The free (tetrabasic) tetraacid, N- (phosphonacetyl) -L-aspartic acid (which will be referred to in part as PALA in the following) is a well-known compound. The present The invention relates to new if- (phosphonoacetyl) -L-aspartic acid compounds (PALA compounds), especially disodium PALA and processes for the preparation of ii- (phosphonoacetyl) -L-aspartic acid compounds.
In particular, the invention relates to the preparation of the known tetrasodium salt and the new disodium salt of iä- (phosphonoacetyl) -L-aspartic acid on a large scale.

Mäuse mit einem Melanom B16 überlebten, wenn sie mit PALA-Tetranatriumsalζ (490 mg/kg i.p.) behandelt wurden,77 bis 86 % länger als Vergleichstiere.suffered to be extended by 64 ° / b when treated with the PALA tetrasodium salt in doses of 188 to 750 mg / kg ip. Lewis lung sarcoma is found to be very sensitive to the PALA tetrasodium salt in mice at ip doses of 240 to 490 mg / kg. Mice with melanoma B16 survived when they were treated with PALA-tetrasodium saline (490 mg / kg ip), 77 to 86 % longer than control animals.
While the synthesis of the tetraacid PALA proceeds smoothly, the production of the sodium salt is very problematic, especially in kg quantities. The process according to the invention is particularly suitable for producing such quantities.
Die neuen erfindungsgemäßen PALA- Verb indungen sind N-(Phosphonacetyl)-Jj-asparaginsäure - dinatriumsalz und der entsprechende P-lthylester und Dibenzylester;The new PALA compounds according to the invention are N- (phosphonoacetyl) -Jj-aspartic acid - disodium salt and the corresponding P-ethyl ester and dibenzyl ester;

Some of these new PALA compounds come in anhydrous form, some in solvated (including hydrated) iTorm. Both forms are suitable for the purposes of the invention. The disodium-PALA compound produced by the process according to the invention is a hydrate which differs from one batch to another in the water content of hydration, but usually contains approximately 0.2 to 2 mol v / water. The compound can also contain ethanol (e.g. about 0.1 to 0.5 mol), acetic acid (e.g. about 0.1 to 0.4 mol) and sodium acetate (e.g. about 0.2 mol) as a solvating agent. Acetic acid and ethanol can be removed by freeze drying. Two to three lyophilizations result in a solvent-free material. The disodium-PALA compound produced by this process can also contain substantial amounts of trisodium-PALA, namely up to JO to 40 % trisodium-PALA. All such solvates of disodium PALA and mixtures of disodium PALA and trisodium PALA fall under the designation disodium PALA (or an equivalent term) as used herein, since these product forms are intended for use as effective antitumor agents or preparations made therefrom are interchangeable. When used as an> anti-tumor agent, the disodium PALA according to the invention corresponds essentially to the known tetrasodium PAIA in terms of activity and toxicity. Because of its anti-tumor effect, disodium PALA can be used according to the invention in the form of medicaments, together with a pharmacologically acceptable carrier. The agents can also include antimicrobial agents and other anti-tumor agents. The agents can be prepared in any appropriate pharmaceutical form depending on the intended route of administration. Examples of such agents include solid agents for oral administration such as tablets, capsules, pills, powders and granules, liquid agents for topical or oral administration such as solutions, suspensions, syrups and elixirs, and agents for parenteral administration such as sterile solutions, suspensions or üjoaulsionen.

When used as an anti-tumor agent, those according to the invention Agent administered in a dosage such that tumor growth is inhibited. A recommended dosage for that Use as an anti-tumor agent (especially for solid (solid) Tumors as indicated above), in mammals, is 50 to 50 mg disodium-PALA / kg in a single daily parenteral dose in the course of treatment (e.g. in the form an intravenous infusion as a 27% aqueous solution). That new calcium salt of PALA corresponds pharmacologically for the purposes according to the invention the disodium PALA and can therefore instead whose or in combination with disodium PALA can be used in the remedies indicated above. The calcium salt of PALA has favorable solubility properties. It dissolves in Water and thus allows easy preparation of preparations, but it dissolves relatively slowly, whereby it is possible to see water soluble inorganic impurities.
The new disodium PALA is advantageously a mobile, free flowing, finely divided solid that can be easily handled, analyzed and weighed for the manufacture of pharmaceuticals. In comparison with the well-known tetrasodium PALA, it is relatively non-hygroscopic. Tetrasodium PALA absorbs moisture from the atmosphere 1.5 times faster than disodium PALA and is difficult to handle and analyze. While tetrasodium PALA is water soluble, disodium PALA beneficially has a solubility in water greater than 950 mg / ml. In the form of a 2 £ / <Jigen characteristic disodium PALA possesses (wt./vol.) Solution in water a pH of about 4, with the trisodium and tetrasodium PALA PALA solutions having a pH of about 6 or 9, is roughly comparable. Disodium PALA is also characterized by a 60-Mc nuclear magnetic resonance spectrum (OTIR spectrum), which typically shows a doublet corresponding to the methylene group (-GHp), in α-position to the -CH group,

An adaptation of the manufacturing process for the pure salts to a larger scale has overcome some potentially serious technical difficulties. The cyclohexylamine salt Dibenzyl-PALA is made by adding about 0.9 to about 1.0 equivalent of cyclohexylamine to an acetone solution of dibenzyl-PALA. The product is insoluble in acetone, while a high percentage of impurities remains in solution. The purity of the product is increased to a good value by recrystallizing from absolute Methanol.
Difficulties arise in the use of dioxane for production of Dibenzyl-PALA. Triethylamine hydrochloride is an insoluble by-product of the reaction and is in large volumes Solvent required to allow adequate agitation. In addition, the reaction is exothermic and so is the use of dioxane limits the possibility of cooling to about 12 ° O, since dioxane solidifies at this temperature.
Instead of dioxane, methylene chloride was used in this reaction used. This solvent offers the following advantages: (a) It is non-flammable .; (b) there are lower cooling temperatures possible ^ (c) the required volume is reduced by half; (d) the removal of the triethylamine hydrochloride by filtering off is not necessary as it is soluble in the reaction mixture and (e) it is no longer necessary da.s solvent to be removed before working up.

Further improvements in the process reside in the fact that the cyclohexylammonium salt can be hydrolyzed directly to the tetranatrium IBALA. As a result, the extra stage of releasing dibenzyl-PALA from the amine salt prior to hydrolysis is no longer necessary.
Another advantage is that the volume of water required for hydrolysis is 63 % less than that used in the original synthesis. This of course makes it possible to process larger batches with the same devices. In the laboratory or pilot plant, using a maximum of 50 liter flasks, the procedure was used to produce disodium PALA in quantities of approximately 2 kg. Taking into account all the modifications described, an approach using 190 and 380 l Pflauderer reactors was successfully carried out. Using the entire capacity, around 50 kg of the desired product could be produced with this system.
The inventive method is only by the size of the Plant restricted.
I'orschunga- bzw. Untersuchungszwecken großen Maßstabs.In the process according to the invention, the purity of the desired disodium PALA product can be improved to the point that it is suitable for parenteral administration Carrier suitable for the treatment of cancer in humans, in particular too. I'm for research or investigation purposes Scale.
This could be achieved by adding (i) the acetic acid and sodium acetate completely eliminated by turbulent precipitation; and (2) dibenzyl-PALA as the cyclohexylammonium salt isolated. In addition, the process has been optimized for ease of scaling up and process problems have thereby been solved.
