Abstract
Optical arrangement with a control means and at least one illumination means whose light beam is scanned by optical imaging means by means of at least one electro-optical deflector and a sample is detected in a sample by the light beam generated light, characterized in that the high-frequency voltages for the deflector are generated multiple resonant.
Description
The present invention relates to an arrangement which allows a preferably microscopic object to be scanned at a very high speed.
Aufgabenstellung:Task:
In fluorescence measurements, especially in fluorescence microscopy, a fluorescence molecule is excited with an excitation light. The excitation energy is given off again a few nanoseconds (ns) later by the molecule with a somewhat longer wavelength. In confocal scanning microscopes, for example, a diffraction-limited laser beam is used as excitation light. Finally, in order to produce an image of the object, the beam must be moved relative to the object and the measurement signal must be registered synchronously.
Since fluorescence molecules do not always return directly to the ground state with fluorescence emission after the excitation but instead remain in a non-fluorescent state for a few milliseconds, it is more advantageous to scan the object several times with short exposure times than simply with a long exposure time. The molecules thus have time to return from the dark states until their repeated exposure. This will both reduce measurement time as a whole and reduce bleaching since less frequently the molecules will be exposed in a detrimental dark state.

To achieve even faster beam positioning, electro-optic deflectors (EOD) and acousto-optic deflectors (AOD) have also been used in the past. Such practically inertia-free deflectors use transparent materials, typically crystals whose refractive index can be influenced by electric fields or ultrasound fields and thus can deflect optical beams. AODs are limited in their speed practically only by the speed of sound in the crystal and thus achieve positioning times in the range of one microsecond. However, a disadvantage here for the fluorescence application is that the deflection is wavelength-dependent, so that this technique could not prevail in confocal scanning microscopy. Electro-optical beam deflectors are even faster and achieve deflections in the nanosecond range. However, their application is very limited by the small deflection angle. In a diffraction-limited laser scanning microscope, only a few 10 points are resolved, even at high coupling voltages of a few 1000 V, so that these beam deflectors have not yet been used effectively. In turn, control voltages in the 1000 V range can no longer be changed as quickly as desired, so that often the deflection speeds are limited by the driver systems.

It has been recognized that electro-optical beam deflectors, despite their small deflection angle, can be advantageously used in non-diffraction limited imaging methods such as the STED method. The resolution is up to about 10 sometimes as high as with diffraction-limited images, so that even with electro-optical deflectors many hundreds of pixels can be resolved. The image fields to be scanned are also usually only a few microns in size, so that electro-optical deflectors can be used effectively despite their small maximum deflection angle and the number of resolution points are not limited as in diffraction-limited microscopes.
In order to achieve a fast quasi-linear beam deflection, the invention proposes to generate the necessary voltages by means of multi-resonant circuits. The necessary signals and their phase relationships can be generated very easily in a function generator and amplified with a low-voltage amplifier. Finally, the high voltages of 1000 V and more arise due to the resonance peaks in the resonant circuits, which can be placed in a compact advantageous manner close to the EOD.
With the arrangement according to the invention, for example, microscopic STED images of 8 × 8 μm can be generated in a resolution of 512 × 512 dots at 1000 images per second.
With the large deflection frequencies, the transit times of the light in the microscope system and also the fluorescence lifetime are no longer necessarily negligible. The integration times of the pixels of 2 ns in the above example correspond to 30 cm for the back and forth of the light to the object and back to the deflector. This means that the detection light in the EOD is no longer completely descanned. If necessary, the detector has to be displaced relative to the EOD scan axis. By means of several detectors next to each other, it would also be possible at the same time to deduce the fluorescence lifetime and thus distinguish different dyes from one another. Also, the early or late fluorescent photons may no longer be registered at the correct position. Corrective measures may also be required for this. In a simple and advantageous case, one could no longer scan the detection light in the EOD and instead select a gap-confocal detection. The location assignment in the fast axis is done exclusively by the timing. Advantageously, a large-area detector in photon counting mode is used. Hybrid detectors such as the HPD R10467U-40 from Hamamatsu appear particularly suitable. In contrast to the GaAs photomultipliers, which can also be used, the hybrid detectors have much higher maximum count rates. In principle, of course, all other photodetectors such as diodes, APDs, PMTs or MPPCs, EMCCDs, etc. can be used without restriction of generality. Also the single photon counting mode is not obligatory, although mostly advantageous. Another variant could also be a linear array of small detectors located in the detection focus.
The STED method mostly uses pulsed lasers with pulse repetition rates of several 10 MHz. In order to avoid aliasing effects, which potentially lead to moiré patterns in the images, it is advantageous to provide a synchronization between the pulse frequency of the laser and the pixel clock of the scanner. By means of suitable temporal clock patterns, it is also possible to illuminate pixels, which are further away from each other in a temporally successive manner, in order, for example, to allow a relaxation of the excitation and dark states before a certain area is illuminated again.
Because of the short pixel integration periods, the number of detected photons per pixel is generally very low. For signal processing and coping with the high data stream of a few hundred million pixels per second, it is advantageous to compress the information for transport to a storage medium. In the case of single-photon detection, this could be done particularly advantageously by means of FPGAs and run-length coding, which can be lossless.
An image is built up in a STED microscope by superimposing some images of the object, since a single image is usually too noisy. The recording process of many successive images of the object could be started and stopped after reaching a desired signal to noise ratio.
