Abstract
Изобретение относится к области клеточных технологий, а именно к анализу культивируемых in vitro клеток, и может быть использовано в биотехнологии, косметологии, медицине (клеточной трансплантологии), для получения охарактеризованного клеточного материала, обладающего лечебными или косметологическими свойствами. Способ определения качества клеточной культуры включает масс-спектрометрический анализ поверхностных клеточных белков. Предварительно промытые живые клетки исследуемой клеточной культуры подвергают витальному воздействию протеазы, отбирают отщепленные фрагменты поверхностных белков и масс-спектрометрически определяют их массы. Контролируют наличие качественных характеристик у исследуемой культуры клеток путем сравнения совокупности полученных масс с совокупностью масс-спектрометрически определенных масс фрагментов поверхностных белков живых клеток с известными качественными характеристиками. Технический результат, достигаемый при использовании патентуемого изобретения, заключается в повышении объективности (точности) идентификации культивируемых in vitro клеток для отбора качественных (с ожидаемыми полезными свойствами) клеток при снижении временных и материальных затрат.The invention relates to the field of cellular technology, namely the analysis of cultured in vitro cells, and can be used in biotechnology, cosmetology, medicine (cell transplantation), to obtain a characterized cellular material with therapeutic or cosmetic properties. The method for determining the quality of cell culture includes mass spectrometric analysis of cell surface proteins. The pre-washed living cells of the cell culture under study are subjected to the vital effect of protease, the split fragments of surface proteins are selected and their masses are spectrometrically determined. The presence of qualitative characteristics of the studied cell culture is monitored by comparing the aggregate of obtained masses with the aggregate of mass spectrometrically determined masses of fragments of surface proteins of living cells with known qualitative characteristics. The technical result achieved when using the patented invention is to increase the objectivity (accuracy) of the identification of cultured in vitro cells for the selection of high-quality (with the expected useful properties) cells while reducing time and material costs.
