Abstract
Angegeben wird ein Verfahren zur Detektion und stufenlo
sen Quantifizierung von diagnostisch relevanten Substan
zen, wobei ein einziger Probenansatz für die Untersuchung
der Probe ausreicht. Als Test können ELISA, RIA, Nuklein
säure-Hybridisierung, Nephelometrie u. a. Bestimmungsme
thoden verwendet werden. Als diagnostsich relevante
Substanzen werden z. B. Antigene, Antikörper der ver
schiedenen Immunglobulinklassen, Nukleinsäuren oder
Stoffwechselprodukte von medizinischer Bedeutung nach
gewiesen. An einer Probe in der gewählten Testverdünnung
mit eingefärbten enzymatisch markierten Antigen-/Anti
körperkomplexen wird die Extinktion gemessen und aus dem
erhaltenen Signal AOD der Titer rechnerisch nach der
Formel:

ermittelt, der dem durch Verdünnungsreihen erhaltbaren
Endverdünnungstiter mit guter Genauigkeit gleichgesetzt
werden kann. Die Werte für alpha und beta werden in
getrennten Versuchsreihen experimentell ermittelt.
A method for the detection and stepless quantification of diagnostically relevant substances is specified, whereby a single sample preparation is sufficient for the examination of the sample. ELISA, RIA, nucleic acid hybridization, nephelometry and other determination methods can be used as the test. As diagnostically relevant substances z. B. antigens, antibodies of different immunoglobulin classes, nucleic acids or metabolic products of medical importance. The extinction is measured on a sample in the selected test dilution with colored enzymatically labeled antigen / antibody complexes and the titer is calculated from the signal A OD obtained using the formula:

determined, which can be equated with good accuracy to the final dilution titer obtainable by dilution series. The values for alpha and beta are determined experimentally in separate test series.
