Abstract

Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung N-substituierter 2-Pyrazolin-5-one der allgemeinen Formel (I), mit der in Anspruch 1 für R angegebenen Bedeutung, das dadurch gekennzeichnet ist, dass man eine Verbindung der allgemeinen Formel (II), mit den in Anspruch 1 für R, X und Y angegebenen Bedeutungen, bei erhöhter Temperatur mit einem molaren Überschuss Alkalihydroxid in einem wässrigen Reaktionsmedium umsetzt und anschliessend durch Zugabe einer Säure einen pH-Wert </= 6 einstellt.

Description

Verfahren zur Herstellung N-substituierter 2-Pyrazolin-5-one
Beschreibung
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung N-substituierter 2-Pyrazolin-5-one der allgemeinen Formel I
R worin
R für Ci-Cs-Alkyl, C5-C8-Cycloalkyl, Phenyl, Naphthyl oder Phe- nyl-Cχ-C4-alkyl steht, die gegebenenfalls einen oder mehrere gegenüber wässrigem Alkali inerte Substituenten aufweisen.
N-substituierte Pyrazolinone sind wertvolle Zwischenprodukte bei der Herstellung von Pharmazeutika und Pflanzenschutzmitteln. So beschreibt beispielsweise die US 4,557,753 die Herstellung von Benzoyl-substituierten 5-Benzyloxy-l-methylpyrazolen, die ausge- hend von l-Methylpyrazolin-5-on hergestellt werden.
In der Regel stellt man N-substituierte Pyrazolone durch Cycli- sierung von ß-funktionalisierten Säurederivaten mit substituierten Hydrazinen her. Eine Übersicht über verschiedene Synthesemethoden gibt die EP-A 240 001. In dieser Schrift wird außerdem ein Verfahren beschrieben, bei dem man ein Hydrazon eines ß-Hydrazinopro- pionsäurederivats in Gegenwart einer Base cyclisiert. Die vorgenannten Verfahren machen alle von substituierten Hydrazinen Gebrauch. Substituierte Hydrazine sind zum einen schwer zugänglich und daher teuer. Zudem sind substituierte Hydrazine in der Regel sehr giftig. Die Herstellung von N-substituierten Pyrazolinonen aus bereits bekannten oder anderweitig zugänglichen Pyrazolvor- stufen ist daher wünschenswert.
Dorn et al. (J. Pract. Chem. 313, 1971, S. 115-128) beschreiben ein Verfahren, bei dem man zunächst 3-Pyrazolidone am 1-Stick- stoff acyliert, anschließend am 2-Stickstoff mit Alkylchloriden oder Dialkylsulfaten alkyliert und dann unter oxidierenden Bedin- gungen die Acylgruppe wieder abspaltet. Hierbei werden N-substituierte 5-Hydroxypyrazole bzw. deren Tautomere 2-Pyrazolin-5-one erhalten. Das Herstellungsverfahren ist jedoch aufgrund der Vielzahl von Reaktionsschritten nicht wirtschaftlich. Außerdem sind Dialkylsulfate sehr toxisch.
Der vorliegenden Erfindung liegt die Aufgabe zugrunde, ein wirtschaftliches Verfahren zur Herstellung der eingangs definierten 2-Pyrazolin-5-one der allgemeinen Formel I bereitzustellen, bei dem man von bereits bekannten oder leicht zugänglichen Pyrazolen ausgeht.
Es wurde überraschenderweise gefunden, dass die Verbindungen der allgemeinen Formel I in guten Ausbeuten erhalten werden, wenn man 5-Halogenpyrazol-4-carbonsäuren oder deren basisch verseifbare Derivate bei erhöhter Temperatur mit wässrigen Alkalihydroxid-Lösungen umsetzt und anschließend die Reaktionsmischung ansäuert.
Demnach betrifft die vorliegende Erfindung ein Verfahren zur Her- Stellung N-substituierter 2-Pyrazolin-5-one der eingangs definierten Formel I, das dadurch gekennzeichnet ist, dass man eine Verbindung der allgemeinen Formel II
worin
für Halogen, und
Y für CN oder eine Gruppe der Formel R' (O)C stehen, worin R' eine Hydroxylgruppe oder einen mit Alkalihydroxid verseifba- ren Rest bedeutet,
und worin R die für Formel I genannten Bedeutungen aufweist,
bei erhöhter Temperatur mit einem molaren Uberschuss Alkalihydro- xid in einem wässrigen Reaktionsmedium umsetzt und anschließend durch Zugabe einer Säure einen pH-Wert < 6 einstellt.
Im Folgenden werden Verbindungen der allgemeinen Formel II mit
Y = R' (O)C und R' = OH auch als 5-Halogenpyrazol-4-carbonsäuren und die Verbindungen mit Y = CN oder Y = R'(0)C mit R' φ OH als
5-Halogenpyrazol-4-carbonsäurederivate bezeichnet. Die Verbindungen der Formel I stehen im Gleichgewicht mit den 5-Hydroxypyrazolen der Formel Ia
Das erfindungsgemäße Verfahren umfasst somit auch die Herstellung der Verbindungen Ia.
Organische Reste R', die unter basischen Reaktionsbedingungen, wie Alkalihydroxid, verseifbar sind, sind dem Fachmann bekannt. Beispiele für geeignete Reste R' sind C1-C4-Alkoxy, wie Methoxy, Ethoxy, n-Propoxy und n-Butoxy, die auch substituiert sein können, weiterhin Phenyloxy und Benzyloxy, die am Phenylring auch substituiert sein können. Beispiele für Substituenten sind Halogen, wie Fluor, Chlor oder Brom, weiterhin Nitro und C!-C4-Alkoxy. weitere basisch verseifbare Reste R' sind NH2 und Halogen. Bevorzugte basisch verseifbare Reste R' sind Cι-C4-Alkoxy, insbesondere Methoxy und Ethoxy. Sofern Y für eine Gruppe CN steht, die Verbindung II also ein Nitrilderivat ist, findet unter den Reaktionsbedingungen der ersten Stufe die Verseifung der Nitrilgruppe zur Carboxylgruppe statt. Das Gleiche gilt, wenn Y für R' (0)C steht und R' eine mit Alkalihydroxid verseifbare Gruppe bedeutet.
Besonders bevorzugte Verbindungen II sind die Carbonsäuren, d.h. Rest R' steht für Hydroxy (= OH). Ebenfalls bevorzugt sind Ver- bindungen II, worin Y für R'(0)C steht und R' Cι-C4-Alkoxy, insbesondere Methoxy oder Ethoxy bedeutet .
Erfindungsgemäß bevorzugtes Halogen X ist Chlor oder Brom, insbesondere Chlor.
Die Art des Substituenten R ist für die erfindungsgemäße Reaktion von untergeordneter Bedeutung. Die für R genannten Bedeutungen stellen Sammelbegriffe für individuelle Reste dar. Hierbei bedeuten:
Cι-C8-Alkyl: eine lineare oder verzweigte Alkylkette mit 1 bis 8 C-Atomen, z. B. Methyl, Ethyl, n-Propyl, iso-Propyl, n-Butyl, 2-Butyl, iso-Butyl, tert.-Butyl, n-Pentyl, 2-Methylbutyl, 2,2-Di- methylpropyl, n-Hexyl, 1-Methylpentyl, 2-Methylpentyl, 1-Ethylbu- tyl, 2-Ethylbutyl, n-Heptyl, n-Octyl und 2-Ethylhexyl. C3-C8-Cycloalkyl: mono- oder bicyclische Kohlenwasserstoffreste mit 3 bis 8 Kohlenstoffatomen, z. B. Cyclopropyl, Cyclobutyl, Cy- clopentyl, Cyclohexyl, Cycloheptyl, Cyclooctyl, Norbornyl und 2.2.2-Bicyclooctyl .
Phenyl-Cι-C4-alkyl steht für eine Cχ-C4-Alkylgruppe, die mit Phenyl substituiert ist, z. B. Benzyl, 1-Phenylethyl und 2-Phe- nylethyl.
Die vorgenannten Reste, sowie Phenyl und Naphthyl, können einen oder mehrere Substituenten aufweisen, die gegenüber wässrigen Alkalihydroxid-Lösungen inert sind. Beispiele für derartige Reste sind Cι-C4-Alkoxy, wie Methoxy, Ethoxy, Propoxy, n-Butoxy, tert.- Butoxy, weiterhin Trifluormethyl, Pentafluorethyl, Trifluorme- thoxy und Pentafluorethoxy. Darüber hinaus kommen für die Reste Cycloalkyl, Phenyl, Naphthyl und Phenyl-C1-C4-alkyl als Alkalistabile Substituenten auch Ci-C4-Alkyl in Betracht. Die drei letztgenannten Substituenten können an der Phenylgruppe bzw. an der Naphthylgruppe auch ein oder mehrere Chlor- oder Fluoratome als Alkali-inerte Substituenten aufweisen.
R steht vorzugsweise für Ci-Cs-Alkyl und insbesondere für Cι-C4-Alkyl, die vorzugsweise nicht substituiert sind, und besonders bevorzugt für Methyl oder Ethyl.
Die Ausgangsverbindungen der Formel II sind dem Fachmann bekannt und gut zugänglich (siehe EP-A 350176). Nicht bekannte Verbindungen der Formel II können beispielsweise in einfacher Weise durch Oxidation N-substituierter 5-Halogen-4-methylpyrazole nach dem in der EP-A 350 176 beschriebenen Verfahren hergestellt werden.
Im erfindungsgemäßen Verfahren setzt man in einem ersten Schritt eine Verbindung der allgemeinen Formel II mit Alkalihydroxid in molarem Uberschuss in einem wässrigen Reaktionsmedium um. Ein molarer Uberschuss an Alkalihydroxid ist bei den Verbindungen der allgemeinen Formel II dann gewährleistet, wenn man pro Mol Verbindung II mehr als 2 Mol Alkalihydroxid einsetzt. In der ersten Stufe werden ein Mol für den Austausch des Halogens X durch Hy- droxy und ein Mol für die Verseifung bzw. Neutralisation der
Gruppe C(0)R' benötigt. Erfindungsgemäß bevorzugt setzt man 3 bis 20 Mol Alkalihydroxid und insbesondere 5 bis 12 Mol Alkalihydroxid je Mol Verbindung II ein. Bevorzugte Alkalihydroxide sind Natriumhydroxid und Kaliumhydroxid, insbesondere Natriumhydroxid. Als wässrige Reaktionsmedien kommen sowohl Wasser als auch Gemische aus Wasser und wassermischbaren organischen Lösungsmitteln in Betracht. Die wassermischbaren organischen Lösungsmittel sind vorzugsweise gegenüber Alkalihydroxid unter Reaktionsbedingungen inert. Beispiele für geeignete organische Lösungsmittel sind
Cι-C4-Alkanole, insbesondere Methanol und Ethanol, weiterhin Di e- thylsulfoxid, Tetrahydrofuran, Dioxan, Glykol, Glycerin, Diethy- lenglykol, Triethylenglykol und vergleichbare. In der Regel wird das wässrige Reaktionsmedium nicht mehr als 50 Vol-%, vorzugs- weise nicht mehr als 30 Vol-% und insbesondere nicht mehr als 10 Vol-% eines organischen, wassermischbaren Lösungsmittels enthalten. In einer bevorzugten Ausführungsform der vorliegenden Erfindung ist Wasser alleiniges Lösungsmittel.
Besonders bevorzugt führt man den ersten Reaktionsschritt in einer wässrigen Alkalihydroxid-Lösung durch, die 10 bis 50 Gew.-% und insbesondere 20 bis 40 Gew.-% Alkalihydroxid enthält.
Erfindungsgemäß wird der erste Reaktionsschritt bei erhöhter Tem- peratur durchgeführt. Unter erhöhter Temperatur versteht man ein Erwärmen auf in der Regel wenigstens 50 °C und vorzugsweise wenigstens 90 °C. In der Regel wird die Reaktionstemperatur 200 °C nicht überschreiten. Ganz besonders bevorzugt führt man die Reaktion bei Temperaturen im Bereich von 120 bis 200 °C durch.
Je nach Reaktionstemperatur wird man den ersten Reaktionsschritt drucklos oder unter erhöhtem Druck durchführen. Bei Reaktionstemperaturen oberhalb 100 °C stellt sich in der Regel ein Reaktionsdruck von 1 bis 10 bar ein. Typische Reaktionsbedingungen sind beispielsweise bei einem rein wässrigen Reaktionsmedium 150 bis 180 °C und 5 bis 7 bar.
In der Regel wird man die Reaktion zu einem nahezu vollständigen Umsatz der Ausgangsverbindung II führen. Unter Umsatz versteht man hier die Umwandlung der Halogengruppe X im Pyrazol II in eine Hydroxylgruppe bzw. die Bildung des entsprechenden Alkoholats sowie die Verseifung der Nitrilgruppe (X = CN) oder der Gruppe -C(0)R', wenn R' Φ OH. Die Dauer zur Erreichung eines nahezu vollständigen Umsatzes hängt naturgemäß von den gewählten Reak- tionsbedingungen ab und kann zwischen 0,5 h und 24 h variieren. Typische Reaktionsdauern in rein wässrigen Systemen liegen in der Regel im Bereich von 2 bis 10 h.
Im zweiten Reaktionsschritt wird das im ersten Reaktionsschritt erhaltene Reaktionsprodukt unter sauren Bedingungen umgesetzt. Hierbei tritt unter C02-Entwicklung die Bildung der Verbindung I ein. Die C02-Entwicklung ist auf die Abspaltung der in der 4-Posi- ιf- if* K> t

xylierung die Reaktionsbedingungen noch einen gewissen Zeitraum, der wenige Minuten bis einige Stunden betragen kann, beibehalten.
Die Isolierung der Verbindung I erfolgt in üblicher Weise durch Aufarbeitung der Reaktionsmischungen des zweiten Reaktionsschrit- tes nach üblichen Aufarbeitungsmethoden, beispielsweise durch extraktive Aufarbeitung der flüssigen Reaktionsmischung mit einem organischen Lösungsmittel oder durch Entfernen des Lösungsmittels und Isolieren der Zielverbindung aus dem dabei erhaltenen Rück- stand. Vor der Aufarbeitung empfiehlt es sich, die Reaktionsmischung der zweiten Reaktionsstufe mit einer Base auf pH-Werte > 6, z.B. pH 6 bis 7, zu neutralisieren. Geeignete Basen sind Alkalihydroxide, Alkalicarbonate, Alkalihydrogencarbonate, Erdalka- licarbonate und Erdalkalihydroxide. üblicherweise wird man Alka- lihydroxide und insbesondere Natronlauge zur Neutralisation einsetzen.
Im erfindungsgemäßen Verfahren ist es aufgrund der anfallenden Salzfracht häufig vorteilhaft, zur Isolierung der Verbindung I das wässrige Reaktionsmedium der 2. Reaktionsstufe, vorzugsweise nach Neutralisation, destillativ oder durch Verdampfen im Vakuum weitgehend oder vollständig zu entfernen und Rückstand mit einem geeigneten organischen Lösungsmittel zu extrahieren. Dabei wird der Fachmann die Lösungsmittel so auswählen, dass das gewünschte Produkt in ihnen löslich ist, nicht jedoch die bei der Neutralisation anfallenden Salze. Typische organische Lösungsmittel zur Extraktion sind C2-C6-Alkohole, wie Ethanol, n-Propanol, iso-Pro- panol, n-Butanol, Isobutanol, Amylalkohol und Isoamylalkohol, aromatische Kohlenwasserstoffe wie Toluol, Ethylbenzol und Xy- lole. Nach Einengen des Extraktes zur Trockne fällt dann die ZielVerbindung I an und kann in üblicher Weise weiter gereinigt und aufgearbeitet werden.
Ebenso kann man das wässrige Reaktionsmedium der .2. Reaktions- stufe, vorzugsweise nach Neutralisation, extraktiv mit einem polaren, mit Wasser nicht oder nur eingeschränkt mischbaren Lösungsmittel aufarbeiten, beispielsweise durch Extraktion mit einem C -C6-Alkohol wie n-Butanol, Isobutanol, Amylalkohol oder Isoamylalkohol, oder mit einem der vorgenannten aromatischen Koh- lenwasserstoffe. Die Extraktion kann in portionsweise oder kontinuierlich durchgeführt werden.
Zur Erläuterung des erfindungsgemäßen Verfahrens wird im Folgenden eine typische Verfahrensvorschrift für die Umsetzung der Ver- bindungen II zu den 2-Pyrazolin-5-onen beschrieben: *» 4* O l to t in O Ul 0 Ul 0 Ul O l

Beispiel 2 (Herstellung von l-Ethyl-2-pyrazolin-5-on = 5-Hy- droxy-1-ethylpyrazol)
4 g 5-Chlor-l-ethyl-4-pyrazolcarbonsäure wurden in 40 g 25 gew.-%iger Natronlauge gelöst und analog der in Beispiel 1 beschriebenen Vorgehensweise umgesetzt. Die Reaktionstemperatur der ersten Reaktionsstufe lag bei 170 °C, der Reaktionsdruck bei 7,5 bar. Die Reaktionsdauer betrug 8 h. Bei Aufarbeitung in der für Beispiel 1 beschriebenen Weise erhielt man 2,3 g der Zielver- bindung mit einem Gehalt von 99,7 % (bestimmt mittels Gaschromatographie). Dies entspricht einer Ausbeute von 89,4 % der Theorie. Der Schmelzpunkt lag bei 88 °C. Das Produkt wurde durch Mischschmelzpunkt mit einer authentischen Probe identifiziert.
Beispiel 3 (Herstellung von l-Methyl-2-pyrazolin-5-on, Aufarbeitung durch flüssig-flüssig-Extraktion)
10 g 5-Chlor-l-methylpyrazol-4-carbonsäure wurden wie in Beispiel 1 zunächst mit 100 g 25 gew.-%iger Natronlauge und anschließend unter sauren Bedingungen umgesetzt. Nach Neutralisation der sauren Reaktionsmischung mit 25 gew.-%iger Natronlauge auf pH 6,5 wurde die Reaktionsmischung in einen flüssig-flüssig-Extraktor überführt und mit Isobutanol bei Siedetemperatur des Lösungsmittels extrahiert. Nach Isolierung der organischen Phase und Abde- stillieren des Isobutanols verblieben 5,8 g l-Methyl-2-pyrazoli- non zurück (Reinheit laut GC: 98,1 %). Der Schmelzpunkt lag bei 112 °C. Die Ausbeute betrug 92,5 % der Theorie.

Claims

Patentansprüche
1 . Verfahren zur Herstellung N-substituierter 2 -Pyrazolin-5-one der allgemeinen Formel I
worin
R für Cx-Cs-Alkyl, C5-C8-Cycloalkyl, Phenyl, Naphthyl oder Phenyl-Cχ-C4-alkyl steht, die gegebenenfalls einen oder mehrere gegenüber wässrigem Alkali inerte Substituenten aufweisen,
dadurch gekennzeichnet, dass man eine Verbindung der allge- meinen Formel II
worin
X für Halogen, und
y für CN oder eine Gruppe R' (O)C stehen,
worin R' eine Hydroxylgruppe oder einen mit Alkalihydroxid verseifbaren Rest bedeutet,
und R die für Formel I genannten Bedeutungen aufweist, bei erhöhter Temperatur mit einem molaren Uberschuss Alkalihydroxid in einem wässrigen Reaktionsmedium umsetzt und anschlie- ßend durch Zugabe einer Säure einen pH-Wert < 6 einstellt.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die Verbindung der allgemeinen Formel II mit wenigstens 3 Mol Alkalihydroxid, bezogen auf 1 Mol der Verbindung II umsetzt.
3. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass man die Umsetzung mit wässrigem Alkalihydroxid bei einer Temperatur oberhalb 90 °C durchführt.
4. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass die Zugabe der Säure bei einer Temperatur im Bereich von 0 bis 100 °C erfolgt.
5. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass Verbindungen der allgemeinen Formel II eingesetzt werden, worin
Y eine Gruppe R' (O)C bedeutet;
R' für Cχ-C -Alkyloxy oder OH steht; X für Chlor oder Brom steht; und
R eine der für Formel I genannten Bedeutungen aufweist.
6. verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass man die Umsetzung in einer wässrigen, 10 bis 50 gew.-%igen Natronlauge durchführt.
