Die Erfindung betrifft die Verwendung von an sich bekannten sowie neuartigen Derivaten der Maduransäure als Inhibitoren proinflammatorischer Cytokine zur Herstellung von Arzneimitteln für die Behandlung von durch diese Cytokine vermittelten Erkrankungen.The invention relates to the use of known and novel derivatives of maduranic acid as inhibitors of pro-inflammatory cytokines for the production of medicaments for the treatment of diseases mediated by these cytokines.

Das Immunsystem ist ein kompliziertes Netzwerk von Wechselwirkungen verschiedenartiger Zellen und deren Mediatoren miteinander. Die Mediatoren sind interzelluläre Signalmoleküle, die zum Beispiel das Wachstum, die Differenzierung und die Funktion von beteiligten Zellen regulieren. (K.F.Arai et al., Annu. Rev. Biochem. 59 (1990), 783) Eine wichtige Mediator-Gruppe sind die Cytokine, zu denen die Kolonie-stimulierenden Faktoren und die Interleukine gehören. Cytokine sind Polypeptide, deren vielfältige molekulare Charakteristiken, Wirkungsmechanismen, physiologische Funktionen sowie deren Rolle bei zahlreichen Erkrankungen gegenwärtig intensiv erforscht werden. So ist bekannt, daß eine Reihe von Cytokinen für die Steuerung der immunologischen Abwehrreaktionen gegenüber Pathogenen verantwortlich sind. Für die Identifizierung körperfremder Partikel oder Substanzen sowie für die Initiierung der Kaskade von Abwehrreaktionen sind B-Lymphozyten und T-Lymphozyten von entscheidender Bedeutung. (H.Holtmann, K.Resch, Naturwissensch. 82 (1995), 178) Für diese Zellen werden die Proliferation, die funktioneile Differenzierung und die Zellaktivität aber auch die Freisetzungsrate anderer Cytokine vor allem durch lnterleukin-2 (IL-2) und lnterleukin-4 (IL-4) geregelt. IL-4 steuert die T-Zell-Differenzierung zur vermehrten Bildung von Th2-Zellen, die bevorzugt IL-4 und IL-5 produzieren. (A.Mori et al., Intern. Pharm. 260 (1994), 247) Nebenwirkungen wie Erhöhung des Augeninnendrucks, erhöhte Infektionsanfälligkeit, Beeinträchtigung des hormoneilen Regelkreises (Osteoporose, bei Kindern Retardierung des Wachstums) können den Einsatz von Glukocortikosteroiden einschränken. Schließlich wurde auch für Cyclosporin A (CsA) eine Hemmwirkung auf die drei Cytokine gefunden. (B.Ryffel, Pharmacol. Rev. 41 (1989), 407) Auch in diesem Fall wurden verschiedene Nebenwirkungen (zum Beispiel Nephrotoxizität) festgestellt. Überraschender Weise wurde nun gefunden, daß sowohl die bereits bekannten, als auch zahlreiche neuartige Maduraphthalazine in der Lage sind, die Wirkung der Cytokine lnterleukin-2, lnterleukin-4 und lnterleukin-5 zu inhibieren. Demzufolge sind diese Verbindungen für die Herstellung von Arzneimitteln zur Therapie von durch diese Cytokine vermittelten Erkrankungen von großer Bedeutung. So wurde beispielsweise nachgewiesen, daß die erfindungsgemäßen Verbindungen in der Lage sind, die für die asthmatische late phase Reaktion charakteristische Einwanderung von eosinophilen Granulozyten in das Gewebe zu inhibieren.

Die Sensibilisierung erfolgt mit zwei subcutanen Injektionen einer Suspension von 10 μg Ovalbumin zusammen mit 1 mg Aluminiumhydroxid in 0,5 ml physiologischer Kochsalzlösung als Adjuvans im Abstand von 14 Tagen. Sieben Tage nach der zweiten Injektion werden die Tiere der Kontrollgruppe, die mit Ovalbumin vernebelt werden sollen, mit Mepyramin maleat (10 mg/kg i.p.) vorbehandelt, um vor dem anaphylaktischen Tod zu schützen.
