Abstract
Изобретение относится к способу определения чувствительности микроорганизмов к химическим агентам или антибиотикам, заключающемуся в том, что микроорганизмы культивируют в жидкой питательной среде с химическим агентом или антибиотиком, затем среду обрабатывают таким образом, что может произойти лизис клеток и распределение фрагментов микроорганизмов по объему среды, а затем в не содержащем микроорганизмов супернатанте измеряют содержание белковых компонентов и сравнивают значения с контрольными или стандартными образцами. Метод включает добавление некоторого количества микробных клеток к жидкой питательной среде, в которую добавлен усиливающий агент, специфичный для определенной группы (рода или семейства) микроорганизмов. Жидкую питательную среду, содержащую микроорганизмы, преинкубируют, а затем, после добавления некоторого количества антибиотика, снова инкубируют. Затем клетки удаляют из инкубационной среды, получая супернатант. Белки супернатанта экстрагируют добавлением спирта с последующим выделением белков. Белковый осадок используют для получения масс-спектра, используемого для сравнения со стандартными масс-спектрами.
