Abstract
Description
The present invention relates to an automatic chemical Analysis device and in particular an automatic chemical Analysis device, the microplates for analysis of samples, e.g. B. blood and liquids taken from patients, in which the analysis process is carried out precisely and efficiently can by adding appropriate microplates both before the reaction as well as after the reaction.
A chemical analysis procedure for performing immunolo analysis of blood samples using microplates, each of which has a number of reaction vessels which are formed therein in a matrix arrangement is known. An automatic chemical analyzer to run such a chemical analysis is also known. At this automatic chemical analyzer are featured given amounts of blood samples and reagents automatically into the Reaction tubes introduced, which are molded into the microplates are and particle patterns on the bottom walls of the reaction vessels due to immunological agglutination reactions occur are measured automatically. Then the respective Particle patterns based on the measured particles kelmuster judges whether she agglutinates or not agglutinates are kidneyed.
To assess the blood type or blood group and the Detection of different antigens and antibodies, e.g. B. HB antigen and syphilis antibody by using very small Performing amounts of blood samples has been a procedure for Detection of agglutination patterns suggested on the ground of reaction vessels through the immunological agglutinations reaction between antigens and antibodies. In Japanese Patent Application, Publication No. 61-59454 is a procedure for the automatic detection of agglutinated Particle pattern and non-agglutinated particle pattern described ben. In this known method are in a microplate a number of reaction vessels arranged in a matrix and at least part of the bottom wall of each reaction vessel is like this designed to have an inclined surface. Example the bottom of the reaction vessel is conical in shape shaped. Then a given amount of a blood cell a blood sample to determine its blood type or group and a predetermined amount of a standard antiserum reagent introduced into the reaction vessel to stop the reactions perform while the microplate is essentially kept at rest becomes. Then the agglutinated particle pattern or not agglutinated particle patterns on the sloping floor surface of the reaction vessel. This means that not agglutinated blood particles along the sloping floor Roll the surface of the reaction vessel down while agglutinating Blood particles hardly down along the inclined surface of the ground roll. Then the blood type of the blood sample is checked Capture the particle pattern assessed on the bottom wall of the reaction vessel is formed after the reaction.
Furthermore, Japanese Patent Publication No. 2-16875 (corresponds to US Patent No. 4,727,033) an automa Described chemical analysis facility in which the analysis method explained above carried out automatically becomes. In this known automatic chemical analysis Different types of analysis can be chosen be carried out by the same device. This means, that the analysis of types and types of blood particles, e.g. B. red blood cells, white blood cells, platelets and Lymphocytes; Components in sera, e.g. B. different types of antibodies, antigens, specific proteins and viruses; and foreign bodies through the use of blood cells, latex part Chen and carbon particles carried out in the analyzer will. This analyzer is very small and therefore requires little space for its installation.
Fig. 1 shows a schematic arrangement of the known automa tical chemical analysis device, as described in the aforementioned Japanese Patent Publication No. 2-16875 be.
A blood sample taken from a patient is contained in a test tube 46 , on which a sample identification sticker 47 is attached, on which sample identification data of the respective patient are recorded. The sample identification data is recorded in the form of a bar code. The test tubes 46 , which contain the blood samples from patients, are placed in a rack 48 and the rack is moved through a rack transport device 49 , e.g. B. a conveyor belt, conveyed into a diluent supply device 52 . In addition to the conveyor path of the frame 48 is a sample identification data reading device 50 , for. B. a bar code reader, arranged to read the identification data located on the sticker 47 , while the frame 48 migrates through the identification data reading device. The sample identification data 51 , which are read by the reading device 50 , are fed to a central processing unit (CPU) 73 for controlling the operation of all components of the analysis device and for evaluating the analysis results.
The blood sample contained in the test tube 46 is diluted by the diluent supply device 52, and then a diluted blood sample is introduced into a sample vessel 53 . The sample vessel 53 is then brought into a sample supply position 54 . At the same time, a microplate 56 , which is arranged in a microplate stack section 57 , is brought into the sample supply position 54 by a microplate transport device 58 . In each microplate 56 , a number of reaction vessels 55 are arranged in a matrix, and at least part of a bottom wall of each reaction vessel is inclined. In the present embodiment, the bottom of the reaction vessel 55 is shaped in a conical shape.
The sample vessels 55 in the microplate 56 are successively brought into the sample supply position 54 and a predetermined amount of a diluted sample is filled into a reaction vessel 55 , which is formed in the microplate 56 , by means of a sample supply nozzle 59 . Furthermore, one of the reagents is filled into the reaction vessel 55 through a reagent supply nozzle 60 . At the sample supply point 54 , a sensor 61 is arranged in order to detect the presence of the microplate 56 . A microplate detection signal 62 generated by the sensor 61 is supplied to the CPU 73 and in number of the micro plates 56, one after the other in the position Probenzuführ be transported 54 is counted on the basis of this signal 62nd
The microplate 56 , in which samples and reagents are filled, is then brought into a reaction belt 63 by means of the transport device 58 through an inlet opening 61-1 of the reaction belt 63 .
The reaction belt 63 includes a lifting mechanism 63-2 to move the microplates 56 down in the reaction belt 63 . In this way, the microplate 56 accommodated in the reaction belt 63 can be held for a predetermined reaction time. It should be noted that the different analyzes are carried out during the same reaction time. During the reaction time, the particles are agglutinated or non-agglutinated to form the agglutinated particle pattern or the non-agglutinated particle pattern. After the agglutination reaction, the microplate 56 is conveyed into a photometric position 65 by means of a transport mechanism 64 through an outlet opening 63-3 of the reaction belt 63 .
Next, the microplate 56 is brought into a photoelectric measuring position 65 and the particle patterns formed on the inclined surfaces of the reaction vessels 55 in the microplate 56 are measured with the aid of a photodetector 66 , e.g. B. CCD camera, photoelectrically detected. A particle pattern detection signal 67 generated by the photodetector 66 is supplied to the CPU 73 . Furthermore, a sensor 68 for detecting the microplate 56 is arranged at the measuring point 65 and a microplate detection signal 67 is fed to the CPU 73 . In the CPU 73 , the microplate detection signals are counted to identify integer numbers of successive microplates that have been brought into the measuring point 65 . After the photometric measurement, the microplate 56 is conveyed out of the measuring point 65 by the transport mechanism 64 .
Fig. 2 is a schematic view which explains the flow of data in the known analysis device described above. In order to correctly process the measured analysis data 67 by the photo detector 66 for a blood sample 71 in order to obtain an output 72 with the aid of an output device 72 , it is necessary to analyze the photoelectrically recorded analytical data 67 on the inclined bottom wall of the Reaction vessel 55 particle pattern formed in the microplate 56 to the sample identification data 51 in a correct manner.
In the known analyzer, as shown in Figs. 1 and 2, in order to correctly assign the measured analytical data 67 to the sample identification data 51 , it is absolutely necessary to confirm that the microplate 56 before the reaction on the reaction belt 63 is identical to the microplate 56 after the reaction. This means that the microplate 56 in which the blood samples and reagents have been brought together must be confirmed to be identical to the microplate which is in the photometric position 65 after the reaction.
However, in the known analyzer, the sensors 61 and 68 are provided at the sample supply point 54 and the measuring point 65 , respectively, to detect the presence of the microplates to generate the microplate detection signals 62 and 69, respectively, so that it is impossible to th the respective microplates Identify 56 . In the known analysis device, the identification of the microplates is carried out by counting the microplate detection signals in order to obtain an integer number of the successive microplates. If the analysis is interrupted due to mechanical or electrical disturbances so that correct counting cannot be carried out, it is therefore no longer possible to carry out the identification of the microplates 56 since the sensors 61 and 68 cannot detect the microplates in question.
It should be noted that a plurality of microplates 56 are used simultaneously in the analyzer and there are no means to identify the respective microplates, so that it is impossible to distinguish these microplates from one another by interrupting the analyzer during the interruption she is watching. Therefore, the reliability of the analysis data can no longer be guaranteed after the resumption of operation.
The present invention is therefore based on the object a new and advantageous automatic chemical analysis create direction in which the respective microplates iden tified and thus the reliability of the analysis results can be improved.
Another object of the present invention is to provide an automatic chemical analysis device, the microplates used in the respective microplates can be identified by inspecting the microplates can.

Fig. 3 is a schematic diagram illustrating the basic structure of an automatic chemical analysis device according to the invention. A blood sample 2-1 is contained in a test tube 2-3 and a sample identification code sticker 2-2 is attached to the test tube. The sample identification code data are read from the sample identification code sticker 2-2 with the aid of a first bar code reader 17 and the sample identification code data 13 thus detected are fed to a CPU 16 .
At a delivery point 7 , a dilution device 3 for diluting a blood sample 2-1 , a sample feed device 5 for feeding predetermined amounts of the respectively diluted samples into reaction vessels 6-1 , which are incorporated in a microplate 6 , are provided. Furthermore, a reagent supply device 4 is seen at the supply point 7 for supplying predetermined amounts of desired reagents into the reaction vessels 6-1 . According to the present invention, 6 microplate identification members 1 for identifying the respective microplates are arranged on each microplate. The Mikroplat ten-identification member 1 carries a first identification code 1-1 , which can be detected by photoelectric means and a second identification code 1-2 which can be recognized by an operator with the eye. A first microplate identification code reading device 11-1 for deriving first microplate identification code data 12-1 is provided at the feed point 7 , at which samples and reagents are introduced into the reaction vessels 6-1 in the microplates 6 . This first microplate identification code data 12-1 is supplied to the CPU 16 . Then, the CPU 16 processes the sample identification code data 13 and first microplate identification code data 12-1 to form an identification table that establishes the mutual relationship between the samples 2-1 and the microplate. A method for forming the identification table is known in the prior art, so that it is not further explained in this description.
After the blood samples and reagents have been supplied, the microplate 6 is transported on a reaction belt 8 . After the reaction, the microplate 6 is brought into a measuring position 10 by the reaction belt 8 . At the measuring point 10 , a photoelectric sensor 9 for detecting particle patterns is seen, which are formed on the bottoms of the reaction vessels 6-1 , and the photoelectrically measured data 14 are supplied to the CPU 16 . At the measuring point 10 , a second microplate identification code reading device 12-2 is provided in order to detect second microplate identification code data 12-2 . This second microplate identification code data 12-2 is also supplied to the CPU 16 . In the CPU 16 , the above-mentioned identification table is searched out in accordance with the second microplate identification code data 12-2 to find the mutual relationship between the respective microplate and the samples. This means that by searching the identification table based on the second microplate identification code data 12-2, it is possible to relate the measured data 14 to the samples that have been filled in the reaction vessels 6-1 of the respective microplate 6 . In this way, it is possible according to the invention at any time to confirm that the microplate 6 into which the samples and reagents have been filled at the filling point is the same as the microplate whose measured data have just been acquired, so that the reliability of the analysis results can be improved.
Fig. 4 is a schematic diagram illustrating an embodiment of an automatic chemical analysis device according to the invention. In the present embodiment, the device is constructed so that the analysis of antigen-antibody reactions can be carried out by using the immunological agglutination reaction. Since the sequential analysis steps are the same as those in the known analyzer, as illustrated in FIGS . 1 and 2, their explanation is omitted.
As explained above, the identification bar code sticker 1 according to the invention is attached to the microplate 6 , which has a number of reaction vessels 6-1 in a matrix arrangement. The reaction vessels 6-1 have a conical bottom. The microplate identification bar code sticker 1 is applied to the microplate 6 when the microplate is inserted in a microplate stack section 22 .
At the feed point 7 , blood samples 2-1 , which are contained in the test tubes 2-4 , are diluted, and predetermined amounts of the diluted blood samples and reagents are introduced into reaction vessels 6-1 in a microplate 6 . Then the micro plate 6 is transported to a reaction line 38 . A first microplate bar code reader 11-1 is provided at the feed point 7 for reading out first microplate identification code data 12-1 . After a predetermined reaction time, the microplate 6 is brought into a measuring point 10 in order to detect photoelectrical particle patterns which are formed on inclined bottom surfaces of reaction vessels 6-1 . At the measuring point 10 , a second microplate bar code reader 12-2 is provided for reading out second microplate identification code data 12-2 .
Sample bar code stickers 2-2 , which are attached to the test tubes 2-4 , which contain blood samples 2-1 , are read by a sample bar code reader 17 and the sample identification data 13 , which are read by the bar code reader 17 , become a CPU 16 Processing of analysis results fed. At the measuring point 10 , the particle patterns, which have formed on the bottoms of the reaction vessels 16-1 , are photoelectrically recorded by a camera 31 and the particle pattern data 14 thus detected are supplied to the CPU 16 .
In the CPU 16 , the first microplate identification code data 12-1 , which were detected by reading the identification bar code label 1 on the microplate 6 with the help of the first barcode reader 11-1 , and the sample identification data 13 are processed to form an identification table showing the mutual relationship between the samples and the microplate. When the particle pattern is photoelectrically detected by the camera 31 , the identification table according to the second microplate identification code data 12-2 , which is read by reading the identification bar code sticker 1 with the help of the second bar code reader 11-2 , is searched out to find samples , to which the respective photoelectrically detected particle patterns belong. Measured particle patterns are analyzed in the CPU 16 in order to obtain analysis results which reflect the agglutination or non-agglutination. In this embodiment, a reaction start time at which a sample and a reagent are introduced into a reaction vessel is recorded in the identification table, and a reaction end time at which a particle is measured photoelectrically is also included therein. Then, a response time is calculated as the difference between the response start time and the response end time, and the response time thus calculated is compared with a predetermined standard response time 16 in the CPU 16 to obtain a judgment result of the response time judgment. Then the analysis results and the evaluation results of the response time are output.
Fig. 5 is a perspective view showing an embodiment of the microplate identification code label 1 attached to a microplate 6 . A number of reaction vessels 6-1 are formed in the microplate 6 and are arranged in a matrix arrangement, and each reaction vessel has a conically shaped bottom. The microplate identifi cation code sticker 1 is placed on the upper surface of the microplate at a position where no reaction vessel is formed. The microplate identification code sticker 1 is made of synthetic resin or paper with an adhesive substance on a lower surface, so that the sticker can be easily peeled off the microplate. On the front surface of the microplate identification code sticker 1 , the bar code 1-1 is recorded, which can be read by the bar code reading devices 11-1 and 11-2 and carries the numerical or alphanumeric data 1-2 directly by an operator is readable with the eyes.
The present invention is not limited to the above written embodiment limited, rather many are changes possible that can be achieved by experts who in the Range of invention. For example, the microplate ten identification code parts made of materials, e.g. B. ceramics or Alloys are made that are hardly affected by reagents can be gripped and attached to the microplate with the help of Adhesives are glued on, which is also hardly caused by reagents tien be attacked. In the case of using a derar It is no longer necessary to use the identification code sticker Lich the sticker from the microplate after each measurement remove. Furthermore, identification code data can be ge forms that they are detected magnetically or electromagnetically can be.
According to the first and second devices for Detect the microplate identification code on the micro plate provided in the feed position and the measuring position, so that the measured analysis data is always correctly related to the samples can be put and the reliability of the Analysis can be improved.
The respective microplates can also be identified and it is therefore possible to measure the measured analysis data to relate the samples, even if the operation of the Analyzer was interrupted.
In addition, even if the microplate Identification code data due to some circumstance not the microplates have been properly grasped by viewing the microplate identification data on the identification identifying stickers, the measured data can Samples can be assigned correctly.
It is therefore no longer necessary to measure the data discard or rerun. It is very advantageous for analysis in a large laboratory in one large number of samples is processed every day.
