Verfahren zur Darstellung in der Estergruppe basisch substituierter
Carbaminsäure-bzw. Monoalkylcarbaminsäure- oder Allophansäureester Die Herstellung
eines basischen N-Dialkylcarbaminsäurederivates ist bekannt. Ebenso ist die Dar.tellun,-
von am Stickstoff arvlsubstituierten C arhaminsäureestern durch t ntsetzung von
Alkantinen mit Derivaten der Arylcarbatninsäure oder aliphatischen Aminen mit Halogenalkvlestern
der Arvlcarbaminsäure bekannt. Bei den vorliegenden Verfahren handelt es sich um
die Herstellung von in der Estergruppe basisch substituierten Cärbaminsäure- bzw.
\lonoalkvlcarbaminsäure- oder Allohhansäureestern -durch Umsetzung von nicht aromatisch
substituierten Carbaminsäurederivaten mit DialkvIaminoalkvlalkoholen. Solche Verbindungen
sind bisher noch nicht ltergestelit worden. 'Man glaubte offenbar. von vornherein
mit einem abweichenden Reaktionsverlauf rechnen. zu müssen. So war zu erwarten,
daß bei dem Versuch, vom Carbaminsäurechlorid aus Dialkvlaminoester aufzubauen,
der stark basische Charakter der Dialkvlaminoalkohole lediglich den bekannten Zerfall
des Carbaminsäurechlorids herbeiführen werde. Letzterer ist ja für den Fall der
Herstellung des eingangs erwähnten Dialky lcarbaminsäurederivates nicht zit befürchten,
da das hierfür zu verwendende Chlorid stabil ist, indem 'es keine 'Möglichkeit zu
einer intrantolekularen Chlorwasserstoffabspaltung bietet. Auch die Anwendung von
in Salzform vorliegenden Dialkvlaminoalkoholen ließ im Hinblick auf die Verwendungsmöglichkeit
eines Lösungsmittels erhebliche Schwierigkeiten voraussehen.Process for the preparation of basic substituted in the ester group

Bei Verwendung von z 1l01. Carbaminsäurechlorid auf i Mol. Dialkylaminoalkohol entstehen
die entsprechenden Allophansäureester. Die gemäß dem vorliegenden Verfahren hergestellten
Produkte sind deshalb wertvoll, weil aus ihnen wichtige pharmazeutische V erbindungen
hergestellt werden können. Beispiele 1. 11,7 g Diäthylaminoäthanol (Molekulargewicht
117) werden in 5o ccm Toluol gelöst und S g Harnstofchlorid, gelöst in io ccm Toluol,
unter Ausschluß von Feuchtigkeit und unter Rühren zugetropft. \ ach z4Stunden Stehen
wird das entstandeneSalz abgesaugt, in absolutem Alkohol aufgelöst und ein unlöslicherAnteil
abgetrennt. 'Nach Abdestillieren
des Alkohol; kristallisiert das
salzsaure Salz noch mit einigen Verunreinigungen. die man durch %Vaschen mit wenig
Aceton und dann wenig Chloroform herauslöst. Schließlich kristallisiert man aus

sich eine Ölschicht ab, die das salzsaure Salz des Carbaminsäureesters enthält.
Man trennt die Ölschicht im Scheidetrichter ab und verrührt sie mit Äther, wobei
sie alsbald kristallinisch erstarrt. Nach mehrmaligem L'mkristallisieren aus absolutem
Alkohol erhält man das Carbaminsäure-ß-diäthylaminoätllvlesterchlorhydrat vom Schmelzpunkt
15.4 bis I53° (s. Beispiel i). Die Ausbeute beträgt über 700,"o der Theorie. Der
freie Carbaminsäureß-diäthylaminoäthylester kristallisiert in derben, bei .42 bis

Reaktionsprodukt mit Äther verrührt, wobei eine kristallinische Ausscheidung eintritt.
Das Produkt wird in konzentrierter Pottaschelösung gelöst und die freie Base mit
Chloroform ausgeschüttelt, aus dem sie sich beim Abdestillieren abscheidet. Zur
Reinigung fällt man noch einmal aus Alkoholpetroläther um. Das Pseudo-i-phenyl-i-(aminoformyl-)oxy-2-dimethylaminopropan
schmilzt bei 13o bis 131'.
4. Zu einer Lösung von 1,;.49 salzsaurem ä-Diätilvlaminoäthanol @l.lo \fol.) in
j5 ccm Chloroform tropft man 16 g Harnstoffchlorid (= ,0 11o1.) in 5 o ccm Chloroform.
Das sich unter Erwärmen abscheidende Öl wird nach längerem Stehen mit Äther verrührt,
wobei es kristallisiert. Nach Zusatz von Carbonatlösung schüttelt man mit Chloroform
ab, der Rückstand daraus wird ausAceton umkristallisiert. 'Man erhält so den ß-Diäthylaminoäthylester
der Allophansäure in farblosen Kristallen vorn Schmelzpunkt 137 bis 138°.
