Abstract

Previously unknown, rapidly acting human insulin analogues having a lower tendency for self-association into dimers, tetramers, hexamers or polymers. The previously unknown insulin analogues are formed by replacing one or more of the amino acid residues in human insulin with naturally occurring amino acid residues. The amino acid residue replacements are preferably more hydrophilic than the natural amino acid residue at the position in question in the molecule. Furthermore, the insulin analogue has the same charge as or a greater negative at neutral pH than human insulin. Preferred amino acid replacements are Asp, Glu, Ser, Thr, His and Ile, and more preferred replacements are Asp and Glu. The previously unknown insulin analogues can be used for preparing rapidly acting insulin solutions.
