Abstract
Description
The present invention relates to a reaction and solubilization medium for the synthesis of peptides. It also relates to a process for the synthesis of peptides in this medium.
Alle diese Methoden haben grundlegende Gemeinsamkeiten:There are a large number of methods for synthesizing peptides in the liquid phase. All of these methods have basic features in common:
- Gegebenenfalls Schutz der Seitenkettenfunktion einer Aminosäure durch eine Schutzgruppe, die am Ende der Peptidsynthese abgespalten werden kann;- If necessary, protection of the side chain function of an amino acid by a protecting group which can be cleaved at the end of peptide synthesis;
- Schutz der Aminfunktion (N α) der Aminosäure durch eine Schutzgruppe, die nach der Kondensation der Aminosäure abgespalten werden kann;- Protection of the amine function (N α) of the amino acid by a protecting group that can be cleaved after condensation of the amino acid;
- Aktivierung der Carbonsäurefunktion der geschützten Aminosäure;- Activation of the carboxylic acid function of the protected amino acid;
- dann die Kondensation der geschützten Aminosäure mit einer aminhaltigen Säure (Aminosäure) oder einem Peptid mit geschütztem endständigen C und freier Aminfunktion;- then the condensation of the protected amino acid with an amine-containing acid (amino acid) or a peptide with a protected terminal C and free amine function;
- Erhalten des Peptids durch Abspalten aller Schutzgruppen nach der Kondensation aller Aminosäuren.- Obtaining the peptide by removing all protecting groups after condensation of all amino acids.
The condensation reaction can be carried out in a homogeneous liquid phase as well as in a heterogeneous phase (e.g. Merrifield synthesis).
In general, the synthesis of peptides requires the protection of the carboxylic acid function of the C-terminated amino acid in the form of esters.
Im Fall von Peptid-Synthesen in homogener Phase ist der Ester ausgewählt aus den folgenden Estern:In the case of peptide synthesis in homogeneous phase, the ester is selected from the following esters:
- aus Methyl-, Benzyl- und tert.-Butylestern;- from methyl, benzyl and tert-butyl esters;
Ester von Polyethern wie bei M. MUTTER et al., Justus Liebigs Annalen der Chemie 1975, Seiten 901 ff., 915; oder auch- from esters of polymers that are soluble in organic solvents, e.g. esters of polyethers as in M. MUTTER et al., Justus Liebigs Annalen der Chemie 1975, pages 901 ff., 915; or also
In the case of peptide syntheses in heterogeneous phase, the ester can in particular be an ester of polymers that are not soluble in organic solvents.
In a non-exhaustive list, the following polymers can be mentioned:
* Styrol/Divinylbenzol-Copolymer, eingeführt von R.B. MERRIFIELD (J.A.C.S. 1963, 85, Seiten 2149-2154);* Styrene/divinylbenzene copolymer, introduced by R.B. MERRIFIELD (J.A.C.S. 1963, 85, pages 2149-2154);
* Polydimethylacrylamid-co-Boc-β-Ala-N'-acroylylhexamethylendiamin gemäß E. ATHERTON und R.C. SHEPPARD (J.A.C.S. 97, 1975, Seiten 6584-6585);* Polydimethylacrylamide-co-Boc-β-Ala-N'-acroylylhexamethylenediamine according to E. ATHERTON and R.C. SHEPPARD (J.A.C.S. 97, 1975, pages 6584-6585);
* auf Kel-F gepfropftes Polystyrol (TREGEAR at al., 1966, US-A-3 700 609 und Chem. Abot. 71 p. 508241 (1969));* polystyrene grafted onto Kel-F (TREGEAR et al., 1966, US-A-3 700 609 and Chem. Abot. 71 p. 508241 (1969));
* Cellulose.* Cellulose.
The peptide synthesis in heterogeneous phase can be carried out in a moving reactor, in a reaction column or according to any other technique (e.g. using a membrane).
The yield and purity of the final product depend considerably on the yield of each individual process step, on the one hand due to the geometric increase in losses and on the other hand due to the problems of separating the desired product from the by-products. This problem increases as the synthesis progresses and the number of amino acids increases.
Dieser Nachteil wird von zahlreichen Autoren bemängelt, so z.B. von:Furthermore, all these methods have the same disadvantage in the special case where the condensation is carried out in a homogeneous liquid medium: their volume productivity (productivity per volume) is very low due to the poor solubility of the protected amino acid or peptide intermediates. This disadvantage is criticized by numerous authors, for example:

(E. Wünsch, Methoden der Organischen Chemie, XV-2, Synthese von Peptiden, Georg Thieme Verlag, 108 (1974)).a) The productivity per unit volume is particularly low when using solvents that are immiscible with water, such as chloroform or ethyl acetate. For example, the 4-nitrobenzyl ester of N-(2-nitrophenylsulfenyl)-α-benzyl-phenylalanyl-N,ω-nitro-L-arginine is synthesized in amounts of 0.016 mol per liter in chloroform.
A. BODANSKY, The Practice of Peptide Synthesis, Springer-Verlag, 140, 1984).The methyl ester of benzyloxycarbonyl-L-prolyl-L-tyrosine is prepared at a concentration of 0.050 mol per liter in ethyl acetate (M. BODANSKY, A. BODANSKY, The Practice of Peptide Synthesis, Springer-Verlag, 140, 1984).
Water-miscible solvents have specific disadvantages and are hardly an advantage. Although they sometimes allow the synthesis of peptides to be carried out in a more concentrated medium, they make it impossible to purify these peptides directly by washing with water, which washing is necessary to remove the reagents introduced in excess and the products of the coupling reaction.
For this reason, the water-miscible solvents are generally distilled off and then replaced by water-immiscible liquid in order to carry out a washing operation. This brings us back to the problem mentioned above, since the cleaning step requires considerable volumes of solvents, while the treatment of the reaction mass is made more difficult by the use of additional distillation and redissolving steps.
The Practice of Peptide Synthesis, Springer Verlag, 129, 1984).Thus, the dipeptide Z-Cys(S-BZl)-TyrOEt, which was prepared in THF (2 mol/l), was transferred to ethyl acetate (0.1 mol/l) before washing with water (M. BODANSKY, The Practice of Peptide Synthesis, Springer Verlag, 129, 1984).
148, 1984).The protected dipeptide Z-Lys(Z)-Gly-OEt, synthesized in acetonitrile (0.13 mol/L), was purified after replacing the initial solvent with ethyl acetate (0.05 mol/L, ibid. 150, 1984), while Z-Ala-TyrOCH3 was prepared in dimethylformamide (0.3 mol/L) before being purified by washing with water in an ethyl acetate solution (0.1 mol/L) (ibid. 148, 1984).
The use of these water-miscible solvents, which are generally polar, leads to further disadvantages in terms of industrial hygiene (DMSO, HMPT) and in terms of chemical selectivity. In general, polar solvents favor the racemization of the N-protected activated amino acid, as pointed out by D.S. KEMP in Peptides, Analysis, Synthesis, Biology; 1, 354-355, 1979.
Thus, the preparation of concentrated solutions of peptide intermediates in solvents that are immiscible with water and compatible with current manufacturing standards (Good Manufacturing Practices) remains an unsolved problem.
For this reason, it is an object of the present invention to provide a reaction environment that increases the yield of peptide condensation products.
Another object of the present invention is to find a reaction medium which makes it possible to facilitate the synthesis and purification processes of peptides, in particular peptides containing 2 to 50 and especially 3 to 20 amine acids or amino acids.
Finally, another object is to provide a reaction medium that allows peptide synthesis in a homogeneous medium at high concentrations (e.g. at least equal to 0.1 M, generally 0.2 M).
These objects of the present invention and others which will be mentioned below are achieved with a reaction and solibilization medium for the synthesis and/or purification of peptides, which contains a diluent A selected from the group of water-immiscible solvents and a phenol B, the mass ratio between this phenol B and the diluent A being between 1/200 and 1, preferably between 1/20 and 1/2, and the medium containing at least one amino acid or at least one peptide in a concentration of at least equal to 10⁻² M.
Die Menge des Phenols B beträgt vorzugsweise mindestens 1/200 des Verdünnungsmittels A.The amount of phenol B is preferably at least 1/200 of the diluent A.
There is no fixed upper limit. Thus, the use of phenol B (as pure product or mixture) without diluent A is also part of the invention, to the extent that the following conditions are met:
- Der Schmelzpunkt des Phenols B beträgt höchstens 50 ºC (in der vorliegenden Beschreibung sind die Nullen keine signifikanten Stellen, außer wenn dies ausdrücklich erwähnt ist);- The melting point of phenol B is not more than 50 ºC (in this description, zeros are not significant figures unless expressly stated);
- das Phenol B kann von dem Reaktionsprodukt mittels Destillation und gegebenenfalls unter reduziertem Druck abgetrennt werden;- the phenol B can be separated from the reaction product by distillation and, if necessary, under reduced pressure;
- das Phenol B ist in jeder Menge mit Wasser nicht mischbar;- Phenol B is immiscible with water in any quantity;
- das Phenol B bildet keine stabile Emulsion, die die Entfernung von Nebenprodukten mittels Waschen mit Wasser beeinträchtigen kann.- Phenol B does not form a stable emulsion, which may affect the removal of by-products by washing with water.
For example, the halogenated phenols, preferably dichlorophenols and especially monochlorophenols, as well as lighter alkylphenols, meet these conditions.
When used alone, the phenols satisfying these conditions also consist of particularly interesting subclasses of phenols which can be used in mixtures with the diluents A.
Preferably, however, a diluent A and a phenol B are used. The mass ratio between the phenol B and the diluent A is advantageously between 1/200 and 1, preferably between 1/20 and 1/2. These values with regard to the mass ratio apply well both to the phenol itself and to those phenols whose molecular mass does not differ by an order of magnitude from the molecular mass of the phenol in the true sense (C6H5OH).
For phenols with higher molecular weights, the molar concentration is about 5 10⁻²M and is preferably between 0.1 and 2 M, in particular between 0.5 and 1.5 M.
If diluents A and phenols B are not miscible with each other in any quantity, the upper limit for the phenol(s) content is the lower of the two limits (i.e. the upper limit for the phenol content and the limit for solubility).
Unter dem Begriff "Phenol B" versteht man Phenole oder Mischungen von Phenolen.The term "Phenol B" refers to phenols or mixtures of phenols.
Diluents A are organic solvents which are sufficiently polar to dissolve at least 1% by mass, preferably at least 2% by mass, of the phenol (C₆H₅OH) and are sufficiently hydrophobic not to be miscible with water in any amount.
Preferably, the water can only be dissolved in amounts of at most 10% by mass of diluent A, preferably at most 1% by mass, and this only in the presence of phenol B as a third solvent.
The diluents A can be mixtures, including hydrocarbon fractions. Of course, under the process conditions, the diluents A must be inert with respect to the phenol and the reagents used for peptide synthesis.
The preferred solvents are selected from the group of aromatic derivatives, ethers, esters and halogenated solvents.
Examples of diluents A include: as halogenated aliphatic derivatives dichloromethane, 1,2-dichloroethane, 1,1,1-trichloroethane; as aromatic derivatives toluene; and as halogenated aromatic derivatives chlorobenzene; as esters ethyl acetate and isopropyl acetate; as ethers tertiobutyl and methyl oxide and anisole.
For economic reasons in industrial application, the diluent A can preferably be distilled under atmospheric pressure or under primary or secondary vacuum.
I) entsprechen:In general, phenol B is selected from the group of compounds corresponding to the following formula (I):
(R&sub1;)n-Ar-O-H (I)(R₁)n-Ar-O-H (I)
in der:in the:
- Ar einen monocyclischen, polycyclischen oder gegebenenfalls heterocyclischen, aromatischen Rest darstellt;- Ar represents a monocyclic, polycyclic or optionally heterocyclic, aromatic radical;

wobei R&sub2; ein Wasserstoffatom oder einen gegebenenfalls hydroxylierten oder mono- oder polyhalogenierten Aryl- oder Alkylrest mit höchstens 8 Kohlenstoffatomen darstellt;where R2 represents a hydrogen atom or an optionally hydroxylated or mono- or polyhalogenated aryl or alkyl radical having at most 8 carbon atoms;
- n die Zahl der Substituenten darstellt und gleich 0 ist oder eine ganze Zahl ist, die höchstens so groß ist wie die Zahl der substituierbaren Positionen an den aromatischen Kernen; und ihren Mischungen.- n represents the number of substituents and is equal to 0 or an integer that is at most as large as the number of substitutable positions on the aromatic nuclei; and their mixtures.
The alkyl groups (defined according to the Dictionnaire de la Chimie, Duval Presses Scientifiques Internationales, PARIS, 6th edition, 1959) can in particular be linear or branched aliphatic radicals containing a maximum of 6 carbon atoms or arylaliphatic radicals.
The number of substitutable positions is easily determined using rules known to the person skilled in the art.
Zum Beispiel gilt:For example:
Wenn Ar = Phenyl : n ≤ 5;If Ar = phenyl: n ≤5;
Ar = Pyridyl : n ≤ 4;Ar = pyridyl : n ≤ 4;
Ar = Naphthyl : n ≤ 7;Ar = naphthyl : n ≤ 7;
Ar = Chinolyl : n ≤ 6.Ar = quinolyl : n ≤ 6.
Vorzugsweise hat das Phenol B eine Zahl von Kohlenstoffatomen von höchstens 30, insbesondere von höchstens 20.Preferably, the phenol B has a number of carbon atoms of at most 30, in particular of at most 20.
It is desirable that the positions adjacent to the phenol function are not substituted or are only occupied by non-bulky groups.
Residues that are connected to the position adjacent to the phenol function via a tertiary or secondary carbon atom are considered bulky.
Monocyclische Verbindungen sind solche, die den besten Kompromiß zwischen Effizienz und Kosten liefern; man bevorzugt solche Verbindungen mit 6 Kettengliedern (Pyridinyl- oder Phenylkern).Monocyclic compounds are those that provide the best compromise between efficiency and cost; those with 6 chain members (pyridinyl or phenyl nucleus) are preferred.
Likewise, it is desirable that the group Z-R2 can represent no more than three, preferably no more than two, hydroxyl groups.
Vorzugsweise sind in der Formel (I) die Reste R&sub1; ausgewählt aus der Gruppe von:Preferably, in formula (I), the radicals R₁ are selected from the group of:

In order not to burden the phenol molecule too much, it is desirable that n in the formula (I) is at most 5, preferably at most 3.
Unter den Phenolen, die die besten Ergebnisse liefern, lassen sich nennen:Among the phenols that provide the best results are:
- gegebenenfalls monosubstituierte Hydroxypiridine;- optionally monosubstituted hydroxypyridines;

The reaction medium according to the invention makes it possible to dissolve an aminated acid (amino acid), which may optionally be protected, at a concentration of at least 0.05 M.
The amino acid can be C- or N-protected; in the latter case, the acid group on the acid function can be activated, e.g. optionally with one or more side-chain acid functions (e.g. diacids, in particular aspartic acid). The amino acids can be natural or synthetic amino acids.
The medium according to the invention also enables the dissolution of a protected peptide.
The medium according to the invention also makes it possible to dissolve unprotected peptides (HCl Pro-Gly-NH₂; HCl His-Trp; 2HCl Arg-Pro) and to carry out the synthesis.
Since the medium according to the invention simultaneously solubilizes amino acids, peptides and blocking or activating reagents, it makes it possible to carry out "in situ" blocking and even activation of the amino acids.
Thus, the medium according to the invention can play the role of a reagent if it contains, in addition to the phenol B and the diluent A, one or more peptides (including amino acids) and/or blocking reagents and/or activating reagents, in view of a gradual or simultaneous introduction (in general to be more practical). An aqueous phase immiscible with the reaction medium according to the invention can be an additional component of the reagent.
The peptide, including the elementary amino acid, may be C- or N-protected; in the latter case, the acid group may be activated via the acid function. Of course, other functions of the amino acids than the acid or amine function intended by the synthesis may be equally protected if required according to the methods conventional in the field.
The medium according to the invention allows a considerable increase in the solubility of the peptides in the organic phase.
It generally makes it possible to advantageously modify the magnitude of the solubility, taking as a basis the solubility of the peptide in the organic diluent A in the absence of the phenolic component.
The environment allows the various steps of peptide synthesis to be carried out under conditions that ensure a good volume yield. In particular, it increases the productivity of the condensation steps.
Among the numerous advantages of the medium according to the invention, it is particularly worth emphasizing the advantage that the condensations of an N-protected and C-activated amino acid or an N-protected and C-activated peptide with a non- C-protected amino acid or a non-C-protected peptide can be slightly facilitated and accelerated when the reaction takes place in the medium according to the invention. This represents a very significant advantage for the synthesis of oligopeptides (with up to about 20 or even 50 chain links). This is one of the reasons why the medium according to the invention must be particularly emphasized when it plays the role of reagent and contains as components for gradual or simultaneous addition:
a) ein Verdünnungsmittel A, das aus der Gruppe von mit Wasser nicht mischbaren Lösemitteln ausgewählt ist;(a) a diluent A selected from the group of water-immiscible solvents;
b) ein Phenol B;b) a phenol B;
c) ein Oligopeptid mit 1 bis 50, vorzugsweise mit 1 bis 20 Aminosäuren, von denen weder das terminale N noch das terminale C geschützt ist.c) an oligopeptide having 1 to 50, preferably 1 to 20 amino acids, of which neither the terminal N nor the terminal C is protected.
Die Menge an Phenol B beträgt vorzugsweise mindestens 1/200 des Verdünnungsmittels A.The amount of phenol B is preferably at least 1/200 of the diluent A.
There is no fixed upper limit for this. Preferably, the mass ratio between phenol B and diluent A is between 1/200 and 1, in particular between 1/20 and 1/2.
These values regarding the mass ratio apply both to the phenol itself and to those phenols whose molecular mass is not one order of magnitude different from the molecular mass of phenol in the narrower sense (C6H5OH).
For phenols with higher molecular weights, the molar concentration per litre is at least 5 10⊃min;²M, preferably between 0.1 and 2 M, in particular between 0.5 and 1.5 M.
If diluents A and phenols B are not miscible with each other in any quantity, the upper limit of the phenol(s) content is the lower of the two limits, namely the limit mentioned above and the limit for solubility.
The content of oligopeptide, which in the present sense also includes a non-condensed amino acid, is preferably at least 10⁻²M, in particular 0.1 M. The upper concentration corresponds to the solubility limit, although it is of course also possible to use a suspension of an oligopeptide if its solubility in the medium is considered insufficient by the person skilled in the art.
The present invention also relates to a process for the synthesis of optionally protected peptides in a liquid medium, starting from a peptide or an amino acid optionally protected at an acid function and adding an amino acid activated at its acid function and protected at its amine function, the reagents being dissolved in the medium according to the invention.
Likewise, the present invention relates to a process for the synthesis of optionally protected peptides in a liquid medium, which process starts from an amino acid or a peptide which is solubilized according to the invention and condenses another peptide whose C-terminal acid function is activated and whose N-terminal amine function is protected, the reagents being dissolved in the medium according to the invention.
For example, the amine function is protected by a carbamate group or by reaction with a β-dicarbonyl compound.
Preferably, the acid function of the N-protected reagents is activated by an organic or inorganic chloride, by an alkyl chloroformate, by a carbodiimide or by an activated ester or by their carbonyldiimidazole.
After condensation of an N-protected amino acid or peptide with the initial peptide or amino acid, the N-terminal amine function is released by any method, preferably by hydrogenolysis or acidic or basic solvolysis or by photolysis.
The initial amino acid or peptide protected at the terminal C (preferably by esterification or by amidation) is generally introduced into the reaction medium, then the amino acid or peptide activated at the acid function and protected at the amine function is added, the order of addition of the reagents being different; after the reaction, the by-products and the excess of reagents are eliminated from the organic phase by washing with an acidic, basic or neutral aqueous solution, then the amine function is released and finally a new amino acid or peptide is Peptide which is activated at the acid function or protected at the amine function.
Surprisingly, the environment potentiates the reactivity of the peptides with the amino acids and considerably improves the kinetics of condensation.
Hydroxybenzotriazol, Imidazol, N-Hydroxysuccinimid etc.A decisive advantage of the process according to the invention is that it allows the removal of excess reagents and condensation by-products from the organic phase by simple washing with water, which is facilitated by the anti-emulsion effect of the phenols, an effect that was established in the course of the experiments that led to the present invention. Excess reagents are, for example, activated and N-protected amino acids or peptides, C-protected amino acids or peptides, coupling-accelerating catalysts such as hydroxybenzotriazole, imidazole, N-hydroxysuccinimide, etc.
The medium according to the invention also makes it possible to increase the productivity of the steps in which the protective groups are cleaved. Furthermore, it also makes it possible to carry out the cleavage of acid-sensitive groups, e.g. tert-butyloxycarbonyl, using common reagents such as trifluoroacetic acid or hydrochloric acid, as well as the cleavage of hydrogenizable groups such as benzyloxycarbonyl, ethers and benzyl esters or nitro groups which protect the guanidine function of the arginine side chain.
The medium according to the invention enables the removal of base-sensitive groups, such as sulfofluorenylmethyloxycarbonyl, using common reagents such as diethylamine or piperidine. It also enables the removal of photolyzable protective groups. It also enables the removal of protective groups by electrochemical reduction.
An important advantage of the homogeneous phase process according to the invention is that it allows the removal of catalysts in the separation steps by simple process steps: the acidic or basic catalysts can be removed by aqueous washing, while the catalysts for hydrogenolysis, such as palladium absorbed on carbon, can be removed by filtration can be removed, leaving the peptide or peptic intermediate in the concentrated organic solution.
Another advantage of the solubilization process is that it is compatible with all the process steps required for the synthesis of peptides and it allows a direct chain between the different process steps (condensation, deprotection and purification by washing with water) without changing the solvent and, if necessary, without isolating the peptide.
It therefore enables the repeatable and especially iterative synthesis of peptides.
Due to the repeatability or reproducibility of the synthesis, the inventive method can preferably be carried out in an automated form.
The process is particularly interesting for the synthesis of peptides that are only slightly soluble in organic solvents, such as peptides that contain a glycyl, arginyl, glutaminyl, asparaginyl, serinyl, threonyl or prolinyl residue. As far as parameters not described in the description are concerned, the process according to the invention is carried out under various, but usual, conditions.
The peptides resulting from the synthesis can, if necessary, be used for the synthesis of drugs, vaccines, agricultural or plant protection products.
The present invention is illustrated by the following examples, which, however, do not limit it.
The solubilities of the peptide intermediate were determined as follows at a temperature of 25 ºC:
A precisely balanced amount of the peptide intermediate was placed in a Pyrex tube. The solvent or mixture of solvents for the experiment was then added using a precision pipette so that the mass ratio of peptide to solvent volume was 8 g/100 cm³.
The tube, closed with a screw cap, was then placed on a HEIDOLPH brand stirrer, model TOP-MIX, which stirred for 60 seconds at its maximum speed.
In case the solid is completely dissolved, the solubility is recorded as > 8 g/100 cm³, otherwise the volume of the solvent is doubled and the process is started again.
At the end of 4 consecutive dilutions without total dissolution, the procedure is repeated with a smaller amount of solid.
If the solubility in a given solvent is determined to be less than 0.05 g/100 cm3, this result is further verified by heating to 40 ºC and continuing to stir for 4 minutes without altering the result.
Remarks: The composition of the solvent and additive mixtures are expressed in weight.

For comparison purposes, an experiment is carried out according to Example 13, in which the addition of 4-tertiobutylphenol or the addition of 2-hydroxypyridine is omitted, which quickly leads to a reaction mixture that can no longer be stirred.
Benzylester des Glycyl-glycyl-L-phenylalanyl-L-leucins. (Gly-Gly-Phe-Leu-OBzl)Benzyl ester of glycyl-glycyl-L-phenylalanyl-L-leucine. (Gly-Gly-Phe-Leu-OBzl)
Given a purification of this tetrapeptide by washing with water at the end of the synthesis, the partition of this peptide between dichloromethane and water is studied as follows.
Ein Vergleichsversuch wurde in Gegenwart von Phenol durchgeführt:A solution of the hydrochloride of Gly-Gly-Phe-Leu-OBzl (0.9 mmol) in dichloromethane (12 cm³) is vigorously stirred in the presence of an aqueous KHCO₃ solution (2 N; 3 cm³). This mixing leads to an emulsion that remains stable for several days. A comparative experiment was carried out in the presence of phenol:
A partition test carried out according to Example 5 after adding 3.9 g of phenol to the organic solution yields, after identical stirring, two clear liquid phases whose decantation is completed in less than 5 minutes.
The HPLC analysis shows that the aqueous phase contains only 0.4% of the tetrapeptide used.
Benzylester des N-Tertiobutyloxycarbonyl-L-phenylalanyl-L-valins (Boc-Phe-Val-OBzl)Benzyl ester of N-tertiobutyloxycarbonyl-L-phenylalanyl-L-valine (Boc-Phe-Val-OBzl)
In a 50 cm³ Pyrex glass reactor, add one after the other at room temperature:
der 2-Hydroxypyridinester des N-tert.-Butyloxycarbonyl-L-phenylalanins (3,6 mmol; 1,23 g);the 2-hydroxypyridine ester of N-tert-butyloxycarbonyl-L-phenylalanine (3.6 mmol; 1.23 g);
das Chlorhydrat des Benzylesters von Valin (3 mmol; 0,73 g);the hydrochloride of the benzyl ester of valine (3 mmol; 0.73 g);
Dichlormethan (15 cm³); dann unter Rühren N-Methylmorpholin (3 mmol).Dichloromethane (15 cm³); then N-methylmorpholine (3 mmol) with stirring.
After 68 hours at room temperature, the reaction mixture is washed successively with 5 cm3 of dilute sulfuric acid (95%, pH 2), with 5 cm3 of water and then with 5 cm3 of a 2N aqueous KHCO3 solution in water.
Es bildet sich dann eine stabile Emulsion.A stable emulsion then forms.
In contrast, if a 9/1 mixture of dichloromethane/phenol is used as the reaction solvent, all washing operations proceed without difficulty and decantation is rapid.
Gegenstand der Untersuchung:Subject of the investigation:
Einfluß des Phenols im Verlauf der peptidischen KupplungInfluence of phenol in the course of peptide coupling
Boc-Phe-O-Py + HCl Val-OBzl + NMMBoc-Phe-O-Py + HCl Val-OBzl + NMM
CH&sub2;Cl&sub2; (mit oder ohne Phenol)CH₂Cl₂ (with or without phenol)
Boc-Phe-Val Obzl + HCl NMMBoc-Phe-Val Obzl + HCl NMM
The coupling is carried out by adding, in methylene chloride under reflux, on the one hand, 1.2 equivalents of the 2-hydroxypyridine ester of N-tert-butyloxy-L-phenylalanine (Boc-Phe-OPy) and, on the other hand, 1 equivalent of the hydrochloride of the benzyl ester of valine (VlOBzl) in the presence of N-methylmorpholine (NMM). The disappearance of ValOBzl and the formation of the peptide are measured. The correlation is perfect with regard to the measurement inaccuracies.
The results obtained are summarized in the following table, which shows the yield of peptide and, more importantly in terms of activation and kinetics problems, the amount of residual ValOBzl. Two different types of experiments are carried out in perfectly homogeneous medium, one with phenol (C6H5OH) in methylene chloride (10% by mass of phenol) and the other without.
The results show an extremely significant difference in kinetics, since 90% of the peptide is obtained in 2 hours in methylene chloride alone and in 20 minutes in the mixture of methylene chloride with 10% by mass of phenol.
The table shows the coupling yields and the percentage of ValOBzl over the course of the reaction time.
Influence of o-cresol on the kinetics of peptic coupling. Reaction Equival. Equiv.
The coupling is carried out by bringing into contact in methylene chloride at room temperature: 1.2 equivalents of the hydroxysuccinimide ester of N-tert-butyloxycarbonyl-L-phenylalanine (BocPheOSU) and 1 equivalent of free leucine (LeuOH) in the presence of 1 equivalent of diisopropylethylamine (DIPEA). Two types of experiments were carried out in heterogeneous medium (leucine is insoluble), on the one hand with 7.2 equivalents of o-cresol and on the other hand without o-cresol.
Man bestimmt die Bildung an Dipeptid BocPheLeuOH. The formation of dipeptide BocPheLeuOH is determined.
The results show a considerable influence of o-cresol on the coupling kinetics. In the presence of o-cresol, the formation of the peptide is multiplied by 6 times in 1 hour of reaction time.
