When carrying out the process according to the invention, the saponification of the nitrile group, cleavage of the protective group R 2 and the coumarin ring closure take place in one go. The reaction products are in a conventional manner, for. B. by cooling and suction, isolated.
The inventive method is generally carried out in such a way that 1 mol of the substituted / i-phenylacrylic acid nitrile of the formula 11 in an aqueous medium, optionally under pressure, in the presence of at least 2 moles of one Mineral acid, for example sulfuric acid, hydrochloric acid, hydrobromic acid, hydriodic acid, hypochloric acid or phosphoric acid, optionally in the presence of a water-miscible organic Solvent, e.g. B. acetic acid, or a water-immiscible solvent, such as chlorobenzene or o-dichlorobenzene, for about 1 to 20 hours at about 80 to 200 ° C, preferably at 100 to 150 ° C, heated and then isolated the reaction product in the usual way.
From Swiss Patent 341 834 it was already known to use u-aryl- / i- (2-alkoxy-4-acylamino) acrylonitrile in a two-stage reaction to the corresponding 3-substituted 7-aminocoumarins transfer, using strong acids in an anhydrous medium as condensation or splitting agents were and after condensation, the aylamino group saponified to the free amino group would. In contrast, it was new and completely surprising that 3-substituted 7-aminocoumarins in a simple manner in high yields and purities in a one-step process by condensation can be obtained in aqueous medium. The method known from US-PS 33 22 794 for the preparation of 7-aminocoumarins from m-aminophenol and / f-hydroxy-u-phenylacrylonitrile or / i-hydroxy-ii-phenylacrylic acid ethyl ester is carried out in the presence of anhydrous aluminum chloride.
It has over the method according to the invention a narrow range of applications, is more difficult to carry out on an industrial scale, and delivers significantly worse bulges.
The process of BE-PS 7 10 223 uses the same starting compounds as the process the US-PS 33 22 794, but avoids anhydrous aluminum chloride as a condensation agent. Through this the disadvantages of a two-stage reaction and even poorer yields have to be accepted will. If a single stage is used (Example 15), the yield drops further. The range of applications is not greater than that of the process according to US Pat. No. 3,322,794. The process of OE-PS 2 5G950, too gives significantly poorer yields than that according to the invention Process whereby the required intermediates, 2. B. p-Acetylaminosalicylanil, the is reacted with phenylacetic acids to the 3-phenyl-7-aminocoumarins, also only with bad ones Yields are accessible.
Overall, it can be stated that the inventive Process has the surprising advantages over the prior art, in a smooth and simple reaction with broad applicability from simple, easily accessible starting materials to provide 3-substituted 7-aminocoumarins in high yield.
Die Ausgangsverbindungen der Formel 11 lassen sich beispielsweise dadurch erhalten, daß man ein Toluolderivat der allgemeinen FormelThe starting compounds of the formula 11 can be obtained, for example, by a Toluene derivative of the general formula

umsetzt, worin R1 eine gegebenenfalls durch eine Alkyl- oder Alkoxygruppe oder Halogenatome substituierte Phenyl- oder Naphthylgruppe oder den über ein Kohlenstoffatom mit der CH2-Gruppe verknüpften Thienyl-, Pyridyl- oder Benzimidazolylrest bedeutet, und anschließend das Reaktionsprodukt in üblicher Weise, beispielsweise durch Abkühlen, gegebenenfalls Ansäuern und Absaugen, isoliert.reacted, in which R 1 denotes a phenyl or naphthyl group optionally substituted by an alkyl or alkoxy group or halogen atoms or the thienyl, pyridyl or benzimidazolyl radical linked to the CH 2 group via a carbon atom, and then the reaction product in the usual manner, for example isolated by cooling, optionally acidification and suction.
This is generally carried out in such a way that at temperatures of about 50 to 120 ° C " a solution of the toluene derivative (III) in an organic, water-miscible solvent, for example an alcohol or dimethyl sulfoxide, with at least one for reducing the nitro group or oxidation of the methyl group, the required amount of aqueous alkaline polysulphide solution is added, the whole thing about 0.5 to 3 hours at 50 to 120 "C holds, then, optionally after previous cooling, about 0.7 to 1 mol of the nitrile (IV) is added, optionally heated for a long time and then the reaction product in the usual way Particularly suitable solvents for this are lower alcohols, for example methanol and ethanol and propanol and dimethyl sulfoxide.
The alkali polysulfides used are those obtained by dissolving sulfur in aqueous alkali hydroxide and / or alkali sulfide are available, the sodium or alkali compounds being preferred Potassium compounds are used.
