Description
The invention relates to the pharmaceutical field and concerns the optimization of the composition of a composition containing a biologically active zinc complex with the peptide alloferon.
Known patent RU 2470031 (Kiselev O.I., Ershov F.I., publ.: 12.20.2012.) describing zinc-containing peptide complexes of the form
This solution was chosen as a prototype.
Complex compounds of peptides with zinc of this type are the main content of patent RU 2470031. However, the structural formula of the complexes proposed by the authors of the patent is based solely on general principles about the properties of zinc and histidine-containing peptides, as well as computer modeling data, being essentially purely theoretical. The complexes declared by the authors refer to a wide range of peptides, the molecules of which contain several (at least three) histidine residues.
The main object of consideration by the authors is the oligopeptide Alloferon (Alloferon-1 patent RU 2172322 Alloferons - immunomodulatory peptides Chernysh S.I., Kim Soo In, Becker G.P., etc.). However, the authors extended their conclusions not only to the entire extensive family of alloferons, but also to other peptides enriched with histidine. The authors of the patent suggested that histidine-containing peptides form chelate-type complexes with zinc, in which zinc is coordinately linked to histidine residues. However, the authors did not confirm the structural formula of the complexes, which is the main content of the invention, with any experimental evidence. The validity of a structural formula without its justification raises serious doubts. The a priori extension of the formula of the complexes and conclusions about their biological activity to a wide range of unstudied peptides also raises doubts.
From the text of the patent it follows that aqueous solutions were used to evaluate the biological properties of the complexes. However, it is known that in an aqueous environment the behavior of complex compounds is complex (O.V. Sergeeva. Reactions in aqueous solutions.
Complex ionic equilibria. Minsk, 2009). This circumstance does not allow us to draw unambiguous conclusions about the structure and composition of complexes based only on theoretical premises. Complex compounds are substances with a fundamentally unstable structure (Grinberg A.A., Introduction to the chemistry of complex compounds, 4th ed., Leningrad, 1971). In aqueous solutions they are always a mixture of different forms, the equilibrium between which is characterized by stability (instability) constants. For complexes of peptides or proteins, as weak ligands, this is expressed to an even greater extent (Inorganic Biochemistry Mir, M., 1978, vol. 1, pp. 152-162, section Complexes of metals with amino acids and peptides). For the alloferon-zinc system, this is aggravated by the fact that, firstly, zinc is a weak complexing agent, and secondly, alloferon has, along with histidine, other functional groups capable of coordination.
The authors of patent RU 2470031 have shown that the biological activity of alloferon complexes with zinc exceeds the activity of the original peptide. However, while providing data on the biological properties of the complexes, the authors of the patent did not provide any data on the method of their preparation. The description does not indicate which zinc salts were used to obtain the complexes, the method of their preparation and the permissible conditions (pH, solvents, ionic strength, additional components) under which these complexes exist are not given.
Thus, patent RU 2470031 is of a purely theoretical nature; it cannot be reproduced or used for practical purposes.
From a practical point of view, it is necessary to establish, firstly, the fact of the presence of alloferon complexes with zinc in an aqueous solution, and secondly, to find the conditions for their existence.
Complex compounds are products of the interaction of substances, one of which has the properties of an electron acceptor - a complexing agent, and the second is an electron donor - a ligand. When mixing two solutions containing metal ions (complexing agent) and a ligand (in this case, the ligand is alloferon), the process of complex formation will begin, i.e. addition of a ligand to a metal ion. The addition of the ligand to the metal occurs stepwise until the amount of the ligand becomes equal to the coordination number of the complexing agent (for zinc it is generally equal to 4). A dynamic equilibrium is established, in which both the formation of the complex and its disintegration occur. Thus, the solution, as a rule, simultaneously contains all
1971).- 1 046126 forms of complexes formed at various stages of complexation, as well as starting substances. Their proportion depends on the strength of the compounds formed (Grinberg A.A., Introduction to the chemistry of complex compounds, 4th ed., Leningrad, 1971).
The stability of complexes is determined by both fundamental factors (the nature of the complexing agent and ligands) and external conditions (temperature, nature of the solvent, ionic strength, solution composition). Among the fundamental factors influencing the stability of complexes are the nature of the central atom and the ligand, the structure of the ligand, and steric factors. Ligands with several binding sites (polydentate ligands) can form cyclic complexes - chelates, which are more stable.
Peptides, as ligands, have several metal binding sites, i.e. are polydentate ligands. Such sites can be: terminal amino and carboxyl groups, the amide nitrogen atom of the peptide bond, as well as side functional groups of amino acid residues. Therefore, peptides are characterized by the formation of chelate-type complexes.
Alloferon, along with the above active groups, contains 4 histidine residues - an amino acid containing an imidazole ring, which has a tendency to form complexes. This gives alloferon the properties of an active ligand. This circumstance is used in the above-mentioned patent RU 2470031. Since the coordination number of zinc is usually 4, and there are 4 histidine residues in the alloferon molecule, the formula of the chelate complex proposed by the authors of the patent RU 2470031 seems obvious. However, the presence of other functional groups in the alloferon molecule that can coordinate with metal ions suggests the possibility of the existence of other structures. The peculiarities of complex formation in an aqueous environment should also affect the composition and structure of the resulting compounds.
В работе T.KowalikJankowsca, L. Biega, M. Kuzer, D.Konopinska. Mononuclear Copper (II) complexes of alloferon 1 and 2: A combined potentiometric and spectroscopic studies. J. Inorg. Biochem. Complex compounds of alloferon and its derivatives with copper ions, formed in aqueous solutions, were carefully studied by a group of Polish scientists and described in a number of articles. In the work of T. Kowalik Jankowska, L. Biega, M. Kuzer, D. Konopinska. Mononuclear Copper (II) complexes of alloferon 1 and 2: A combined potentiometric and spectroscopic studies. J. Inorg. Biochem. 103, 2009, 135-142, the authors conduct a detailed analysis of the equilibrium in the alloferon - copper system (at their equimolecular ratios), based on potentiometric titration and spectrometry data. Copper and zinc as complexing agents have similar properties, so the data obtained for copper can be taken into account when studying alloferon complexes with zinc.
The work of Polish researchers has shown that in the alloferon-copper system in aqueous solutions many chelate-type compounds are formed, the composition and quantity of which very much depends on the pH of the medium. It was shown that at all pH values, copper ions and alloferon form complexes that are in equilibrium with the original components. It has been established that the copper ion forms a bond with histidine residues and certainly with the terminal amino group of alloferon. The number of copper bonds with histidine nitrogen depends on the pH of the medium and increases with increasing pH value. It can be associated with one, two or three histidine residues. The important thing is that one of the four histidine residues always remains free, not associated with a metal ion. This structure of the complexes differs from the formula given in patent RU 2470031, which considers binding specifically to 4 histidine residues. The authors of the Polish article showed that binding at 4 histidines is possible, but only if the terminal amino group is blocked, for example, by acetylation.
An important circumstance discovered by the authors of the article is that complex compounds of different composition and structure are simultaneously present in the copper alloferon system. Thus, polynuclear complexes were found in it, in which several metal ions are coordinated with one alloferon molecule. Thus, the alloferon - copper system in an aqueous solution has a complex composition; it simultaneously contains both complex compounds of various compositions and the starting substances - alloferon and copper salt.
Similar patterns should be observed for aqueous solutions of the alloferon-zinc system. However, it is important to keep in mind that zinc in the Irving-Williams series is inferior to copper in complex-forming activity (O.V. Sergeeva. Reactions in aqueous solutions. Complex ionic equilibria. Minsk, 2009, 70 pp.). Therefore, zinc complex compounds are usually less stable than copper ones. However, it can be assumed that the nature and patterns of existence of complex compounds in the alloferon-zinc system will be close to those described above for copper.

Carrying out the invention
During the development, zinc acetate and zinc chloride were used as zinc salts. Only these zinc salts are included in drugs for parenteral administration, primarily in various forms of insulin. The solvent used was 0.9% solutions of sodium chloride and sodium acetate in water. Other isotonic solutions (Ringer's solution, etc.) are not applicable in this case, because additionally contain calcium ions, as well as phosphates and carbonates, which form insoluble compounds with zinc.
Solutions containing alloferon and zinc salts were studied using potentiometric titration and mass spectrometry.
RU 2470031.It has been established that in the system alloferon - zinc salt in an aqueous environment the process of complexation occurs. In this case, the solution always contains an equilibrium mixture of the initial components - alloferon and zinc salt, as well as the complex compounds they form. It was found that zinc and alloferon in solution form complex compounds of various compositions. Complexes are predominantly formed in which the alloferon molecule is associated with one zinc ion. However, along with them, the solution contains complexes in which two zinc ions are bound to an alloferon molecule, as well as dimers in which one zinc ion is bound to two alloferon molecules. Thus, in the alloferon - zinc system in an aqueous solution there is a complex mixture of substances, including initial components and complexes of various structures and compositions. It is obvious that the composition and structure of these complexes differ significantly from the formula given in patent RU 2470031.
It was found that the composition and concentration of the formed complexes depend on pH, the ratio of allo
An increase in pH increases the concentration of complexes. The same effect is achieved by increasing the amount of zinc salt above the equimolecular one. The use of 0.9% solutions of sodium chloride or sodium acetate with a pH in the range from 6.5 to 8.0 as a medium made it possible to obtain a composition in which the degree of complexation is close to quantitative.
Композиция получается путем смешения равных объемов растворов ацетата или хлорида цинка и аллоферона в 0,9% растворе натрия хлорида или натрия ацетата, с последующей корректировкой рН до указанной величины 6,5-8,0 (предпочтительно 7,5).As a result, the developed composition includes complex compounds of alloferon and zinc of various structures, initial alloferon and zinc salt (acetate or chloride), 0.9% solution of sodium chloride or sodium acetate and has a pH in the range from 6.5 to 8.0, mainly 7 ,5. In this case, to prepare the composition, alloferon and an equivalent or greater amount of zinc salt are taken. The composition is obtained by mixing equal volumes of solutions of zinc acetate or chloride and alloferon in a 0.9% solution of sodium chloride or sodium acetate, followed by adjusting the pH to the specified value of 6.5-8.0 (preferably 7.5).
The biological activity of the claimed compositions was confirmed by experiments. As comparison drugs, we used the drug Allokin-alpha, a lyoflysate for the preparation of a solution for subcutaneous administration, containing 1 mg of pure alloferon without excipients, as well as the standard antiherpetic drug Acyclovir. The antiviral activity of the developed composition, shown previously (patent RU 2470031) using the example of the influenza virus, was verified by tests with the herpes virus. The test result confirmed the data of patent RU 2470031, showing higher antiviral activity of the composition in comparison with alloferon. The immunomodulatory properties of the developed composition, shown by the example of the induction of interferon gamma and interleukin-18, are also higher than those of the pure alloferon peptide.

Ион с m/z 1265,5896 - однозарядный ион, ион с m/z 633,2992 является двухзарядным ионом [М+2Н]2+ и трехзарядный ион [М+3Н]3+ с m/z 422,5351.In fig. The ion with m/z 1265.5896 is a singly charged ion, the ion with m/z 633.2992 is a doubly charged [M+2H] 2+ ion, and the triply charged ion [M+3H] 3+ with m/z 422.5351.
In fig. Figure 2 shows the mass spectrum of a 0.001 M solution containing alloferon and zinc salt at a molar ratio of alloferon - zinc of 1:1. Comparison of the spectra of Fig. 1 (alloferon) and Fig. 2 (composition) shows that signals of new ions appeared in the system, the presence of which can only be explained by the formation of complex compounds of alloferon with zinc. The low intensity of the signals of new ions indicates that the content of free alloferon in the solution is significantly higher than the concentration of the complex.
C52H76N22O16Zn.Spectrum Fig. 2 contains a signal with m/z 443.1731, corresponding to the triply charged ion of the zinc complex [M+Zn+H] 3+ , as well as a signal with m/z 664.2556, which corresponds to the doubly charged ion of the [M+Zn] 2+ complex. Both signals correspond to a complex with the gross formula C 52 H 76 N 22 O 16 Zn.
Такое же совпадение теории и практики было получено и для двухзарядного иона.- 4 046126 shim than 1:2 were not studied. Shown in FIGS. 4 and fig. 5, the real and theoretical mass spectra of the triply charged ion of the complex practically coincide, which indicates the correct interpretation of this signal in the mass spectrum as corresponding to a complex with the gross formula C 52 H 76 N 22 O 16 Zn. The same agreement between theory and practice was obtained for a doubly charged ion.
C104H152N44O32Zn.In addition, the spectra contain signals with m/z 465.1425 and 697.2097. They correspond to a complex with the gross formula C 52 H 72 N 22 O 16 Zn 2 , in which alloferon is bonded to 2 zinc atoms at once. A comparison of the real and theoretically calculated mass spectrum of the triply charged ion with m/z 465.1425 confirms this identification. The presence of a triply charged ion with m/z 886.6705, corresponding to an alloferon dimer with zinc C 104 H 152 N 44 O 32 Zn, is also observed.
Thus, mass spectrometric research shows that in an aqueous environment, alloferon in combination with zinc salts forms complex compounds of various forms and compositions.
6.Despite the existing technical limitations, a mass spectrum of the alloferon zinc system in saline solutions was obtained. In fig. Figure 6 shows the mass spectrum of the composition in a 0.9% sodium chloride solution. The intensity of doubly and triply charged signals with m/z 664.2558 and 443.1728 shows that in an isotonic solution a high content of alloferon complexes with zinc of the composition C 52 H 76 N 22 O 16 Zn is achieved. The signal intensity of the doubly charged ion of the complex with m/z 664.2558 in the spectrum of Fig. 6 is significantly higher than for the alloferon ion m/z 633.29776, and the triply charged one m/z 443.1721 is at its level (m/z 422.53426). The mass spectrum of the composition in sodium acetate solution is similar to Fig. 6.
The dependence of the content of complex compounds in a solution of alloferon with zinc on the pH value of the solution was studied.
The results (Fig. 7-9) show that the content of complex compounds of alloferon with zinc depends on the pH value. As the pH value increases in the range from 4 to 8, an increase in the intensity of the corresponding signals is observed in the mass spectra of aqueous solutions of complex compounds of zinc and alloferon, which may indicate an increase in their concentration in the solution due to a change in pH.
The maximum intensity of the doubly charged zinc complex m/z 664.2558 is observed at pH 8. The signal of the triply charged ion of the complex is absent at this pH. The above results of mass spectrometric studies have most likely shown that:
аллоферон с солями цинка в водном растворе образует комплексные соединения;alloferon forms complex compounds with zinc salts in aqueous solution;
в водном растворе одновременно присутствуют комплексы аллоферона с цинком различного состава и строения, а также свободный аллоферон;in an aqueous solution, complexes of alloferon with zinc of various compositions and structures, as well as free alloferon, are simultaneously present;
концентрация комплексов зависит от рН раствора, с его ростом их концентрация увеличивается с максимумом при рН 8,0;the concentration of complexes depends on the pH of the solution; as it increases, their concentration increases with a maximum at pH 8.0;
концентрация комплексов зависит от соотношения аллоферон - цинк и ионной силы раствора.the concentration of the complexes depends on the ratio of alloferon - zinc and the ionic strength of the solution.
8,0.Conclusion: the composition with the maximum concentration of complex compounds includes alloferon, zinc salt in an amount equal to or greater than equimolecular in a 0.9% solution of sodium chloride or sodium acetate with a pH from 6.0 to 8.0.
Example 2. Potentiometric titration.
ZnCl2.The solutions under study had a concentration of 0.001 M for alloferon and zinc at a molar ratio of 1:1. Solvent -0.9% sodium chloride solution. The volume of titrated solutions was 30 ml. To prepare solutions, zinc acetate dihydrate Zn(CH 3 COO) 2 *2H 2 O and anhydrous zinc chloride ZnCl 2 were used.
For the work, a potentiometric automatic titrator ATP-02 was used, which makes it possible to obtain a titration curve in integral and differential forms. Titrations were performed of both the composition of alloferon with zinc salts and solutions of the initial components - alloferon, acetate and zinc chloride. Titration was carried out separately in the acidic and alkaline regions. Each titration was carried out three times. The average titration data are presented in integral (Fig. 10) and differential (Fig. 11) form.
The titration curve of alloferon has two clearly defined jumps at pH 4.75 and 9.4, which quantitatively precisely correspond to the presence of 4 histidine residues and one terminal COOH group.
The titration curves for mixtures of alloferon + zinc salt are of a significantly different nature. Firstly, the introduction of zinc salt into the alloferon solution significantly (from 7.5 to 6.5) changes the pH value of the solution. This circumstance alone suggests the presence of interaction between the components in the system under study.
Secondly, the jumps in the titration of solutions of the compositions in comparison with the curve for alloferon are greatly blurred (the amplitude of the dE/dV jump on the differential curves is sharply reduced). Thirdly, the curves contain new titration jumps. These differences cannot be described by simply adding the titration curves of pure alloferon and pure solutions of zinc salts (Fig. 10). The differential curves (Fig. 11 and in more detail Fig. 12) show this more clearly. These circumstances clearly indicate the presence of interaction between alloferon and zinc salts. Such an interaction can only be complex formation. However, the small amplitude of titration jumps indicates a weak connection of alloferon with zinc.
- 5 046126- 5 046126
In the acidic region, the disappearance of the jump at pH 4.75, characteristic of alloferon, and the appearance of a jump at pH 4.25 are observed. There is also a new, but very weak, jump at pH 6.5, more pronounced in the case of zinc acetate.
The most important changes take place in the alkaline region. Thus, there is practically no jump at pH 9.4, which is characteristic of both alloferon and zinc salts. This indicates that the content of free alloferon and zinc ions in the solution in the alkaline region is not high. Therefore, they are mainly found in a complex compound with each other. In this case, two new jumps appear: one at pH 8.0, the second around 10.0, which can be interpreted as jumps in the titration of various forms of the complex. In the alkaline region at high pH values (above 8.0), the presence of complexes with deprotonated forms of alloferon is possible.
The most interesting is the narrow region between the jumps at pH 6.5 and 8.0. In this region, an uncharged alloferon molecule can interact with zinc salts to form a chelate-type structure.
The data obtained show that:
аллоферон и соль цинка в растворе вступают во взаимодействие друг с другом, взаимодействие аллоферона и солью цинка с образованием комплексных соединений наблюдается во всем диапазоне рН от 4,0 до 10,0, область рН от 6,5 до 8,0 является предпочтительной для фармацевтических целей, данные потенциометрического титрования подтверждают закономерности, выявленные при массспектрометрических исследованиях (пример 1) системы аллоферон - цинк.alloferon and zinc salt in solution interact with each other, the interaction of alloferon and zinc salt with the formation of complex compounds is observed over the entire pH range from 4.0 to 10.0, the pH range from 6.5 to 8.0 is preferable for pharmaceuticals purposes, potentiometric titration data confirm the patterns identified during mass spectrometric studies (example 1) of the alloferon - zinc system.

In order to assess the immunomodulatory activity (IA) of the claimed compositions, as well as to identify the contribution of the complex compounds contained in them, the level of induction of cytokines (IFN-γ and IL-18) was determined both by the compositions and the initial components - alloferon and zinc salt (acetate) . In addition, the induction of cytokines caused by the use of both components (alloferon and zinc salt), introduced into the body separately over time, was determined.

1.- 6 046126 the maximum level of IFN-γ induction is observed. To determine the induction of interleukin-18, the method described in patent RU 2482866 was used; the time point for determining the level of IL-18, based on the data of this work, was 36 hours after the introduction of the sample. For the control group, the confidence interval was calculated from dilution measurements of the same sample. In the experimental groups, the confidence interval was calculated relative to three samples from three animals for each measured point. The results of testing the samples under study are given in table. 1.

The protective effect of the developed compositions was assessed on a model of herpes infection. The drugs used for comparison were: 1/ Allokin-alpha, a lyophilisate for preparing a solution for subcutaneous administration - a drug used in the treatment of genital herpes, containing 1 mg of alloferon per ampoule, 2/ Acyclovir - a drug used in standard therapy of herpes.
White outbred mice (females) weighing 16-20 g were obtained from the Rappolovo nursery (Leningrad region) and kept on a standard diet in regulated vivarium conditions. The selection of animals into experimental groups was carried out using random sampling. The animals were divided into 5 groups of 20 animals: four experimental ones, in which the animals received injections of composition 1, composition 2, the drug Allokin-alpha, the drug Acyclovir, respectively, and one control, in which the animals received injections of 0.9% sodium chloride solution .
The study used herpes simplex virus type 2 (HSV-2) strain G (ATCC VR-734). Infection was carried out vaginally. Damage to the vaginal epithelium was carried out using a scarifier for vaginal smears, after which the animals were infected with the virus at a dose of 1.7x105 TCID50 in a volume of 30 μl.
Allokin-alpha and compositions 1 and 2 were administered to animals intraperitoneally at a dose of 1.25 mg/kg once a day for 3 days after infection according to the treatment regimen: 1, 2 and 3 days after infection. Acyclovir was administered according to the same regimen at a dose of 50 mg/kg. The control group of animals received an injection of 0.9% sodium chloride solution.
The animals were observed for 14 days.
