Description
It is known that the intensity of the fluorescent radiation with the concentration of the fluorophore increases in the irradiated volume. For media with unchangeable optical properties and refractive indices is determined by calibration or Modeling the concentration scaled to the intensity.
Regardless of whether a fiber or a flat detector for registration tion of fluorescence radiation, it was previously not possible to use simple methods (continuous radiation and detection in contrast to considerably more time-consuming method) information about the distance to obtain the fluorescence centers to the detector. Known arrangements are only suitable for the concentration of the fluorophore as an average over the to measure total excited volume. A spatial representation of the fluo Resence distribution in scattering media is not possible.
Furthermore, changing optical properties of the medium result Disturbance of the fluorescence signal.
From US 5,022,757 it is known the spatial distribution of fluorescent substances scanning by laser radiation. However, the scanning takes place here point by point in succession, so that obtaining an overall picture is time-consuming. In addition, two laser beams must be in each case for spatial scanning overlay a point in space.
Furthermore, DE 37 18 202 C1 describes the imaging of fluorescent substances with a Video camera known. However, the possibility of a spatial one is missing here Presentation.
The invention has for its object a device for determining spatial distribution of fluorescence centers in cloudy media in comparison to create little effort.
According to the invention, this object is achieved by means of the claims 1 and 7 specified characteristics. Advantageous further developments are in the respective Subclaims specified.
In the case of a similar basic solution according to the task, different devices are intended guides as modules the scaling of the fluorescence radiation by means of the Backscattering the excitation radiation and the tomographic reconstruction of Allow fluorescence images. Areas of application a. typically the laboratory as well as procedural analysis as well as medical Diagnostics using the body's own and enriched fluorescent dyes.
Surprisingly, it has been shown that when using detectors different cross sections and different numerical apertures the ge measured intensity of the fluorescent radiation in percent different Fluorescence centers of different distances are distributed.
Furthermore, there is a change in the reflection intensity of the excitation light Information about changed optical properties and optical coupling conditions included.
According to the invention, the cloudy medium to be examined, which is both solid, can be liquid as well as gaseous by a source of known geo stimulated with light of one or more wavelengths.
The excitation radiation as well as the fluorescence radiation are of one principle any number of detectors with basically any individual configuration (e.g. CCD cameras or individual fibers or fiber bundles) both in remission as well as, if possible, registered in transmission. The device allows the Repeat the procedure for each known positioning of the excitation source and the detectors, the detector arrangement not always rigid with the Excitation arrangement is connected. According to the invention, the fact is used that that both through the cross section of the detector and through its opening angle (numerical aperture) a selectivity in the spatial distance of the detected radiation is specifically achieved. The total intensity of fluorescence radiation is thus additively composed of parts from fluorescence at different distances.
A separation of these parts and thus a reconstruction of the spatial According to the invention, the concentration of the fluorophore is distributed in that by calibration on phantoms or by numerical simulations of the Relationship between concentration and fluorescence intensity is determined.
This is described mathematically as follows. Let I i be the total intensities of the fluorescence radiation, which are registered by the i-th detector with a certain opening cross-section and a certain aperture at a certain distance. These intensities are additively composed of parts that come from different spatial distances 1 , 2 , ..., n or can be assigned directly to different voxels in the fluorescent medium:
Ii = Ii,1 + Ii,2 + . . . + Ii,n I i = I i, 1 + I i, 2 +. . . + I i, n
These individual portions of the intensity are proportional to the concentration of the fluorophore over a wide range:
Ii,k = gi,k . ck.I i, k = g i, k . c k .
The weights g are dependent both on the geometry of the source and the detector and on the distance from the fluorescence center. They can be determined empirically by calibration on phantoms or by model calculations. Combining both equations creates a system of equations that links the intensities with the concentrations:
I = G × c I = G × c
Volumenelementen. I is the vector of the intensities from different detectors, G the known matrix of the weights and c the vector of the concentrations in different distances or volume elements.
According to the invention, the number of discrete spatial distances chosen should be that of the differently configured arrangements do not exceed, so that the linear system of equations is determined or over-determined. In the first case it is Solution clearly. Singularities are not relevant from a practical point of view, because in the interest of the stability of the solutions the spatial subdivision is always the same can be chosen that a neighborhood does not occur on such points.
If a lower resolution is chosen (dimension of vector c smaller than that of vector I ), the measurement values which are superfluous in the sense of the reconstruction algorithm can be used with the aid of statistical methods for noise suppression.
Outside the linear range, quadratic and higher-order dependencies can also be included in the reconstruction of the spatial fluorophore distribution:
Ii,k = gi,k . (ck) + g2 i,k . (ck)2 + ...I i, k = g i, k . (c k ) + g 2 i, k . (c k ) 2 + ...
The higher order weights g 2 etc. are to be determined empirically. The new system of equations thus created is also inverted using known methods of numerical mathematics.
Both technical realizations by individual Detectors physically and after registration with large apertures syn be generated theoretically. It does not matter whether there are several detectors positioned or aligned at the same time or a detector differently and used sequentially for measurement, as long as only the individual Positions correspond to those defined in the matrix G.
It is thus possible to quantitatively determine the concentration proportions from the intensities to reconstruct according to a chosen resolution. The maximum resolution is thereby by the number of detectors implemented in total (factually or synthetically) certainly.
The complex technique of separating runtime effects with the help of Correlation methods are thus replaced by a simple method.
Furthermore, the optical properties influence both the spread of the Excitation as well as fluorescence radiation. Again, through calibration the influence of spatial or temporal on phantoms or model calculations Fluctuations in these parameters to correct the scaling of the concentration of the Fluorophors can be determined via the fluorescence intensity.
In a preferred application example are therefore in a fiber bundle both the backscattered excitation radiation and the fluorescence radiation one or more excitation wavelengths with any number of optical fibers of known numerical aperture registered. These signals are read out individually and evaluated in a central processing unit.
In a further application example, are spatially free using a positionable source and one or more freely positionable cameras (fly ing optics) with variable opening cross-section (motor cover) in principle any number of configurations registers the different intensities. This Signals are read out individually and also in a central processing unit evaluated with regard to synthetically produced apertures.

A preferred application example is medical diagnostics using the embodiment from FIG . B. of indocyanine ( 32 a and b). This dye is excited at approx. 740 nm and the fluorescence radiation has a wavelength of approx. 785 nm. In this spectral range, biological tissue can be irradiated relatively deep (up to 10 mm). The detection takes place via a multifiber bundle 33 . In the first recording, for example, the inner ring of the fiber bundle 33 a is used for excitation and detection. As in Fig. 1a and b can be seen, it is possible to detect a marker 32 concentration fluorescence. The choice of the aperture and the distribution of excitation and fluorescent radiation is realized in the assembly 34 in a known manner. In a second step, the excitation and detection takes place via the aperture 33 b spanned by the outer fiber ring. As also the basic principle in Fig. 1a and b can be seen, while the fluorescent marker concentration 32 a and b detected. The evaluation unit 35 reconstructs the fluorescence image. The diagnostic procedure delivers results online and does not burden the patient with ionizing radiation. In this example, the method can also be used endoscopically in tumor diagnosis, in metabolism monitoring and in fluorescence angiography.
The methods of image synthesis associated with the process differ on the methods of density reconstruction within the field of view of a detector to be considered separately from this device. In particular, however, with With the help of all embodiments of the described device, sectional images of the investigating body with regard to the spatial distribution of fluorophores be won.
