VERFAHREN ZUR HERSTELLUNG VON ÄTHINYLDERIUTEN DER. STMOIDE
Die Erfindung betrifft ein. neues Verfahren zur Herstellung von Äthinylderivaten
der Steroide, d.h, zum Finführen
.Gruppe in das Molekül der St6,0i)ide. PROCESS FOR THE PRODUCTION OF ETHINYLDERIUTS OF. STMOID

Die in der letzten Zeit immer wichtiger werdenden Äthinylderivate
der Steroide werden nach den in der Literatur beschriebenen Verfahren im allgemeinen
derart hergestellt, daZ die als Ausgangsstoff angewendeten Steroide bzw. deren substituierte
Derivate in Anwesenheit von geeigneten Kondensationsmitteln, meistens von Alkalimetallen
oder deren Verbindungen, mit Acetylen umgesetzt werden. Dabei wird zuerst durch
die Reaktion des Kondensationsmittels mit dem Acetylen das ent--
sprechende Alkaliacetylid gebildet, welches dann mit dem
zu äthinylierenden Steroid in Reaktion tritt, wodurch_das
Alkalisalz des gewünschten Äthinylderivats entsteht, aus wel-
chem durch Zersetzen der Idkaliverbindung das gewünschte End-
produkt erhalten wird.
Bei den aus der Literatur bekannten Ausführungsweisen
dieser Reaktionen wird das mit dem Steroid in Reaktion tre-
tende Alkaliaeetylid aus einem Alkalimetall, Alkalihydrod
oder Alkalialkoholat mit gasförmigem Acetylen derart herge-
stellt, daß man in die Schmelze des Alkalimetalls oder in das
die Alkaliverbindung enthaltende Reaktionsgemisch Acetylen-
gas einleitet. Die Bildung des Acetylids ist eine Gleichge-
wichtsreaktion: deren Gleichgewicht im Interesse der wirt-
schaftlichen und erfolgreichen Ausführung des Verfahrens in
die
Riahtung der, Acetylidbildung verschoben werden muß.
Dieses
Verschieben des Gleichgewichts kann im Fall von Anwendung von
polaren Lösungsmitteln, wenn das ,Alkalimetall in der Form
von
Hydroxyd oder Alkoholat anwesend ist und das Reaktionsgemisch
ein homogenes System bildet, durch die Anwendung eines groJen
Überschusses vom Acetylengas erreicht werden. Wird aber in
einem ,polaren Lösungsmittel gearbeitet, wobei das filkalime--
tall z.B. in der Form eines Ukaliamids angewendet wird, dann
scheidet sich das in ,polaren Lösungsmitteln unlösliche Aeetylid
aus dem Reaktionsgemisch ab, es entsteht also ein heteroge-
nes System, -wodurch die Verschiebung des Reaktionsgleichge-
wichts in die Richtung der Acetylidbildung von vornherein
gewährleistet ist.
Bei den bisher bekannten Verfahren wird aber die praktische Ausführung
der Äthinylierungareaktionen durch zahlreiche Schwierigkeiten erschwert und der
Umsetzungsgrad wird auch durch das Eintreten von Nebenreaktionen herabgesetzt, Wird

Es müssen ferner bei der derartigen Ausführung der Reaktion ziemlich lange Reaktionszeiten
(20 bis 130 Stunden) =gewendet- werden, wodurch Jm alkalischen Medium Zersetzungsreaktionen
auftreten, schädliche und verfärbende Nebenprodukte gebildet werden. Bei der in
apolaren Lösungsmitteln, also in heterogenen estem ausgeführten Äthinylierungsreaktion
treten zwar die obenerwähnten Nebenreaktionen weniger auf, es werden aber die Reaktionsgeschwindigkeit
und der Konversionsgrad im heterogenen System erheblich herabgesetzt, Weitere Nachteile
können in solchen Fällen aus der Gefährlichkeit des =zuwendenden Kondensation: '',tels
(z.B. in Fall von Alkaliamiden), ferner aus den mit der Handhabung des als Lösungsmittel

Es wurde gefunden, daf die zur Bildung von Radikalaniwleu_#A#@gen

radikalanionbildenden aromatischen Verbindungen verläuft nämlich rasch und in einer
mit den Augen leicht verfolgbaren (die ursprünglich grüne Lösung entfärbt sich bei
der Bildung des Acetylids) und durch das Ausscheiden des Acetylids aus den Reaktionsgemisch
in praktisch quantitativer Weise. Die durch die Reaktion entstehende Dihydroverbindung
ist ein gutes Lösungsmittel für die übrigen Reaktionsparner und durch ihre Mitwirkung
wird das entstandene Acetylid in feiner Dispersion gehalten, Durch die Einwirkung
des in feiner Verteilung und infolge dc;r stabilisierenden Wirkung der Dihydroverbindung
in groSer Konzentration anwesenden iLlkaliacetylids wird dann das gewünschte äthinylierte
Endprodukt rasch (in günstigen Fällen innerhalb von 30 bis 60 Minuten) und mit sehr

Die praktische Ausführung des erfindungsgemäßen Ver-
fahrens kann vorteilhaft in der folgenden Weise erfolgen:

der organischen Phase erhaltenen Rückstand die nicht reagierte aromatische Verbindung
(z.B. Naphthalin) und deren noch anwesendes Dihydroderivat durch Extraktion mit
einem g»oicneten Lösungsmittel, z.B, mit Dichlormethan entfernt und das als Rückstand

das rohe Produkt nur in minimalem Maß durch Nebenprodukte verunreinigt ist. Dieser
Vorteil ist einerseits der in protonenfreiem Lösungsmittel, in heterogener Phase,
mit fein verteiltem Alkaliacetylid durchgeführten Reaktion und andererseits der
kurzen Gesamtreaktionsdauer (höchstens 4 bis 5 St=--den, gegenüber den bisher üblichen
?.0 bis 130 Stunden) zu verdanken, Die weiteren Einzelheiten des erfindungsgc;mä
)en Verfahrens werden durch die nachstehenden 'Beispiele veranschaulicht.
Beispiel 1:
In einer Lösung von 10,26 g Naphtalin in. 80 ml Totra-
hyärofuran werden bei Zimmertemperatur, unter Stickstoffstrom
1984 g Natriummetall aufgelöst, was 1,5-2 Stunden in
Einspruch
nimmt. Dann wird Acetylengas bei Zimmertemperatur in das
Reaktionsgemisch eingeleitet, bis die ursprünglich dunkel-
grüne Farbe des Reaktionsgemisches verschwindet (in etwa
20-30 Minuten). Das das entstandene Natriumacetylid enthaltende
Reaktionsgemisch wird dann mit der Lösung von 5,68 g Östra-
--1, 3, 5 (10) -trien-3-methoxy-17-on in 180 ml Tetrahydrofuran
versetzt und das Einleiten von Acetylengas irird noch eine
weitere Stt-.ude fortgesetzt.
Der pH- Wert des Reaktionsgemisches wird mit 3 N Salz--
säure unter Stickstoffstrom auf 6 bis 6,5 gestellt. Die orga-
nische Phase wird von der wä2uigen Phase getrennt, über
wasserfreien Natriumsulfat getrocknet und in Vakuum bei
5000 verdampft. Aus den Naphthalin., Dihydronaphthalin
und
3-Päethoxy-17«--äthinyl-östra-1,3,5(10)-trien-3,17ß-diol ent-
haltenden Reaktionsgemisch werden das Naphthalin und das Di-
hydronaphthal in bei Zimmertemperatur mit r_.- 'T - ptar ausgewa-
schen; nach Filtrieren und Trocknen des Rückstandes werden
5,7 g 3-rJlethoxy-17a-äthinyl_östra 1,3,5(10)-trien-3,17ß-diol
(92 % d.Th.) erhalten; F. 149-151°C. Das Gemisch dieses Pro-
dukts mit Einen authentischen Muster derselben Verbindung
zeigt keine Schmelzpunktdepression.

Beispiel 2:
In einer Lösung von 20,52 g Naphthalin in 160 ml
Tetrahydrofuran werden unter Stickstoffstrom, bei Zimmer-
temperatur 3,68 g Natriummetall gelöst. Nach dem Auflösen
wird Acetylengas bis zum Verschwinden der grünen Farbe (et-
wa 30 Minuten) in das Reaktionsgemisch eingeleitet, dann
wird unter weiterem Einleiten von Acetylengas die Lösung von
12j72 g 6,5 (10) -Östren--3, l7-dion-3-dir.iethylac etal in
120 ml
Tetrahydrofuran zugesetzt. Nach einer halben Stunde wird das
Einleit3n von Acetylen unterbrochen und das ReaktionsGemisch
unter Stickstoffstrom mit r,9 ml 3 N Salzsäure versetzt und
15 Minuten unter schwachem Rückflur erhitzt Nach dem Abküh-
len wird das Reaktionsgemisch mit festem Natriumhydrogenkar-
bonat neutralisiert, die organische Phase wird abgetrenntt
über wasserfreiem Natriumsulfat getrocknet und im Vakuum
unter 500C zur Trockne verdampft. Das Naphthalin und das Di--
hydronaphtahlin werden aus der Rückstand mit einem 1:4 Gemisch
von Benzol und Heptan ausgewaschen, Nach Filtrieren und
Trocknen werden 11,45 g 17a hthinyl -17ß-hydroxy- A.4-östren-
-3-on (96% d.Th.) erhalten, F. 201-2040C, Das GLmisch dieses
Produkto mit einem authentischen Muster derselben Verbindung
zeigt keine Schmelzpunktdepression.
Analyse:

Beispiel 3:
12t72 g A5(10)-Östren-3,17-dion-3-diriethyla^etal
werden in der im Beispiel 2--aeswlariebenen Weise äthinyliert.

derselben Verbindung zeigt keine schmelzptuaktdepression.

Das nach Beispiel 2 hergestellte, Natriurtacetylid ent-
haltende Reaktionsgemisch wird mit der Lösung von
1392 g
@r_dros-i.p3-en-3ß-o1-17-orz-3°aceta t in 100 ml Tetrahydrofuran
versetzt tuid das Einleiten von Acetylen wird bei ZiL-tertenpe-
r2°@ttr weitere 2 Stunden fortgesetzt. Das Reaktionsgemisch
wird
in der in Beispiel 2 beschriebenen ;eise aufgearbeitet p mit
den; Unterschied, da.-) das Naphthalin und das Tetrahy dronap-lä.tha--
lin aus dem. verdampften 'eichstarzd des Reaktionsgemisches

Das nach Beispiel 1 hergcatellte, I3atriunaeetilyd
enthaltende Reaktionsgemisch wird mit der Lösung von 5,16 g
Östr-4-en-17-on in 60 ml Tetrahydrofuran versetzt und d:s
Einleiter- von Acetylengas wird eine weitere Stunde fort-
gesetzt. Das erhaltene Natriunsalz von 17a Äthinyl-östr-,4-
-en-17P-ol wird iü Reaktionsgemisch durch Zugabe von einer
10%igen wä£<rigen Amoniunchlz@iiaaösung hydrolisiert. Die
ab-
getrennte organische Phase wird mit einer 10 öigen wälirigen
Ammoniunchloridlösung, dann mit einer gesättigten Kochs,-.lz--
lösung gewaschen,- über wasserfreier. Natriumsulfat getrocknet
und i. Vakuum verdampft. Aus den Rückstand werden das
Naph-
thalin und das Dihydronaphthalin ait n-Heptan ausgewaschen.
Es werden 5,4 g 17a-Äthinyl-östr-4-en-17ß-ol (95% d.Th,) er-
halten, F. 158-160°e, Das Gemisch dieses Produktes mit einen
authenti8cheäi Muster derselben Verbindung zeigt keine Schnelz-
punktdepression.
