Изобретение относится к рекомбинантной плазмидной ДНК с фрагментами каппа-казеина человека, рекомбинантному штамму Escherichia coli- продуценту пептида - аналога каппа-казеина человека, обладающего апоптотической активностью по отношению к опухолевым клеткам, и рекомбинантному пептиду - аналогу каппа-казеина человека, обладающему апоптотической активностью по отношению к опухолевым клеткам, и может быть использовано в биотехнологии. Рекомбинантная плазмидная ДНК pFK4 обеспечивает более высокий синтез рекомбинантного пептида RL4, являющегося аналогом фрагмента каппа-казеина человека, имеет молекулярную массу 2,41 MDa и размер 3653 п.о. Рекомбинантный штамм бактерий Escherichia coliXL1-Blue/pFK4 депонирован в Межинститутском Центре коллективного пользования СО РАН "Коллекция экстремофильных микроорганизмов и типовых культур" ИХБФМ СО РАН под № ЭМТК 1347 и является продуцентом пептида RL4 - аналога фрагмента каппа-казеина человека, обладающего апоптотической активностью по отношению к раковым клеткам. The recombinant plasmid DNA pFK4 provides a higher synthesis of the recombinant peptide RL4, which is an analogue of the human kappa-casein fragment, has a molecular weight of 2.41 MDa and a size of 3653 p. The Escherichia coliXL1-Blue / pFK4 recombinant bacterial strain was deposited at the Inter-Institute Center for Collective Use of the Siberian Branch of the Russian Academy of Sciences "Collection of Extremophilic Microorganisms and Type Cultures" IHBFM SB RAS under number No. EMTC 1347 and is a producer of peptide RL4 - an analogue of the human kappa-casein fragment; to cancer cells. The obtained recombinant peptide RL4 is an analogue of the human kappa-casein fragment, having a molecular weight of 10.2 kDa, consisting of a methionine residue, a fragment of human kappa-casein with 49 to 122 amino acid residues and a C-terminal histidine tract, and having apoptotic activity against cancer cells.
