Предложен способ перфузионного культивирования клеток, позволяющий в промышленных объемах продуцировать, в частности, терапевтические антитела, аффинные к С5-компоненту комплемента, и анти-IgE антитела. К особенностям осуществления способа относятся регулирование скорости протока питательной среды в реакторе, которое позволяет замедлить "старение" клеточной культуры, и регулирование отвода суспензии клеток из биореактора, что позволяет снизить уровень варьирования критических гликанов в полученных антителах.The invention relates to pharmaceutical biotechnology, more specifically to methods for producing therapeutic antibodies. A method of perfusion cell culture is proposed, which makes it possible to produce in industrial volumes, in particular, therapeutic antibodies affinity for the C5 component of complement, and anti-IgE antibodies. The features of the method include regulation of the flow rate of the nutrient medium in the reactor, which allows to slow down the "aging" of the cell culture, and regulation of the withdrawal of the cell suspension from the bioreactor, which makes it possible to reduce the level of variation of critical glycans in the obtained antibodies.
