher Fragmente ist dem Fachmann bekannt. Vorzugsweise handelt es sich bei den erfindungsgemäßen Antikörpern um monoklonale Antikörper. Die erfindungsgemäßen Antikörper können gemäß Standardverfahren hergestellt werden, wobei vorzugsweise JNK/SAPKs-Peptide oder synthetische Fragmente davon als Immunogen dienen. Diese Peptide bzw. die Fragmente davon können durch Gewinnung des entsprechenden Gens, Klonierung und rekombinante Expression erzeugt werden. Verfahren zur Gewinnung monoklonaler Antikörper sind dem Fachmann bekannt .
In einer besonders bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens erfolgt die Immunpräzipitation der endogenen, im Zellysat enthaltenen JNK/SAPKs dadurch, dass diese mit spezifischen Antikörpern inkubiert und danach an Protein-A-Sepharose-Kügelchen gebunden werden. Dabei sollten die Antikörper gegen alle JNK/SAPKs gerichtet sein bzw. gegen ein Epitop, das allen JNK/SAPKs gemeinsam haben, damit der Assay quantitativ ist, d.h. die Gesamtaktivität aller JNK/SAPKs erfaßt wird. Die weiteren Verfahrensschritte können dann mit den von den Protein-A-Sepharose-Kügelchen freigesetzten JNK/SAPKs oder noch-gebundenen JNK/SAPKs erfolgen, jedenfalls muß gewährleistet sein, dass durch Bindung an den Antikörper die biologische Aktivität der JNK/SAPKs nicht gehemmt oder sonstwie nachteilig beeinflußt wird.
In einer noch mehr bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens sind die spezifischen Antikörper anti-JNKl-, anti-JNK2- und/oder anti-JNK3-Antikörper, die gegen gemeinsame Epitope von JNKl, JNK2 und JNK3 gerichtet sind (Davis et al . , Biochem. Soc. Sympl . 1999, 64, S. 1-12). Die vorstehend beschriebenen Antikörper sind im Handel erhältlich. Im nachfolgenden wird die Erfindung am Beispiel des Transkriptionsfaktors c-Jun beschrieben. Dies ist aber nicht als Beschränkung der Erfindung zu verstehen. Bezüglich der übrigen vorstehend aufgezählten Transkriptionsfaktoren ist analog zu verfahren.
Die Durchführung der Kinase-Reaktion mit den gewonnenen JNK/SAPKs erfolgt in Gegenwart von ATP und eine oder beide N- terminale Phosphorylierungsstellen enthaltendem GST-c-Jun. Bedingungen für Kinase-Reaktionen sind dem Fachmann bekannt und die Reaktion kann z.B. gemäß dem nachstehenden Beispiel durchgeführt werden. Der Fachmann kann auch das für diesen Reaktionsschritt erforderliche GST-c-Jun selbst reinigen oder rekombinant herstellen (van Dam et al . , EMBO J. 1995, 14: 1798-1811) Geeignetes GST-c-Jun-Protein ist auch im Handel erhältlich (siehe dazu die Angaben im nachstehenden Beispiel) . GST-c-Jun ist ein rekombinant in Bakterien exprimiertes c-Jun- Protein, das an GST fusioniert ist. GST ermöglicht die Aufreinigung des rekombinanten c-Jun-Proteins über Glutathion- Sepharose-Beads .
Der Begriff "GST-c-Jun" umfaßt alle Formen des Proteins, die noch einer Phosphorylierungsreaktion durch JNK/SAPKs zugänglich sind, somit auch Formen, die gegenüber den natürlichen Formen Veränderungen aufweisen, z.B. ein anderes Glykosilierungsmuster, Austausch, Deletion und/oder Addition von Aminosäure (n) etc. Dazu zählen auch verkürzte Formen von GST-c-Jun, wobei die verkürzten Formen C-terminal oder N- terminal verkürzt sein können, jedenfalls noch mindestens eine Phosphorylierungsstelle aufweisen, vorzugsweise besitzen sie noch beide Phosphorylierungsstellen.
In der am meisten bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens ist das für die Kinase-Reaktion verwendete GST-c-Jun GST-c-Jun 1-166 oder GST c-Jun- 1-79, die z.B. von der Firma Santa Cruz, Heidelberg erhältlich sind, wobei die Zahlen auf die Positionen der verbliebenen Aminosäuren hinweisen. Ein für den Nachweis des phosphorylierten GST-c-Jun geeigneter Antikörper kann gemäß üblicher Verfahren hergestellt werden, wobei bezüglich dieses Antikörpers bzw. dessen Herstellung die vorstehenden Ausführungen zu anti-JNK-Antikörpern sinngemäß gelten. Dieser Antikörper muß zwei Hauptvoraussetzungen erfüllen: Er muß spezifisch an GST-c-Jun binden und zwischen der phosphorylierten und unphosphorylierten Form unterscheiden können, d.h. nur an die phosphorylierte Form binden. Solche Antikörper können z.B. dadurch hergestellt werden, dass pho sphoryl i er t e s GST-c-Jun (vorzugsweise beide Phosphorylierungsss teilen enthaltend) oder ein die phosphorylierten Bindungstelle (n) (z.B. Ser63 und/oder Ser73) enthaltendes Fragment als Immunogen verwendet wird und dann nach Antikörpern gescreent wird, die nur noch an die phosphorylierte Form des GST-c-Jun binden, nicht jedoch an die unphosphorylierte.
Für das e r f i ndung s gemäße Verfahren liegt der Konzentrationsbereich von GST-c-Jun vorzugsweise zwischen 0,2 - 5 μg pro 50 μl Kinase-Reaktion, wobei der Bereich von 1-3 jw.g/50 μ l Kinase-Reaktion bevorzugt ist. Der Konzentrationsbereich des verwendeten Zellysats liegt vorzugsweise zwischen 50 - 1000 μg Protein pro 50 μl Kinase- Reaktion, wobei der Bereich von 100 - 300 μg bevorzugt ist. Vorzugsweise ist der anti-p-GST-c-Jun Antikörper im Vergleich zu GST-c-Jun im Überschuß vorhanden, um eine quantitative Messung zu erhalten, wobei die Konzentration des Antikörpers von seiner Affinität abhängt.
Die quantitative Bestimmung der (absoluten) JNK/SAPKs- Aktivität kann durch Vergleich mit einer Kontrollreaktion erfolgen, die JNK/SAPKs mit bekannter Aktivität enthält.
In einer weiteren bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens erkennt der anti-p-GST-c-Jun- Antikörper spezifisch die Phosphorylierung an Ser73 und/oder Ser63. Die Bestimmung der Aktivität von JNK/SAPKs über die Bindung des anti-p-GST-c-Jun-Antikörpers kann über verschiedene allgemeine Verfahren erfolgen, z.B. Wes tern-Blot , Immunfluoreszenz, ELISA, Radioimmunassay (RIA) etc., wobei Western-Blot für das erfindungsgemäße Verfahren bevorzugt ist. Vorzugsweise trägt der anti-p-GST-c-Jun-Antikörper eine nachweisbare Markierung, wobei als Markierung eine fluoreszente Verbindung, ein kolloidales Metall, eine chemilumineszente Verbindung, eine biolumineszente Verbindung, eine phosphoreszente Verbindung oder ein Enzym besonders bevorzugt ist. In der am meisten bevorzugten Ausführungsform des erfindungsgemäßen Verfahrens wird die JNK/SAPKs-Aktivität über einen primären polyklonalen Kaninchen anti-p-GST-c-Jun- Antikörper und einen im Handel erhältlichen sekundären anti- Kaninchen-Meerrettich-Peroxidase-Antikörper und verstärkte Chemilumineszenz bestimmt. Der anti-p-GST-c-Jun-Antikörper kann auch an einen festen Träger über übliche Verfahren absorbiert sein. Desweiteren besteht die Möglichkeit, daß der primäre Antikörper biotin-konjugiert ist und ein Streptavidin- Peroxidase gekoppelter Sekundärantikörper verwendet wird. Falls eine Bestimmung im Fluoreszenz-Mikroskop oder FACS gewünscht ist, wird vorzugsweise ein Fluorescein- oder Rhodamin-gekoppelter Sekundär-Antikörper eingesetzt.
Die vorliegende Erfindung stellt auch Kits bereit, die für das erfindungsgemäße Verfahren von Nutzen sind und vorzugsweise einen anti-p-GST-c-Jun-Antikörper oder ein Fragment davon enthalten und ein eine oder beide Phosphorylierungsstellen enthaltendes GST-c-Jun-Protein sowie einen Antikörper mit dem sich JNKl, JNK2 und/oder JNK3 durch Immunpräzipitation anreichern lassen. Der Kit kann außerdem JNK/SAPKs oder aktive Teile davon enthalten, z.B. zur Kontrolle oder zur genauen Quantifizierung sowie Protein A-Sepharose-Beads zur Immunpräzipitation. Je nach Ausgestaltung des Kits kann der Antikörper an eine weitere Einheit konjugiert sein, beispielsweise eine Markierung und/oder er kann auf einem festen Träger (Substrat) immobilisiert sein. Der Kit kann auch einen zweiten Antikörper zum Nachweis von p-GST-c- Jun/Antikörper-Komplexen enthalten. Der Antikörper oder das Fragment davon können frei vorliegen oder an einen festen Träger immobilisiert sein, beispielsweise eine Plastikschale, ein Teströhrchen, eine Mikrotiterplatte, ein Teststäbchen etc. Der Kit kann auch Anleitungen enthalten, die die Verwendung der enthaltenen Verbindungen in einem Assay zur Bestimmung der JNK/SAPKs-Aktivität erläutern. Der Kit kann weiterhin geeignete Reagenzien zum Nachweis von Markierungen oder zur Markierung positiver und negativer Kontrollen, Waschlösungen, ECL-Reagenzien (Fa. Amersham, Braunschweig), Verdünnungspuffer etc . enthalten.
Das erfindungsgemäße Verfahren ist auch für Zwecke von Nutzen, bei denen eine auf der Aktivierung von JNK/SAPKs beruhende Krebstherapie durchgeführt wird. Somit betrifft die vorliegende Erfindung auch ein Verfahren zur Beurteilung der Wirksamkeit einer Krebstherapie, dadurch gekennzeichnet, dass die therapeutische Aktivierbarkeit der JNK/SAPKs über das vorstehend beschriebene erfindungsgemäße Verfahren bestimmt wird, vorzugsweise zu verschiedenen Zeitpunkten im Verlauf der Therapie.
Ein weiterer Vorteil des erfindungsgemäßen Verfahrens ist, daß JNK/SAPKs vollständig aus einem Zell-Lysat immunpräzipitiert werden. Deshalb kann man mit dieser Methode nicht nur die Phosphorylierung von c-Jun, sondern selektiv die Phosphorylierung aller Substrate der JNK/SAPKs analysieren. Zu diesen Substraten zählen z.B. die Transkriptionsfaktoren (TCF)/Elk-1, c-Jun, Jun B, Jun D und ATF-2. Der Nachweis der phosphorylierten Proteine erfolgt über einen Antikörper, der spezifisch die phosphorylierte Form der entsprechenden Transkriptionsfaktoren erkennt.
Kurze Beschreibung der Figuren:
Fiσur 1 : Schema zum erfindungsgemäßen Assay
JNK/SAPKs werden mit spezifischen Antikörpern (FL, C-17, Fa.
Santa Cruz) immunpräzipitiert, an Protein-A-Sepharose gebunden und anschließend mit nichtradioaktivem ATP und GST-c-Jun- Protein inkubiert. Die Aktivität von JNK/SAPKs führt zur Phosphorylierung des GST-c-Jun-Proteins an Ser-63 und Ser-73. Die Menge der Phosphorylierung, die die Aktivität von JNK/SAPKs reflektiert, kann über Western-Blot Analyse mittels eines phospho-c-Jun-spezifischen Antikörpers nachgewiesen werden.

Lysate von JURKAT-Zellen, die 4 Stunden mit Cisplatin (1 μg/ml) behandelt worden waren, um die JNK/SAPKs zu aktivieren, wurden zur Immunpräzipitation von JNK/SAPKs mit den polyklonalen Kaninchen-IgG-Antikörpern FL und C17 (Santa Cruz) und Protein-A-Sepharose verwendet . Die Kinase-Reaktion wurde durch Inkubation des Immunpräzipitats mit Kinase-Puffer und unmarkiertem ATP (Spur a) oder mit Kinase-Puffer, unmarkiertem ATP und GST-c-Jun 1-166 als Substrat (Spur d) durchgeführt. Alternativ wurde der Kinase-Puffer nur mit GST-c-Jun 1-166 und unmarkiertem ATP ohne Immunpräzipitat inkubiert (Spur b) . Die verschiedenenen Reaktionsgemische oder nur Kinase-Puffer (Spur c) wurden mittels 12% SDS-PAGE aufgetrennt und mittels Western-Blot analysiert. Das MG (in kDa) ist auf der linken Seite der Figur angegeben. Die Phosphorylierung des GST-c-Jun- Proteins wurde mittels einem polyklonalem phospho-c-Jun-Ser73- spezifischen IgG-Antikörpers (New England BioLabs) und verstärkter Chemilumineszenz nachgewiesen. Erklärung zum Verfahren: Bei der verstärkten Chemilumineszenz wird ein Peroxidase-gekoppel ter Sekundärantikörper an den Primärantikörper gebunden. Gibt man eine Lösung zu, die das Substrat "Luminol" enthält, wird Luminol von der Peroxidase zersetzt und es kommt zu einer Lichtreaktion. Die Menge des entstehenden Lichtes kann quantitativ auf einem Röntgenfilm oder im Luminometer erfaßt werden.
Das folgende Beispiel erläutert die Erfindung. Beispiel 1
Nicht-radioaktiver Nachweis der Aktivität von JNK/SAPKs
Insgesamt lxlO7 JURKAT-Zellen (Zellbank des Deutschen KrebsforschungsZentrums, Heidelberg; Kultivierung bei 37°C in einem Inkubator in RPMI-1640 Medium, welches 10% Hitzeinaktiviertes fötales Kälberserum (Fa. Biochrom, Hamburg) , 25 M HEPES und 2 mM L-Glutamin (Fa. GIBCO/Life Technologies, Eggenstein, Deutschland) enthält) wurden in eiskaltem PBS gewaschen und anschließend in 400 μl Flag-Puffer (20 mM Tris, pH-Wert 7,5, 137 mM NaCl, 2 mM EDTA, 1% Triton X-100™, 10% Glycerin, 25 M ß-Glycerinphosphat, 2 mM Natriumpyrophosphat, 1 mm Na3V04, pH-Wert 10,0, 1 mM PMSF, 10 μg/ml Aprotinin, 10 μg/ml Leupeptin) lysiert. Die Lysate wurden durch Zentrifugation (20 min., 2°C, 13.000 UpM) geklärt und der Zellkern-freie Überstand, der nur die Proteine des Zytoplasmas und der Membranen enthielt, wurde in ein neues Gefäß überführt. Nach Bestimmung des Proteingehaltes wurden je 200 μg Protein mit je 1,5 μl polyklonalem IgG-Kaninchen-Antikörper gegen JNKl und JNK3 (C-17, sc-474; Fa. Santa Cruz, Heidelberg, Deutschland; gerichtet gegen die Aminosäuren 369 bis 384 des C-Terminus) oder gerichtet gegen JNKl, JNK2 und JNK3 (FL, Fa. Santa Cruz, Aminosäuren 1 bis 384 von JNKl) zugegeben und das Gemisch wurde 45 min. bei Raumtemperatur durch Rotation vermischt. Danach wurden 25 μl einer Suspension von Protein-A- Sepharose-Kügelchen (Sigma, Deisenhofen, Deutschland) zugegeben und das Gemisch wurde 1 Stunde bei Raumtemperatur unter Rotation inkubiert. Die Kügelchen wurden zur Entfernung unspezifisch gebundener Proteine zweimal mit Flag-Puffer gewaschen und einmal mit Kinase-Puffer . Die Kinase-Reaktion wurde in einem Gesamtvolumen von 50 μl in Gegenwart von nichtmarkiertem ATP (100 μm) und GST-c-Jun-Fusionsprotein (2 μg) durchgeführt, das die beiden N-terminalen Phosphorylierungsstellen Ser63 und Ser73 enthält, die für die Phosphorylierung durch JNK/SAPKs essentiell sind (Figur 1) . Dazu wurden die Protein-A-Sepharose-Kügelchen in 50 μl Kinase- Puffer suspendiert, der aus 25 mM HEPES, pH-Wert 7,4, 25 mM MgCl2, 25 mM ß-Glycerophosphat, 100 μM ATP (Röche, Mannheim, Deutschland), 0,1 mM Na3V04 und 2 μg GST-c-Jun 1-166 bestand, das wie von van Dam et al . , EMBO J. 14 (1995), 1798-1811, beschrieben hergestellt worden war, oder alternativ 2 μg GST- c-Jun 1-79 (Santa Cruz) . Nach Inkubation für 25 min. bei 37° wurde die Reaktion durch Zugabe von 30 μl 3 x SDS- Auftragspuffer (187,5 mM Tris-HCl, pH-Wert 6,8, 6% SDS, 30% Glycerin, 10% ß-Mercaptoethanol, 0,3% Bromphenolblau) beendet. Die Produkte wurden mittels 12% SDS-PAGE unter reduzierenden Bedingungen aufgetrennt und auf ECL-Membranen (Amersham, Braunschweig, Deutschland) übertragen. Phosphoryliertes GST-c- Jun-Protein wurde mit einem polyklonalen Kaninchen-IgG- phospho-spezifischen anti-c-Jun(Ser73) -Antikörper (New England Biolabs) angefärbt, der in einer Verdünnung von 1:5.000 verwendet wurde. Dieser Antikörper erlaubte die spezifische Bestimmung der JNK/SAPKs-induzierten Phosphorylierung von c- Jun an Ser73, wobei diese Stelle für die c-Jun-abhängige transkriptioneile Aktivität wichtig ist. Gebundene Antikörper wurden über anti-Kaninchen-Meerettichperoxidase-Konjugat (Santa Cruz, Heidelberg) in einer Verdünnung von 1:5.000 und anhand verstärkter Chemilumineszenz nachgewiesen. Die Spezifität des Assays wurde durch die Durchführung der Kinase- Reaktion in Abwesenheit oder Gegenwart von GST-c-Jun, Immunpräzipitats oder ATP bestimmt. Nur die Kinase-Reaktion, die alle Reagenzien zusammen enthielt, ergab ein starkes Signal (Figur 2) . Eine schwache Bande, die in der Reaktion ohne GST-c-Jun sichtbar ist (Spur a) dürfte durch die Phosphorylierung von endogenem Jun-Protein entstanden sein, das zusammen mit JNK/SAPKs präzipitierte. Die Doppelbande entspricht mono- und bi-phosphoryliertem Jun-Protein. Dieser Assay erforderte nur Entwicklungszeiten im Sekundenbereich und zeigte nur eine sehr geringe Hintergrundbindung. Er ist somit hochempfindlich und es kann davon ausgegangen werden, dass dieser Assay um Größenordnungen empfindlicher ist im Vergleich zu den bisherigen auf 32P-Markierung basierenden Assays.

Claims

Patentansprüche
1. Verfahren zur Bestimmung der Aktivität von JNK/SAPKs, wobei das Verfahren die folgenden Schritte umfaßt:
(a) Herstellung eines Zellysats oder Gewebelysats ;
(b) Aufreinigung von endogenen JNK/SAPKs;
(c) Durchführung einer Kinase-Reaktion mit den nach Schritt (b) erhaltenen JNK/SAPKs in Gegenwart von ATP und eine oder beide N-terminale Phosphorylierungsstellen enthaltendem GST- Transkriptionsfaktor-Protein;
(d) Nachweis der Menge an phosphoryliertem GST- Transkriptionsfaktor-Protein durch Zugabe eines spezifischen anti-p-GST-Transkriptionsfaktor- Antikörpers zu dem Gemisch aus Schritt (c) und Bestimmung der Aktivität der JNK/SAPKs anhand der Menge an gebundenem anti-p-GST-Transkriptionsfaktor- Antikörper .
2. Verfahren nach Anspruch 1, wobei in Schritt (b) die endogenen, im Zeil- oder Gewebelysat enthaltenen JNK/SAPKs über Immunpräzipitation aus dem Lysat isoliert werden.
3. Verfahren nach Anspruch 2, wobei die Isolierung der endogenen JNP/SAPKs dadurch erfolgt, dass diese mit spezifischen Antikörpern inkubiert und danach an Protein- A-Sepharose-Kügelchen gebunden werden.
4. Verfahren nach Anspruch 3, wobei die spezifischen Antikörper anti-JNKl-, anti-JNK2- und/oder anti-JNK3- Antikörper sind.
5. Verfahren nach einem der Ansprüche 1 bis 4, wobei der Transkriptionsfaktor (TCF)/Elk-1, c-Jun, Jun B, Jun D oder ATF-2 ist.
6. Verfahren nach einem der Ansprüche 1 bis 5, wobei in Schritt (c) beide N-terminale Phosphorylierungsstellen enthaltendes GST-c-Jun verwendet wird.
Verfahren nach Anspruch 6, wobei das GST-c-Jun GST-c-Jun 1-166 oder GST c-Jun 1-79 ist.
8. Verfahren nach einem der Ansprüche 1 bis 7, wobei in
Schritt (d) der Antikörper spezifisch die
Phosphorylierung an Ser73 und/oder Ser63 von c-jun erkennt .
Verfahren nach einem der Ansprüche 1 bis 8, wobei in Schritt (d) die Menge an gebundenem Antikörper über Western-Blot bestimmt wird.
10. Verfahren nach einem der Ansprüche 1 bis 9, wobei der Antikörper eine nachweisbare Markierung trägt.
11. Verfahren nach Anspruch 10, wobei die Markierung eine fluoreszente Verbindung, ein kolloidales Metall, eine chemilumineszente Verbindung, eine biolumineszente Verbindung, eine phosphoreszente Verbindung oder ein Enzym ist.
12. Verfahren zur Beurteilung der Wirksamkeit einer Krebstherapie, dadurch gekennzeichnet, dass die therapeutische Aktivierbarkeit der JNK/SAPKs in einer Probe gemäß einem Verfahren nach einem der Ansprüche 1 bis 11 gemessen wird.
3. Kit zur Durchführung des Verfahrens nach einem der Ansprüche 1 bis 12 , enthaltend ein eine oder beide N- terminale Phosphorylierungsstellen enthaltendes GST-c- Jun-Protein und einen anti-p-GST-c-Jun-Antikörper.
