They are produced by the thyroid gland in the form of thyroxine (T4) and 3,5,3'-triiodo-L-thyronine (T3). THs form part of the thyroid axis, also known as the hypothalamic-pituitary-thyroid axis, or HPT axis, which includes a complex endocrine and paracrine feedback loop linking brain and endocrine system tissue to exert global control of parameters such as metabolic rate, lipid secretion, cardiac function, muscle and bone growth, and many others (see, for example, Robins and Cotran: Pathologic Basis of Disease, Kumar, V. et al., eds. Subjects are randomly assigned to one of seven separate treatment groups: daily oral placebo (PO); loading dose of compound 1250 mg followed by daily dose of compound 15 mg PO; a loading dose of compound 1,250 mg followed by a daily dose of compound 1 of 10 mg PO; loading dose of Compound 1,250 mg followed by every other day (QOD) dose of Compound 1 10 mg PO; a compound loading dose of 1,100 mg followed by a daily compound dose of 5 mg PO; a loading dose of Compound 1 of 100 mg followed by a daily dose of Compound 1 of 10 mg PO; and a loading dose of Compound 1 of 100 mg followed by an every other day (QOD) dose of Compound 1 of 10 mg. Compound 1 is provided in the form of a tablet containing either 5 mg or 10 mg of Compound 1.
