Verfahren zur Herstellung sekundärer aromatischer Aminoketone Durch
Patent 520 079 ist ein Verfahren geschützt, welches gestattet, am Stickstoff
alkylierte (oder arylierte) Oxyaminoacetophenone leicht und in befriedigender Ausbeute

Es wurde nun gefunden, daß die Anwendung des gleichen Verfahrens allgemein
sehr vorteilhaft zur Gewinnung von Oxyphenyla,lkylaminoalkylenketonen aus denentsprechenden
an der Hydroxylgruppe acylierten Haloggenverbindungen führt. Diesen kommt die allgemeine
Formel
zu, wo Ac eine Acylgruppe, R einen beliebigen zweiwertigen aliphatischen Kohlenwasserstoffrest
und Hal ein Halogen bezeichnet. Setzt man diese mit den Alkalisalzen von Al-1,vlamiden
um und verseift die entstandenen Verbindungen, so erhält man in guter Ausbeute Aminoketone
der Formel
wo R' einen einwertigen Kohlenwasserstoffrest bedeutet. Stellt R etwa die Gruppe
dar, so entsteht ein Aminoketon mit endständiger Aminogruppe, bedeutet R z. B. den
Rest = C H - C 113, so ist die Alkylaminogruppe des entstandenen Aminoketons

Beispiel i 6,7g, p-Ben7oyloxy-a-broi-npropioplienon der Formel
(F. 87', erhalten durch Bromieren der entsprechenden Benzovlverbindung vom
F. 117',
die man ihrerseits äurch Schütteln von p-Oxypropiophenon mit
Benzoylchlorid und --"Zatronlauge gewinnt, in Schwefelkohlenstofflösung) werden
in wenig Aceton gelöst und mit 4,5 g des Kaliumsalzes des p-Toluolsulfons.quremetliylamids
2 Stunden geschüttelt. Die entstandene kristallinische Masse wird abgesaugt, mit
Wasser und Alkohol gewaschen und der Rückstand mit 15 ccm etwa 5o'/,iger jodwasserstoffsäure
2o Minuten gekocht. Die Flüssigkeit wird nach dem Erkalten zur Entfernung von Benzoesäure,
Jod und Toluolsulfonsäurejodid mit Äther ausgeschüttelt und die wäßrige Lösung vorsichtig
zur
Trockne eingedampft. Der in derben Kristallen verbleibende Rückstand stellt das
Hydrojodid der Ketobase
dar. Man kann zu derselben Verbindung auch in folgender Weise gelangen: Beispiel
2 Die durch einstündiges Kochen von p-Oxypropiophenon mit Essigsätireanhydrid erhältliche
Acetylverbindung liefert mit i Mol Brom in Schwefelkohlenstofflösung ein Bromketon
der Formel
13,5 9 desselben werden mit 17,3 9 Benzoesäuremethylamidkalium
C,H,-CO-NK-CH,9 in Acetonlösung einige Stunden geschüttelt und vom ausgeschiedenen
Bromkalium abgesaugt. Die nach dem Eindunsten der Lösung auf ein kleines Volumen
erhaltene Masse wird nach dem Waschen mit Wasser und Alkohol, wie in Beispiel i
angegeben, behandelt, wobei die dort beschriebene Verbindung in fast quantitativer

Reaktionsprodukt wird abgetrennt und mit 75ccm 5o'/,iger Jodwasser-, stoffsäure
etwa 2-o bis 30 Minuten verkocht. Dann wird einmal mit Äther ausgeschüttelt,
die wäßrige Lösung abgetrennt und unter guter Kühlung ammoniakalisch gemacht. Das
ausfallende o-Oxyphenylmethylarnino-2-propanon-i wird mit alkoholischer Salzsäure
in sein gut kristallisierendes Hydrochlorid verwandelt. Ausbeute 5o bis 6o
