Anti Cancer Drugs 3, 507 (1992)). Die bisher publizierten Methoden zur Erfassung von Mistellektinen in Mistelextrakten mit einem Enzym-Immuno-Assay (P. Ziska u. a. in: T.C. Bog-Hansen u. a. Lectins vol IV, 473 (1985)) oder einem Lektinbindungs-Enzym-Immuno-Assay (O. Vang u. a. in: T.C. Bog- Hansen u. a. (1989)) beschreiben den Einsatz von polyklonalen Mistellektin-spezifischen Antikörperpräparationen, deren begrenzte Sensitivität und Reproduzierbarkeit infolge des unterschiedlichen Antikörper-Repertoires zu einzelnen Antigendeterminanten von Mistellektin I-III keine differenzierte Analyse des Gehaltes an Mistellektin I, II und III ermöglichen. Die Standardisierung von therapeutisch angewendeten Mistelextrakten sowie deren arzneimittelrechtliche Kontrolle hinsichtlich der quantitativen Bewertung der Mistellektine I-III ist bisher nicht möglich (H. Das erfindungsgemäße Verfahren ist dadurch gekennzeichnet, daß als Fangantikörper für die Mistellektine I, II und III monoklonale Antikörper der Maus-Hybridomzellinien SIFIN Anti ML-A-5H8 (AC-1) bzw. Der immobilisierte monoklonale Antikörper wird mit einer Probe der Flüssigkeit in Kontakt gebracht, die Mistellektine neben anderen pflanzlichen Inhaltsstoffen, extrahiert aus Mistelpflanzenmaterial bzw. mit proteinchemischen Methoden gereinigtes Mistellektin I, II und III jeweils getrennt als Standardvergleichssubstanz enthält. Der sich ausbildende festphasengebundene Antikörper- Antigen-Komplex wird entweder mit dem enzymmarkierten monoklonalen Antikörper der Maushybridomzellinie SIFIN Anti ML-A-5F5 (AC-1) oder der Zellinie SIFIN Anti ML-A-5H8 (AC-2) umgesetzt. Anschließend wird nach Auswaschen der nicht an den festen Trägern gebundenen Substanzen in bekannter Weise die festphasengebundene Enzymaktivität ermittelt. Nach einer geschüttelten Inkubation (30 min/35°C) und anschließendem Waschen (5mal mit PBS-Tw) wurden in alle Vertiefungen der Platte 100 µl des in PBS-Tw (1 : 5000) verdünnten markierten (POD) monoklonalen Antikörpers SIFIN Anti ML-A-5F5 pipettiert. Der sich anschließende Inkubations- und Waschvorgang war identisch mit den vorangegan genen. Die Entwicklung und Bewertung der festphasengebundenen Enzymaktivität erfolgte analog zu Punkt 3 und ergab die in Abb. 2 dargestellte Kompetition der Bindung des Mistellektin-spezifischen mAk SIFIN Anti ML-A-5F5 an Mistellektin I durch Mistellektin spezifische Immunglobuline in Patienten-seren. Die Bewertung der unterschiedlichen Kompetition von SIFIN Anti ML-A-5F5 Epitopen auf den nativ über den festphasengebundenen mAk SIFIN Anti ML-A-5H8 präsentierten Mistellektin I bzw. III durch mistellektinspezifische Immunglobuline in 10 unterschiedlichen Patientenseren ist in Tab. 4 zusammengefaßt. - a) Umsetzung der Probe mit dem nachzuweisenden Mistellektin I und/oder Mistellektin II und/oder Mistellektin III sowie der gereinigten Lektinstandards (Mistellektin I, II und III) mit dem immobilisierten monoklonalen Antikörper SIFIN Anti ML-A-5H8;
- b) Entfernung nicht gebundener Bestandteile und Einwirkung des enzymmarkierten monoklonalen Nachweisantikörpers SIFIN Anti ML-A-5F5 oder seiner enzymmarkierten Antikörperfragmente;
- c) Entfernung nicht gebundener Bestandteile, Manifestation des gebundenen Enzymanteils und Berechnung der Konzentration im Vergleich zur Lektinstandardkonzentration (Mistellektin I) auf an sich bekannte Art und Weise.
