Abstract
Изобретение относится к области медицины, в частности к клинической микробиологии, и может быть использовано для преодоления устойчивости грамотрицательных бактерий вида Klebsiella pneumoniae, продуцирующих металло-β-лактамазу (МβЛ), к карбапенемам. Указанный технический результат достигается тем, что клодроновая или этидроновая кислоты из группы бисфосфонатов ингибируют стандартный реактив - металло-β-лактамазу P.aeruginosa, рекомбинантную, экспрессированную в Escherichia coli. Достигнутый результат позволил выявить антимикробный эффект комбинации клодроновой или этидроновой кислот с карбапенемами (имипенемом или меропенемом) в отношении референс-штамма K.pneumoniae АТСС 70603, резистентного к карбапенемам, в присутствии стандартного реактива МβЛ, инактивирующего эти антибиотики. Предложенное техническое решение позволяет моделировать получение штаммов грамотрицательных микроорганизмов с различным уровнем резистентности к карбапенемам и оценивать эффективность ингибирования МβЛ.The invention relates to the field of medicine, in particular to clinical microbiology, and can be used to overcome the resistance of Gram-negative bacteria of the species Klebsiella pneumoniae, producing metallo-β-lactamase (ML), to carbapenems. This technical result is achieved by the fact that clodronic or etidronovy acid from the group of bisphosphonates inhibit the standard reagent - metallo-β-lactamase P. aeruginosa, recombinant, expressed in Escherichia coli. The achieved result allowed to reveal the antimicrobial effect of the combination of clodronic or etidronic acids with carbapenems (imipenem or meropenem) in relation to the reference strain K.pneumoniae ATCC 70603, resistant to carbapenems, in the presence of the standard МβЛ reactive inactivating these antibiotics The proposed solution allows to simulate the production of strains of gram-negative microorganisms with different levels of resistance to carbapenems and to assess the effectiveness of inhibition of MβL.
