Abstract
Description
The present invention relates to a self-adhesive Adhesive plasters for delivering drugs to the skin.
The cutane administration of drugs by means of a plaster ger products is known. Dosage forms of this type offer the Possibility to equal the drug over a longer period of time dispense moderately and therefore often as the restrict annoying oral medication. In addition, the transdermal application of drugs offers in many cases the possibility of supplying the organism To reduce the amount of drug. This is especially true for such drugs that after oral administration during the first liver passage in pharmacologically inactive metabolites being transformed. It is known that this also as "first pass metabolism "referred to effect by a transdermal Administration of the drugs can be avoided.
A number of embodiments are described in the literature ben, with the help of the transdermal application of drug fen can be accomplished. For example, in US 15 77 259 describes a plaster-like preparation consisting of one Top layer, a drug-containing reservoir and one microporous membrane used to control drug delivery serves. The role of the membrane that controls drug delivery can also be applied through a layer of pressure sensitive adhesive such as the one below is described in EP-A1-00 33 615. In addition to these in one A number of other patent applications described so-called membrane controlled systems have the so-called matrix systems in which the drug is incorporated into a self-adhesive matrix, acquired a special meaning. The matrix takes over at the same time the function of a drug reservoir, one that Release speed control element and one PSA layer. Such a system can be a multilayer Have matrix, as described in DE-OS 33 15 245. In a special embodiment, it should be graded by a Drug concentration in the individual layers one more or less uniform release of the drug can be achieved. Furthermore, a number of monolayers are used in the literature Matrix systems of different compositions are described.
The used to make such self-adhesive matrices Pressure sensitive adhesives are characterized in that they are in water are practically not swellable. The contact often leads to Water, e.g. Sweaty skin, even causing adhesion to the Skin is significantly reduced, or that the plaster is not more sticks. At the same time, the selection of drugs that can be administered with the aid of such systems. As in B. C. Lippold: Biopharmacy, 2nd ed., Wiss. Publishing company Stuttgart, 1984, page 116, is carried out according to the Higuchi equation is the released amount of a drug a topical preparation proportional to its satiety in this system. Because of the more or less pronounced Lipophilicity of the in conventional medicated plasters The pressure sensitive adhesive used is the selection of drugs that can be used suitable for administration by means of such systems, on non-polar Substances limited, since only these are sufficiently soluble in lipophilic systems.
Another significant disadvantage of conventional single-layer self-adhesive plaster is that it is occlusive Act. As a result, the sweat on the skin Application site is secreted, not unhindered to the Environment can be delivered. This causes this to swell Skin areas that cause a change in skin permeability and can lead to skin irritation.
The invention has for its object a self-adhesive Adhesive plasters for delivering drugs to the skin to provide, which avoids occlusive conditions, a sufficient release rate especially for ensures polar drugs and their adhesion to the skin is also preserved under the influence of moisture.
This task is solved by a self-adhesive adhesive plaster, which is characterized by the following features:
- a) eine Deckschicht (1) mita) with a top layer ( 1 )
-
- b₁) at least one water-swellable polymeric matrix former,
- b₂) at least one water-insoluble, tackifying substance in a weight ratio of b₁) to b₂) of 1:10 to 10: 1 and
- b₃) comprises one or more absorption mediators.
The polymer matrix ( 2 ) contains polymeric substances that swell in water, which absorb the sweat that forms on the skin and thereby prevent or severely restrict occlusive conditions.
The use of such water-swellable polymers Substances in pharmaceutical plasters are known. So z. B. in the DE-OS 29 20 500 a pharmaceutical preparation in the form of a Described films that are suitable for transdermal use is. The film consists of a water-swellable copolymer sat from alkyl esters of acrylic and methacrylic acid. The drug Amorphous material is embedded in the film. The manufacturer lung is carried out in such a way that in an aqueous polyacrylic Latdispersion first, if necessary, the auxiliaries and - if it is are very well water-soluble drugs - these in the desired amount dissolves. The dispersion is then on a poured out flat, limited area and possibly in the case of increased Let the temperature dry. After pouring and drying becomes a solution of the drug in an organic solution medium applied to the film and then evaporated, whereby the drug should diffuse into the film.
Die EP-A2-0 86 997 beschreibt ein von der Zusammensetzung her ähnliches, bezüglich des Herstellungsverfahrens modifiziertes System, bei dem zunächst die wäßrige Polyacrylatdispersion gefriergetrocknet, anschließend mit dem Arzneistoff in einem organischen Lösungsmittel gelöst und schließlich zu einem Film ausgegossen und getrocknet wird.EP-A2-0 86 997 describes a composition Similar, modified with regard to the manufacturing process System in which initially the aqueous polyacrylate dispersion freeze-dried, then with the drug in one dissolved organic solvent and finally to a film is poured out and dried.
Both systems are very complex to manufacture. They have but above all the disadvantage that the drug-containing film does not have sufficient adhesion to the skin. For this Basically, according to EP-A2-00 86 997, there should be an additional one on the film Adhesive layer can be applied.
The self-adhesive adhesive plaster according to the invention consists in essentially consisting of a top layer and a one-sided layer applied self-adhesive layer of a polymer matrix. The Polymer matrix serves both as a carrier for the dissolved in it Drug as well as an adhesive layer necessary for an adequate Adheres to the skin. The layer of the polymer matrix can be provided with a removable protective layer, the front the application of the adhesive plaster is removed. The polymer matrix comprises at least one water-swellable polymeric matrix image ner and at least one water-soluble tackifying substance. The weight ratio of matrix former to tackifying substance is 1:10 to 10: 1, preferably 1: 1. Furthermore, the Polymer matrix one or more absorption mediators. The Drug is evenly distributed in the polymer matrix layer.
