Abstract

Recovery or pure recombinant M-13-ss DNA (I) comprises (1) pptn. of recombinant M13 phages, excreted into the medium from an E. coli strain, by treatment with acetic acid; (2) filtering the resulting suspension through a glass fibre filter; then (3) treating the filter with a soln. contg. chastropic ions to extract the envelope proteins. Also new is an appts. for this process. Specifically, for pptn., acetic acid is added to a final concn. of 1-10% and the extract soln. is 4-8m perchlorate of pH 6.5-8.5. After extraction, the filter is washed with aq. EtOH and the purified DNA eluted with an aq. soln. of 0.2-2mM Tris-HCl (H 7-8) contg. 0.05-0.2mM EDTA.
