Bekannt ist, daß nach UV-Bestrahlungen erhöhte PGD2- und PGE2-Werte gefunden werden. Anschließend erfolgt eine Hyperpigmentierung. Nordlund (J. Invest Dermatol 86,4 433-437,1986) hat gezeigt, daß epidermale Zytokine wie TNFα, IL-1 und IL-6 ein negatives feed-back auf die Proliferation und Melanogenese der normalen humanen Melanozyten ausüben. Diese Autoren haben diese Zytokine als Inhibitoren der normalen humanen Melanozyten charakterisiert, d. h. ein erhöhter TNF- Spiegel in diesem System führt zur erhöhten Inhibition bzw. zur Depigmentierung. Durch die Hemmung des Inhibitors kann man eine Erhöhung der Melanogenese erzielen. Es wurde nun überraschenderweise gefunden, daß die genannten Carbacyclinderivate einerseits topisch appliziert eine langanhaltende Hyperpigmentierung hervorrufen und andererseits, daß sie die TNF-Synthese auf mRNA-Ebene hemmen. Carbacyclinderivate sind monoklonalen Antikörpern, die gegen TNF gerichtet sind, vorzu ziehen, da sie nicht nur auf bereits sezernierten TNF wirken, sondern sie können auch prophylaktisch eingesetzt werden und sind im Gegensatz zu den monoklonalen Antikörpern auch nach topischer Applikation wirksam. Herzerkrankungen, koronarer Thrombose, des Herzinfarkts, peripherer Arterienerkrankungen,
Arteriosklerose und Thrombose, Therapie des Schocks, Inhibierung der Bronchokonstriktion,
Inhibierung der Magensäuresekretion und Zytoprotektion der Magen- und Darmschleimhaut;
antiallergische Eigenschaften, Senkung des pulmonalen vaskulären Widerstandes und des
pulmonalen Blutdruckes, Förderung der Nierendurchblutung, Anwendung an Stelle von
Heparin oder als Adjuvans bei der Dialyse oder Hämofiltration, Konservierung von Blut
plasmakonserven, besonders von Blutplättchenkonserven, Inhibierung von Geburtswehen,
Behandlung von Schwangerschaftstoxikose, Erhöhung der zerebralen Durchblutung und
Antiproliferation.

HmB45 ist ein Marker der zur Charakterisierung der Reifestadien der Melanozyten entwickelt wurde. Exprimiert wird er z. B. als Reaktion auf einen aus Endothelzellen stammenden Faktor, bei erhöhter Vaskularisation sowie bei Melanozytenstimulation in der fetalen Haut. Elektronenmikroskopisch läßt sich zeigen, daß die Anzahl der Melanosomen in den Zellen erhöht ist. Eine gesteigerte Fortsatzbildung mit einer größeren Verteilung der Melanosomen wird ebenfalls gefunden.
