Abstract
A protein-like, physiologically active substance having an antineoplastic effect, which substance is induced by administering at least substance which can stimulate the reticuloendothelial system to a rabbit and then injecting the rabbit with endotoxin from a Gram-negative bacterium, is purified. The process comprises bringing a crude solution of the protein-like, physiologically active substance into contact with a basic ion exchanger, with the physiologically active substance being adsorbed with the ion exchanger, eluting the adsorbed physiologically active substance and subjecting the eluate, which contains the physiologically active substance, to gel filtration using a gel which is suitable for separating a substance having a molecular weight in the range of 30,000-70,000. The resulting purified preparation of the physiologically active substance can be used as an antineoplastic agent for treating malignant tumours. The physiologically active substance is isolated by subjecting the purified preparation to affinity chromatography on immobilized Cibacron Blue F3G-A, gel filtration, affinity chromatography and immobilized concanavalin A, preparative polyacrylamide gel electrophoresis and gel filtration, in the given order.
