Description
The invention relates to the marking of organic connections with Technetium-99m according to the preceding claim. These compounds, in particular the metabolites of cell metabolism, occur, inter alia, in the form of alcohols, sugars, sugar derivatives such as nucleic acid building blocks, nucleoproteins, amino sugars, polysaccharides, glycosides and conjugates of steroids, hormones, pharmaceuticals, drugs and aromatic compounds, including heterocyclic hydroxyl compounds or α -Oxocarboxylic acids. Such labeled substances are required as radioactive spray substances in medicine and biology for scintigraphic and / or autoradiographic localization of concentrations of these substances in organisms, organs, tissues and cells on a large scale.
Techniques for marking with technetium 99m or for the production of injectable preparations ten of the Tc-99m became known, with which predominantly only those connections could be marked, which are characterized by distinguished acidic substituents (DE-OS 25 34 985, DE-OS 23 22 758). The marking was done by pushing back one Hydrolysis by reaction of a mixture of those to be labeled Substance with a reduc containing predominantly tin-II and a neutral to weakly acidic solution a connection of the technetium isotope.
Nature 215 (1967) 1397). A very broad group of biologically active compounds, their Labeling with technetium isotopes for reasons of outstanding radiation physical properties of the Tc with respect to the Quality of the scintigraphic image and the minimal beam oil pollution is desirable, contains as essential moles cooling components aliphatic or aromatic polyhydroxyver bonds (polyols), their labeling according to known methods, in which reduction of the pertechnetate in the presence of the labeling compound by ascorbic acid or Sn (II) verbin in acidic to neutral solution, only then to Target leads if the molecule is immediately over be neighboring strongly acidic groups (J. Nucl. Med. 7 (1966) 364 u. Nature 215 (1967) 1397).
Steigman, Internat. Sympos. on Radiopharmaceuticals, Atlanta, Georgia Febr. 12-15, 1974). Only a few other biologically active polyols could some markings are achieved (Richards and Steigman, Boarding school Sympos. on Radiopharmaceuticals, Atlanta, Georgia Feb. 12-15, 1974). So the yields were with the sugar alcohols xylitol, d-mannitol, sorbitol and galacticol between 50% and 70%, with hydroxycarboxylic acid lactic acid 7%, Glyceric acid 3%, ribionic acid 0%, xylonic acid 22%, d-glucon acid 82%, galactonic acid 0% and only with the d-gluco heptonic acid 94%. Seen as a whole, so far there have been none Methods have become known which are a reproducible markie tion with a high yield for all of the inputs allow named substances.
The method according to the invention enables an in vivo signal lization of changes in the biochemical reaction situation in order to thus all pathological conditions, which ultimately can be found in hi Express stological and anatomical changes through signaling and localization of the accompanying deviations the biochemical cell reactions and the biological regulations able to diagnose early and safely.
Bei dem einzustellenden pH-Wert 11-12 erfolgt dann die Markierungsreaktion mit außerordentlich hoher Ausbeute (90-99%).The invention is based on the object of a method creating the label of organic compounds, especially the metabolites of cell metabolism, with Techne tium-99m if a high marking rate is granted and which allows marking at the site of use. This object is achieved by the method according to the invention. The pH value to be set is 11-12 Labeling reaction with extremely high yield (90-99%).
To get the mark you want in medical practice on site the application, that is in the clinic immediately before the app lication to be able to carry out comfortably will be a Mi creation of the organic compound with the alkaline buffer and the reducing agent in solid, preferably lyophysisier ter, mold made and dispensed in ampoules or Carpulen® packs. The labeling reaction itself is only just before injection by adding a solution of technetium compound which, for example, from one of the known Tc genes is won.
To the high achievable with the inventive method Marking yields also do not exceed those for marking unsaturated orga required hydroxyl groups to be able to transfer niche substances, it is possible to hydroxylate them by a suitable chemical reaction and to make this marking process accessible.
For the pharmaceutical application of the herge according to the invention established Tc-labeled compounds result in numerous new opportunities, which include the following:
