Abstract
Description
The invention relates to new leukotriene B₄ derivatives, processes for their preparation as well as their use as medicines. The new connections are optically active Structural analogues of previously known leukotriene B₄ antagonists using a six-membered ring included as a basic structural element (DE-A 39 17 597, DE-A 42 27 790.6). Leukotriene B₄ (LTB₄) was developed by B. Samuelsson and co-workers as a metabolite of arachidonic acid discovered. In biosynthesis, the enzyme 5-lipoxygenase is initially used as central intermediate, the leukotriene A₄ formed, which then by a specific Hydrolase is converted into the LTB₄.

Out of this results that LTB₄ is an important inflammation mediator for inflammatory diseases is where leukocytes migrate into the diseased tissue.
The effects of LTB₄ are at the cellular level through the binding of LTB₄ triggered a specific receptor.
LTB₄ spielt offensichtlich bei entzündlichen, allergischen und immunologischen Prozessen eine entscheidende Rolle.LTB₄ is known to cause leukocyte adhesion to the blood vessel wall. LTB₄ is chemotactically active, i. H. it triggers a directional hike from Leukocytes towards a gradient of increasing concentration. Furthermore due to its chemotactic activity, it indirectly changes vascular permeability, a synergism with prostaglandin E₂ being observed. LTB₄ plays obviously one with inflammatory, allergic and immunological processes decisive role.
Leukotrienes and especially LTB₄ are involved in skin diseases associated with inflammatory processes (increased vascular permeability and edema, cell infiltration), increased skin cell proliferation and itching, such as for eczema, erythema, psoriasis, pruritus and acne. Pathologically increased Leukotriene concentrations are either the cause of the formation of many dermatids involved, or there is a relationship between the persistence of the Dermatids and the leukotrienes. Significantly increased leukotriene concentrations have been found, for example, in the skin of patients with psoriasis or atopic dermatitis measured.
Leukotrienes and especially LTB₄ are also involved in diseases of internal organs, for which an acute or chronic inflammatory component has been described, e.g. B .: joint diseases (athritis); Respiratory disorders (asthma, Rhinitis and allergies); inflammatory bowel diseases (colitis); as well as reperfusion damage (on cardiac, intestinal or kidney tissue), which can be caused by Closure of blood vessels arise.
Leukotrienes and especially LTB₄ continue to have an impact on education from white blood cells in the bone marrow, to the growth of smooth muscle cells, from keratinocytes and B lymphocytes. LTB₄ is therefore for diseases with inflammatory processes and with diseases with pathological increased cell formation and growth involved.
Erkrankungen mit diesem Erscheinungsbild stellen z. B. Leukämie oder Artherosklerose dar.Diseases with this appearance represent z. B. leukemia or atherosclerosis.
By antagonizing the effects of LTB insbesondere in particular, the active substances and forms of administration of this invention specific cures for the disease of humans and animals, in which leukotrienes in particular are pathological Role-play.
In addition to therapeutic options that result from antagonizing the LTB₄ Effect with LTB₄ analogues could also derive the usefulness and potential Use of leukotriene B₄ agonists for the treatment of fungal diseases the skin (H. Katayama, Prostaglandins 34, 797 (1988)).
