Description
Process for cleaning the antibiotic 106 - 7 The invention relates to a process for cleaning the antibiotic io6-7-It has been found that the growth of the microorganism known under the name Streptomyces garyphalus in suitable fermentation media is a valuable product known as "antibiotic io6-7" produces known antibiotic which is an effective therapeutic agent against gram negative and gram positive organisms.
This antibiotic and process for its production by fermentation are described in detail in German Patent 933 053 .
The antibiotic io6-7 is an amphoteric substance that is both acidic as well as basic properties. So this antibiotic z. B. salts through Reaction with bases. For example, by reacting this product with bases such as sodium hydroxide, potassium carbonate, calcium hydroxide and magnesium hydroxide the sodium, Potassium, calcium and magnesium salts of the antibiotic io6-7 are formed. Acid addition salts this antibiotic, such as the hydrochloride, the sulfate and the like., Are by addition the corresponding acid to an aqueous solution of the base of the antibiotic.
The isolation and purification of the antibiotic io6-7 has been particularly difficult because this product does not respond to the methods commonly used to isolate and purify antibiotics, such as adsorption on charcoal or aluminum oxide or extraction with common solvents such as ether and butanol. A method for isolating and for achieving substantial purification of this antibiotic is described in German patent specification 933 053 . According to this method, the aqueous solution of the antibiotic io6-7 is treated with a strongly acidic cation exchange resin to adsorb the antibiotic, and the antibiotic is then eluted from the adsorbate with an aqueous solution of a base. A product of considerably greater activity can be obtained from the basic eluate obtained in this way, the effectiveness of which is up to about thirty times that of the starting material.
The present invention is a new method for further cleaning of the antibiotic io6-7.
In the context of the present invention it has been found that strongly basic anion exchange resins in their hydroxyl form react with the antibiotic io6-7 to form complexes of antibiotic and resin, which provides a valuable means of isolating and purifying this antibiotic. Thus, when a solution of the impure antibiotic io6-7 is brought into intimate contact with a strongly basic, anion-exchanging quaternary ammonium resin in its hydroxyl form, the antibiotic is preferentially adsorbed to form a resin complex and can be removed from the complex by elution with an anion-forming solution, e.g. B. the solution of an acid, a base or a salt can be recovered. If this method is applied to a solution of the antibiotic io6-7, which was obtained by adsorbing the antibiotic from a Fennentationsbrühe on a strongly acidic, cation-exchanging resin and washing out the resin adsorbate in an aqueous solution of a base according to the German patent 933 053 so one can achieve a substantial purification of the antibiotic. By carrying out the method according to the invention, a crystalline antibiotic io6-7 with an effectiveness of about 225 to 300 units / mg can easily be obtained from the aqueous eluates.
The resins which can be used according to the process according to the invention are strongly basic, anion-exchanging quaternary ammonium resins whose exchangeability is based on the quaternary amine groups present in the polymer. Resins which can be used in the context of the invention are available commercially under the names Amberlite IRA 40o, Amberlite IRA 410, Amberlite XE 98, Amberlite XE 75, Amberlite XE 58, Permutit S, Ionac A 293 and Dowex 2. These resins are converted to their hydroxyl form by washing with an alkaline solution such as aqueous sodium hydroxide. After the resin thus obtained has been washed out with water to remove the excess alkali, the resin is suitable for use in the process according to the invention.
This adsorption of the antibiotic can take place from solutions which contain the product in various concentrations. In general, however, the best results are achieved if an aqueous solution is used as the starting material which contains the antibiotic io6-7 in partially purified form, such as. B. the solution obtained by adsorbing the antibiotic io6-7 from a fermentation broth on a strong, cation-exchanging resin and eluting the resin adsorbate with an aqueous solution of ammonium hydroxide. The pu of the solution of the antibiotic from which the product is adsorbed can vary from about 5 to ig. In general, it is advantageous for the pH of the solution of the antibiotic to be 8 or above as there is some evidence that the antibiotic is fickle at lower pR values.
The recovery of the antibiotic io6-7 from the resin adsorbate can easily be accomplished by elution with a solution which contains a source of anions. Suitable eluents for this purpose are: acidic solutions, such as the aqueous solutions of organic and inorganic acids, e.g. B. acetic acid, propionic acid, carbonic acid, hydrochloric acid and sulfuric acid, basic solutions such as the aqueous solutions of strong organic bases, e.g. B. quaternary ammonium hydroxides, or inorganic bases, e.g. B. sodium hydroxide and potassium hydroxide, or aqueous solutions of salts such as sodium chloride, sodium sulfate, potassium chloride, calcium chloride, and the like. The weaker bases, such as aqueous solutions of ammonium hydroxide, can also be used, but are generally less advantageous than the solutions of the strong ones Bases. If the elution is carried out with acidic solutions, it has been found that the fractions of the eluate with a pH in the range from about .7 to 4.5 contain the active antibiotic. If basic eluents are used, the antibiotic 106-7 is found in the eluate fractions with a pI, from about 7 to 14-Particularly advantageous and in the context of the invention generally preferred is the use of a 20% acetic acid solution as the eluent.
The acidic and basic eluates obtained according to the invention contain the antibiotic io6-7 in an average strength of about 25o to 275 units / mg. A further purification to produce a practically pure antibiotic io6-7 can be carried out by various extraction and / or precipitation processes.
Adsorption and elution can be achieved by simply mixing the resin with the solution or preferably by pouring the solution through a with the resin-filled column take place. The latter method is generally preferred, because it is the easy determination of for complete adsorption of the antibiotic required contact time and the point at which the antibiotic is in the escaping liquid begins to flow through.
According to a further embodiment of the invention, the resin which contains the adsorbed antibiotic io6-7 can be washed or dried immediately, whereby a valuable, effective complex of antibiotic io6-7 and resin is obtained. It has been found that these resin complexes are very stable and can be valuable as feed additives for animals and therapeutic agents.
The effectiveness of the io6-7 antibiotic will be in the same way determined in which the analysis of streptomycin is carried out, namely after the Cylinder plate method using M. pyogenes var. Aureus and a streptomycin standard.
