Abstract
Description
The invention relates to a method for producing cellulose fibers for the removal of leukocytes from those containing leukocytes Blood fluids, especially platelet preparations, through Filter and a filter, which such a cell in a filter container contains loose fibers.
Die Filtrierung von leukozytenhaltigen Blutflüssigkeiten, z. B. mensch lichen Blutplättchenkonzentraten, über Zellulosefilter zwecks Entfer nung vorhandener Leukozyten führt man durch, um bei wiederholter Transfusion eine Alloimmunisation der betreffenden Patienten zu ver meiden (Abstract XIX Congress of the Ing. Soc. of Blood Transfusion, 19, S. 497, Poster P-TU-307-2 und P-TU-307-3). The filtration of leukocyte-containing blood fluids, e.g. B. human platelet concentrates, via cellulose filters for removal Existing leukocytes are carried out in order to avoid repeated Transfusion an alloimmunization of the patient concerned ver avoid (Abstract XIX Congress of the Ing.Soc. of Blood Transfusion, 19, p. 497, posters P-TU-307-2 and P-TU-307-3). The cellulose fibers consist of cleaned cotton, d. H. a natural product, which in terms of fiber diameter and length and thus also the filter properties are subject to large fluctuations. Furthermore Cotton is a biological material that despite previous cleaning to a certain extent bacterial is contaminated and the risk of endotoxins and pyrogenic reak contains in itself.
The invention has for its object to indicate how to Cellulose fiber comes out better for the intended use are suitable.
To this end, the present invention teaches from a procedural point of view that cellulose ester fibers can be treated with one in one At least non-solvent for cellulose ester dissolved ester splitting agent partially converted into cellulose fibers.
The invention is based on the previously unused knowledge from that with a chemical production of the cellulose fibers without great difficulties a very homo regarding the fiber dimensions gene and pharmaceutically perfect quality is achievable.
There are several options for the further configuration within the scope of the method according to the invention. So one will use cellulose acetate fibers as the starting material after preferred embodiment. The conversion of the cellulose ester to cellulose can in principle by all known methods, such as. B. with the aid of an acid hydrolysis catalyzed by acids, bases, metal ions, nucleophiles and enzymes, an ester alcohol hydrolysis hydrolysed by acids or bases or an ester reduction using LiAlH 4 (J. March, Advanced Organic Chemistry, Third Edition 1985 , John Wiley and Sons Inc.). To this extent, however, hydrolysis of the cellulose ester fibers with sodium hydroxide in ethanol is preferred. In terms of concentration, it is advisable to use a solution of 0.1 to 139 g / l, in particular 0.5 to 2 g / l, of sodium hydroxide in ethanol. This solution is expediently allowed to act on the latter in an amount of 0.1 to 100 ml / g, in particular 3 to 5 ml / g, of cellulose ester fibers. The hydrolysis is complete after two to three hours at room temperature; However, complete conversion of the cellulose ester fibers is not always necessary. After the treatment, the fibers are expediently cleaned by washing to remove sodium ethanolate which has not reacted and reaction products formed, e.g. B. Ethyl acetate and acetic acid. We recommend washing the fibers with ethanol, water and then again with ethanol.
The invention also relates to a filter for removing leu cocytes from leukocyte-containing blood fluids, especially blood platelet preparations containing cellulose in a filter container fibers that are produced by the method according to the invention. Cellulose fibers with a Y-shape and with an average diameter of 15 µm.
The invention is described below using an exemplary embodiment explained in more detail:
200 g Y-shaped cellulose acetate fibers with a constant diameter 17 µm were placed in a solution (1000 ml) for three hours of 1% (w / v) sodium hydroxide entered in absolute ethanol.
The hydrolysis or more precisely solvolysis was below at room temperature Exclusion of carbon dioxide carried out. After three hours it was The sodium ethanolate is removed by placing the fibers in a funnel were transferred. The fibers were then four times with 500 ml of absolute ethanol washed. Then the fibers for at least one hour with 1000 ml of an acidified solution of absolute ethanol (21 ml conc. HCl per liter ethanol) and after decanting the solution again as indicated above washed. Then the fibers were at 50 ° C to the weight con punch dried. Finally, the fibers were again with two Liter of salt solution (0.9% by weight NaCl), extensively with water as well finally washed with ethanol and dried again at 50 ° C net. The fibers had a diameter after this treatment from 15 µm.
The cellulose fibers thus produced were placed in a filter container entered and used to filter human platelet con centrates used to remove the leukocytes. The filter removed 97 to 98% of all existing leukocytes and no more than about 5% of platelets. The same result was achieved with Zel Lulose fibers obtained in a further, separately manufactured batch.
