Abstract
Description
The invention relates to the use of highly ordered proteins, those with island-like clusters of a catalytically active tive metal are occupied, according to the first claim.
Under the term "Metallic nanostructures" are particularly in the publication special mechanical components in the field of microsystem technology nik, for example probes for electron microscopy or metallic tubes, understood. The basic building block of this construction parts are self-assembled, multidimensionally geometrically high ordered proteins such as B. bacterial cell envelope proteins, Collagen, actin and tubulin filaments and DNA strands. To Her The components are made of the proteins with a metal layer covered. The manufacturing process runs in two Steps: First, cluster-like metal island structures, for example platinum clusters, aba device. The metallic island structures are in a second Step used as nucleation centers to remove all of the protein by reducing dissolved metal ions with another Me tall to coat.
The document contains the remark made in the first step obtained regular arrangements of platinum clusters on the Proteins are "especially with regard to the application of the kata lytic activity of the deposited clusters interesting ".
The inventors of the cited WO 97/48837 are the authors of a publication publication (R. Kirsch et al. "Three-dimensional metallization of microtubules ", Thin Solid Films 305 (1997) 248-253), in which the manufacturing process of metallic nanostructures and the structure of which is discussed in detail. In this publication chung is explained in particular for which Pro record the catalytic activity of the deposited clusters is settable. Thereafter it follows that the catalytic activity the deposited cluster is exploited to by reduction of the metal ions of a dissolved salt, those with the catalytic ones proteins occupied by active platinum or palladium clusters as completely as possible to be coated with another metal, d. H., to metallize the proteins. In experiments with Protei those that were populated with very few metal clusters, it was found that the metallization of the proteins excludes Lich used on the catalytically active metal clusters. In the The remark contained at the beginning of WO 97/48837 is there to understand in such a way that the catalytic activi the cluster works on the reduction of metal ions and the associated deposition of the resulting metal on the with the Clusters of occupied proteins. In the catalyti mentioned The activity of the clusters is therefore not one Catalysis in the real sense, in which the catalyst in the chemical reaction remains unchanged. Rather, the Me tall cluster in the course of the reduction of the metal ions from the emerging nickel metal covers, making their catalytic Activity is inevitably canceled because they are then for the metal ions to be reduced are no longer accessible.
The invention is based, for the higher ranking task Proteins with clusters of a kata lytically active metal are occupied, a new possible use propose.
The object is achieved by the one described in the first claim Use resolved. In the further claims are preferred Refinements of the use proposed according to the invention specified.
In principle, all in the proteins mentioned above are used become. Microtubules are particularly preferred. Microtubules consist of globular tubulin (alpha, beta dimers, molecular weight 110,000) and are formed through a self-assembly process. Adjacent heterodimers are opposite each other by approx. 10 ° offset from the horizontal, making it a helical arrangement ingestion. Tubulin dimers build with alternating al pha, beta-tubulin binding protofilaments with a diameter of approx. 4 nm, which bind together longitudinally (more typical wise 13 or 14) and microtubules. The microtubules have an outer diameter of approx. 25 nm and can be used Reach length between 1 and 10 µm or more. The micro tubules are dynamic structures, i. that is, they can rapidly changing needs in the cell through assembly or customize disassembly processes.
In the case of using the clustered microtubules as Ka Talysatoren is such a constant disassembly process however not desirable. The microtubules, but also other pro tones are therefore preferably chemical and therefore structural fixed. Is particularly suitable for chemical fixation Glutaraldehyde. This makes the protein molecules intra- and intermolecularly linked, whereby the assembly / disassembly dynamics and the chemical and mechanical stability of the proteins is improved. Through the Fixation can occur, for example, when the metal clu is deposited the reaction temperature on the proteins up to 100 ° C be raised without attacking the metal / protein structure becomes. This is both for the representation of the clustered Proteins as well as when used to perform kata lysed reactions advantageous.
The proteins must have an island-like cluster be catalytically active metal. The type of metal depends on the intended purpose, d. that is, after the chemical reaction to be catalyzed. Metals that catalyze a variety of chemical reactions are at for example, platinum, palladium, gold, ruthenium or rhodium neither alone nor in combination with each other. Metal cluster with a diameter of 1 to 6 nm are for the invention Suitable for use. Preferably all are with the Clusters can be assigned to the protein with the clusters sets so that the highest possible cluster density on the protein surface is achieved.
According to the Pro with the metal clusters teine used as a catalyst for chemical reactions. Trä In principle, gerfixed catalysts have the advantage that they simply recovered after completion of the catalyzed reaction can be. In the use according to the invention To understand the term "catalyst" in the usual way, näm Lich that the cluster-occupied proteins in the course of the by them catalyzed reaction remain unchanged and not - as with State of the art initially cited - even a change be subjected.
The clustered proteins to be used according to the invention offer significant advantages: because the ratio of surface to Volume increases with decreasing particle size ell the material properties of small particles more strongly from the Surface atoms as determined by the classic lattice atoms. The number of active centers on the surface required for the Ak activity of a heterogeneously catalyzed reaction is crucial are therefore generally reduced with decreasing particle size to. In the use according to the invention, the metal clusters are during the reaction chemically and sterically as catalytically active tive surface freely available. The agglomeration of the metal clu ster is prevented by its fixation on the protein surface different. The protein serves as a spacer. Chirality at Proteins are both created by asymmetric carbon atoms (Primary structure) as well as the helical folding (secondary structure). The clustered proteins can therefore affect the stereoselectivity of chemical reactions.
It has been shown that the cluster-occupied proteins in particular which are suitable for the catalysis of chemical hydrogenation. Examples of such chemical hydrogenations are hydrogenation of carbon multiple bonds and the hydrogenation of nitro groups.
The invention is described below using exemplary embodiments and explained a figure.
The figure shows the results obtained in Example 3.
The assembly of microtubules from a protein solution with a final concentration of 1 mg / ml tubulin was carried out in a buffer solution of 20 mM PIPES (1,4-piperazine diethanesulfonic acid, pK a 6.8), 80 mM NaCl, 0.5 m MgCl 2 , 1 mM EGTA (ethylene glycol bis (2-aminoethyl) tetraacetic acid) by adding 10 mM GTP (guanosine triphosphate) and 10 µM taxol at a temperature of 37 ° C. In order to monitor the assembly, the turbidity of the assembly solution was recorded with a spectrometer at 360 nm until saturation was observed (approx. 20 min). The microtubules are first fixed in 0.05%, then after 5 min in 3% glutaraldehyde with vigorous stirring. After 20 min, the reaction solution is dialyzed against 1000 ml water / buffer 10: 1 overnight to remove the remaining glutaraldehyde from the reaction mixture.

3 ml of different colloidal gold suspensions (Polyscience: Au particle diameter 5 nm, 10 nm, 20 nm and 60 nm) are concentrated to approx. 200 µl with the centrifuge, mixed with 100 µl assembled and fixed microtubules and within 30 min immobilized on the microtubules at room temperature. The approx. 2 nm Au colloids are visualized in the presence of 100 µl of the microtubule solution by reducing 100 µl of a HAuCl 4 solution (1 g / ml in water) with 20 µl of a NaBH 4 solution ( 0.5 eq) at 0 ° C. The Au colloids thus produced serve as nucleation seeds in the reduction of 400 µl Na 2 PdCl 4 solution (0.0125 M in a buffer / water mixture of 1:10, pH 6.8) with 40 µl of an aqueous solution the trisodium salt of citric acid (0.25 M in water) at 70 ° C for 30 min. In order to remove excess, non-reduced metal salt from the reaction mixture, dialysis is carried out overnight.
Die Gesamtmetallmasse wird durch ICP-Messungen (Inductively Coupled Plasma) bestimmt.780 .mu.l of the solutions prepared according to Examples 2a and 2c are mixed with 49.22 ml of water and 10 ml of an aqueous crotonic acid solution (0.723 M) and subjected to 1,013 bar of hydrogen at 25 ° C. The decrease in H 2 per unit time and metal mass is measured. The total metal mass is determined by ICP measurements (Inductively Coupled Plasma).
The results obtained according to this example are in the figure shown. The figure contains a diagram in which on the Or dinate the catalytic activity in [ml / mg.min] and on the Ab The diameter of the metal clusters is plotted. The Volume is converted to standard conditions (Nml). The dunk The bars indicate the initial hydrogenation activity based on the Ge velvet metal mass determined by ICP. The bright ones Bars relate to the initial hydrogenation activity the palladium mass. The Pd / Au ratios are above the bars (Molar ratio). With the Pd / Au ratio of 3.9 only the particle size of the Au portion (20 nm), not the Particle size of the Pd portion (1 to 2 nm) can be determined exactly; therefore "20 + x" was given on the abscissa.
The diagram shows that the catalytic activity decreases with the increase in the cluster or particle size.
