Abstract

The gene for resistance to phosphinothricin (PTC; 2-amino-4-methylphosphino-butyric acid) is new. It is derived from the total DNA of Streptomyces viridochromogenes DSM 4112 (selected for resistance to phosphothricylalanyl-alamino, PTT) by cutting with BamHI, cloning the resulting Ak6 fragment and selection for PTT resistance. DNA was isolated from colonies of DSM 4112 resistant to 100 microg/ml PTT, cut with BamHI and fragments from pSVH1 by ligation with a thiostreptone-resistance fragment described in EP 158201). Recombinants were used to transform S.lividans TK23 and PTT-resistant colonies isolated. These colonies contained the plasmid pPRI. The 4kb fragment in pRI was cut first with SstII-SstI and then with kgl II to isolated a 0.8 kb fragment able to impart resistance to S.lividans. The DNA sequence of this 0.8 kb fragment, and of possible amino acid sequences derived from it, and reproduced in the specification. The gene codes for a PTC-octeyltransferase which selectively acylates the L-form of PTC in a racemate. This enzyme can be isolated from cultures but more conviently is used in the form of cells.
