Description

Osteoblastenvorläufer und Knochenbildung aus humane NabelschnurblutzellenOsteoblast precursors and bone formation from human umbilical cord blood cells

BeschreibungDescription
Anwendungsgebietfield of use

The invention relates to a method according to the preamble of claim 1.

Stand der TechnikState of the art

Delayed or non-existent consolidation of fractures and the reconstruction of bone defects represent a major challenge from both a surgical and an economic point of view. Since resources for autologous transplants are limited and there is a non-negligible risk in secondary interventions to obtain the body's own bone material, alternatives to autologous cancellous bone transplantation have always been sought (1). The precursors of mature osteoblasts are found in bone marrow and can be successfully isolated and enriched from it. It is known that placental residual blood from the umbilical cord of newborns represents an alternative to bone marrow as a stem cell pool for hematopoiesis (2). To date, the existence of osteoblasts or their precursors in umbilical cord blood has not been described.

Nachteile des Standes der TechnikDisadvantages of the state of the art

In the case of autologous cancellous bone grafting, resources are limited. Furthermore, the operation poses a significant risk and, as a second procedure, increases the patient's trauma.

Aufgabe der ErfindungObject of the invention

Alternatives Knochenwachstum (Ex vivo und In vivo).Alternative bone growth (ex vivo and in vivo).

Lösung der ErfindungSolution of the invention

Osteopoiesis by osteoblasts or their precursors derived from human umbilical cord blood cells.

Vorteile der ErfindungAdvantages of the invention

In contrast to the limited resources of autologous cancellous bone grafting, the existence of cord blood banks means that the resources for transplantation of osteoblasts and osteoblast precursors are almost unlimited. The large number of cord blood transplants available means that it is still possible to use HLA-matched osteoblasts and osteoblast precursors in order to minimize any problems with tissue compatibility. Furthermore, replacing autologous cancellous bone transplantation with allogeneic transplantation of osteoblasts and their precursors from cord blood offers economic advantages, as a second operation to remove the cancellous bone is not necessary. Furthermore, the patient is less traumatized.

BeispielbeschreibungExample description

Ex vivo, it has been possible to stimulate precursor cells of osteoblasts from umbilical cord blood to mature into osteoblasts and then expand them. This means that osteoblasts and their precursors from umbilical cord blood can be used as autologous or allogeneic transplants (tissue-compatible) for osteogenesis, e.g. in osteopetrosis patients. The cells can be administered to the relevant patients by infusion. For this purpose, the osteoblasts can be generated or expanded in vitro from the relevant precursor cells. This leads to a significant improvement in the healing of age-related fractures, accident-related bone fractures, closing of bone defects or skeletal extensions.
