Claims (15)
- BESTEMMELSE AF KONCENTRATIONEN AF EN BESTANDDEL I ET FLUID FRA ET DYR VED SPEKTROSKOPISK ANALYSE AF ET ANDET FLUID PATENTKRAV 1Fremgangsmåde til kalibrering af et infrarødt spektroskopiapparat beregnet til at frembringe spektre med henblik på bestemmelse af koncentrationen af en bestanddel C1 i et fluid F1 fra et dyr, hvilken fremgangsmåde er kendetegnet ved, at den omfatter følgende trin: a) - opnåelse af prøver af fluidet F1 og af et fluid F2, der er et andet end fluidet F1, fra et dyr, der hører til en gruppe af repræsentative dyr, hvilket F2 indeholder detekterbare mængder af mindst én bestanddel C2, der er et produkt eller et direkte eller indirekte medprodukt af metabolismen af bestanddelen C1, b) - måling af koncentrationen af bestanddelen C1 i fluidprøverne F1 ved hjælp af en referencefremgangsmåde og registrering af de opnåede referenceværdier i en centralenheds hukommelse, c) - frembringelse af det komplette IR-absorptionsspektrum for fluidprøverne F2 i frekvensintervallet fra 650 cm'1 til 4000 cm'1 og registrering af de opnåede spektre i centralenhedens hukommelse, d) - beregning af korrelationskoefficienten for hver bølgelængde af de opnåede absorptionsspektre i c) mellem absorbansværdierne for fluidprøverne F2 og referenceværdierne for bestanddelen C1 og identifikation af spektrale intervaller, for hvilke absorbansværdierne korreleres med referenceværdierne for koncentrationen af bestanddelen C1 i fluidprøverne F1, og e) - beregning på basis af mindst ét af de spektrale intervaller af absorptionsspektrene for fluidprøverne F2 afen matematisk model, der kan forudsige koncentrationen af bestanddelen C1 i fluidet F1.
- 2. - Fremgangsmåde ifølge krav 1, kendetegnet ved, at, i trin d), de korrelerede spektrale intervaller identificeres ved hjælp af multivariate regressionsalgoritmer i frekvensintervallet fra 650 cm'1 til 4000 cm'1.
- 3. - Fremgangsmåde ifølge et af de foregående krav, kendetegnet ved, at prøverne af fluiderne F1 og F2, som opnås i trin a), stammer fra prøver taget samtidigt fra hvert dyr af en gruppe af repræsentative dyr.
- 4. - Fremgangsmåde ifølge et af de foregående krav, kendetegnet ved, at den omfatter følgende supplerende trin: f) - søgning blandt de bestanddele, der kan være til stede i fluidet F2, efter mindst én bestanddel C2, der mindst har én karakteristisk absorptionsfrekvens, der falder sammen med ét af båndene i det spektrum, for hvilket der eksisterer en korrelation, g) - bestemmelse af koncentrationen af den mindst ene bestanddel C2 i hver af fluidprøverne F2, h) - beregning af korrelationsforholdet R1, der eksisterer mellem i) koncentrationerne af den mindst ene bestanddel C2 i fluidet F2 og ii) koncentrationerne af bestanddelen C1 i fluidet F1.
- 5. - Fremgangsmåde ifølge et af de foregående krav, kendetegnet ved, at den omfatter følgende supplerende trin: f)- søgning blandt de bestanddele, der kan være til stede i fluidet F2, efter n bestanddele C2n, der hver har mindst én karakteristisk absorptionsfrekvens, der falder sammen med mindst ét af båndene i det spektrum, for hvilket der eksisterer en korrelation, g')- bestemmelse af koncentrationen af de n bestanddele C2n i hver af fluidprøverne F2, h')- beregning af værdien af en indikator le, der er proportionel med koncentrationen af mindst én af de n bestanddele C2n, h")-beregning af korrelationsforholdet R2, der eksisterer mellem i) værdierne er indikatoren le i fluidet F2 og ii) koncentrationerne af bestanddelen C1 i fluidet F1.
- 6. - Fremgangsmåde ifølge et hvilket som helst af de foregående krav, kendetegnet ved, at fluidet F1 er blod fra et sundt eller sygt dyr, og fluidet F2 er et andet fluid fra det sunde eller syge dyr, der er valgt blandt mælk eller urin.
- 7. - Fremgangsmåde ifølge et af de foregående krav, kendetegnet ved, at bestanddelen C1 er et stof, hvis koncentration i fluidet F1 er en direkte eller indirekte indikation for et metabolisk problem eller en særlig metabolisk tilstand.
- 8. - Fremgangsmåde ifølge det foregående krav, kendetegnet ved, at bestanddelen C1 er et ketonlegeme, en fedtsyre, et carbohydrat, et protein, et glycoprotein eller et hormon.
- 9. - Fremgangsmåde ifølge det foregående krav, kendetegnet ved, at bestanddelen C1 er BHB (β-hydroxybutyrat), der er til stede i blodet hos et sundt dyr i en tilstand af energiunderskud eller en tilstand af acetonæmi.
- 10. - Fremgangsmåde ifølge et af kravene 4 til 9, kendetegnet ved, at den mindst ene bestanddel C2 er en bestanddel, der er kendt for at have eller nyligt er identificeret ved afslutning af trin f) som havende en direkte eller indirekte relation med en metabolisk vej for bestanddelen C1.
- 11. - Fremgangsmåde ifølge et hvilket som helst af de foregående krav, kendetegnet ved, at den mindst ene bestanddel C2 er en bestanddel, der er tilstede i mælken fra et sundt dyr i en tilstand med energiunderskud eller acetonæmi, valgt blandt fedtsyrer, proteiner, glycoproteiner, carbohydrater.
- 12. - Fremgangsmåde ifølge det foregående krav, kendetegnet ved, at den mindst ene bestanddel C2 er en umættet, ikke-esterificeret fedtsyre, fortrinsvis oleinsyre eller stearinsyre.
- 13. - Fremgangsmåde ifølge krav 5, kendetegnet ved, at indikatoren le er defineret som forholdet mellem indholdet af en fedtsyre eller en fedtsyreklasse og proteinindholdet.
- 14. - Fremgangsmåde til bestemmelse af koncentrationen af en bestanddel C1 i et fluid F1 fra et dyr ved hjælp af et spektroskopiapparat, der er egnet til at frembringe absorptionsspektre, kendetegnet ved, at den omfatter: - anvendelse af en fremgangsmåde til kalibrering af spektroskopiapparatet ifølge et af kravene 1 til 13, hvilken model forudsiger koncentrationen C1 ud fra et spektrum for en prøve af et fluid F2fra dyret, der er et andet end fluidet F1, hvilket fluid F2 indeholder detekterbare mængder af mindst én bestanddel C2, som er et produkt eller et direkte eller indirekte medprodukt af metabolismen af bestanddelen C1, - frembringelse af et spektrum for en prøve af fluidet F2 fra dyret og registrering af prøvens spektrale informationer, - anvendelse af den matematiske model til de spektrale informationer for fluidprøven F2 til bestemmelse af koncentrationen af bestanddel C1 i fluidet F1.
- 15. - Fremgangsmåde ifølge det foregående krav, hvor koncentrationen af BHB bestemmes i blodet fra et dyr ved anvendelse af den matematiske model til de spektrale informationer, der er opnået ud fra en mælkeprøve fra dyret.
