16 70 494.2) sind neue borhaltige heterocyclische Verbindungen und Verfahren
zu deren Herstellung beschrieben worden. Diese Verbindungen, die der.allgemeinen
Formel
worin Z für einen ankondensierten o-Phen-ylen-, Naphthylen- oder Thienylenring steht,
der gegebenenfalls durch einen oder mehrere Substituenten aus der Gruppe, welche
Alkylreste, Halogenatome, verätherte Oxy- oder Mercaptogruppen, Alkyl-, Aralkyl-oder
Arylsulfonylgruppen oder Nitrogruppen umfaßt, substituiert ist, H 1 einen
gegebenenfalls durch Alkylreste, verätherte Oxy- oder Mercaptogruppen, Halogenatome,
Amino- oder Acylaminogruppen, Trifluormethylgruppen oder Nitrogruppen substituierten
ein-oder mehrkernigen aromatischen oder heteroeyclischen Rest bedeutet und R 2 fär
ein Wasserstoffatom, ein Katior oder für den Rest
steht ,
entsprechen, weisen u. a. ausgeprägte ,Jirkungen
gegenüber Mikroorganismen, insbesondere gegenüber gramnegativen Bakterien auf. Besonders
wertvoll sind diejenigen Verbindungen der Formel I, in denen R 1 die Gruppe
der Formel II
worin R 3 für eine Amino- oder eine Ac71.aminogruppe, R für ein Halogenatom,
eine Trifluormethy'gruppe 4 - Z
oder einen niederen Alkylrest und R
5 für e#n Wasserstoffatom, ein Halogenatom, eine TriUuorme-"-hylgruppe, einen
niederen Alkylrest oder einen ankondensierten o-Phenylenring stehen, bedeutet. Vorzugsweise
steht darin R 3 für eine Aminogruppe, R 4 für ein Chlor- oder Bromatom und
R 5 für ein Wasserstof f atom. Bedeutet H 3 eine Acylaminogruppe,
so leitet sich der darin #L-enthaltene Acylrest vorzugsweise von einer niederen
a14 phatischen, d.h. von einer einen niederen Alk-,#,lirest enthaltenden Carbonsäure
ab. In den Verbindungen der Formei I steht R 2 bevorzugt fÜr ein Wasserstoffatom
oder fär ein Kation, wobei im letzteren Fall salzartige Verbindungen resultieren,
die wesentlich besser als die Produkte mit R 2 = H löslich sind. Z

Es
wurde jetzt Überraschenderweise gefunden, daß die Verbindungen der Formel I, insbesondere
'die-bevorzugten Glieder dieser Gruppe von Verbindungen, die antibakterielle Wli--kung
von Antibiotika zu steigern vermögen und daß Ant-ibiotika die Wirksamkeit der Verbindungen
der Formel I erhöhen, d.h."da13 sich diese Verbindungen gegenseitig in hohem Maße
hinsichtlich ihrer antibakteriellen Wirkung synergistisch beeinflussen. Dieser günstige
Einfluß der Verbindungen der Formel I wurde z. B. für die antibakterielle Wirkung
von 6-Acylaminopenicillansäuren ("Penicillinen"), Tetracyclinen, Chloramphenicol,
Spiramycin, Kanamycin, Streptomycin, Dihydrostreptomycin, Golistin und weiteren
Antibiotika experimentell bewiesen. (Die vorstellend genannten liamen der Antibiotika
sollen für die Definition der vorliegenden Erfindung auch z. B. Salze und weitere
antibakteriell wirksame Derivate der Antibiotika, wie z. B. Ecter des Chloramphenicols,
Mannich-Derivate von Tetrac-,vcljnen usw. sowie Gemische von Antibiotika wie
Z. B. Streptomycin-Penicillin-Kombinationspräparate umfassen). EJInige der
dabei erhaltenen Ergebnisse sind in den folgenden Tabellen zusammengestellt. D--rin
bedeutet "Verbindung A" das 1-Hydro-xy-2-(2'-Chlor-Lk'-aminophenylsulfonyl)-benzo-2,3,1-diazaborin,
das in Form seiner Lösung in Kaliumphosphatpuffer vom pH 7, d.h. in Form

Es ist besonders hervorzuheben, daß durch den synergistischen Einfluß
der Verbindung A nicht nur die mehr oder weniger stark ausgeprägte antibakterielle
Wirkung de-. Antibiotika gesteigert wird, sondern daß darüber hinaus sogar für die
Praxis als völlig unzureichend anzusehende Wirkungen von Antibio-bka (vergl. Tabelle
1, Spiramycin sowie Tabelle 2, Tetrac-yclin).so gesteigert werden, daß die
Antibiotika dann in solchen Konzentrationen die Testkeime hemmen, die therapeutisch
ohne weiteres anwendbar bzw. erreichbar sind. Diese synergistische Beeinflussung
wurde auch im Tierexperiment gefunden. Beispielsweise wurde als DC 50 ( das
ist die Dosis der Prüfsubstanz in mg/kg Körpergewicht -nach deren Verabreichung
50 % der Versuchstiere die Infektion überleben) bei oraler Verabreichung
an mit E. coli infizierte Mäuse für die Substanz A ein Wert von
165 mg/kg, für Chloramphenical ein Wert von 64 mg/kg und fÜr Ampicillin ein
Wert von 90 mg/kg gefunden. Wurden aber z. B. Gemische gleicher Gewichtsteile
dieser Wirksubstanzen zur Therapie verwendet, so Überlebten 50 919 der Tiere
die Infektion bei einer oralen Verabreichung von je 32 mg/kg der Substanz
A und Chloramphenicol bzw. von je 60 mg/kg der Substanz
A und Ampicillin. Entsprechende Ergebnisse wurden auch erhalten, wenn die
Wirksubstanzen in anderen Mengenrelationen verabreicht wurden. Außer mit der "ubstanz
A, d.h. dem 1-Hydroxy-2-(2'-chlor-4f-aminophenylsulfonyl)-benzo-2,3,1-diazaborin
wurden synergistische Einflüsse auf die antibakteriellen Wirkungen von Antibiotika
auch mit anderen Verbindungen der Formel I beispielsweise mit 1-Hydroxy-2-(2'-chlor-4'-acetamino-phenylsulfonyl)-benzö-2,3,1-diazaborin,
1-Hydroxy-2-(p-tolylsulfonyl)-benzo-2,3,1-diazaborin, 1-Hydroxy-2-(2'-chlor-4'-acetaminophenylsulfonyl)-thieno-E2,3-el-2,3,1-diazaborin,
1-Hydroxy-2-(2'-methyl-4'-aminophenylsulfonyl)-benzo-2,3,1-diazaborin, 1-Hydroxy-2-(21-trifluormeth71-4'-aminophenylsulfon
-,71)-benzo-2,3,1-diazaborin,
1-Hydroxy-2-(21-chlor-4'-amino-phenylsulfonyl)-7-methyl- oder auch -7-methoxj-benzo-2,3,1-diazaborin
festgestellt. So hemmt z. B. das 1-H-,ydro--#.-j-2-(2'-chlor-4'-acetaminophenylsulfonyl)-benzo-2,3,1-diazaborin
("Verbindung B") E. coli in einer Konzentration von 60 y/ml. (Durch
Ampicillin wird - vergl. Tabelle 1 - dieser Keim in einer Konzentration
von 4 y/ml gehemmt). Eine Hemmung von E.coli wird aber auch durch Konzentrationen
von 8 y/ml der Verbindung B und 2 y/ml Ampiciliin bzw. 20 y/ml der Verbindung
B und 1 y/ml Ampicillin bewirkt. B. proteus mirabilis wird durch die Verbindung
B in einer Konzentration von 80 y/ml und von Ampicillin in einer Konzentration
von 6 y/ml gehemmt. Eine Hemmung des Wachstums dieses Keimes bewirken aber
auch jeweils folgende Konzentrationspaare:

Auch aus diesen Ergebnissen resultiert die ausgeprägte gegenseitige synergistische
Beeinflussung dieser Wirk-Stoffe. Die Verbindungen der Formel I, in denen R2 für
ein Wasserstoffatom steht, bilden, wie bereits oben erwähnt, mit Basen Salze. Zum
Gegenstand der vorliegenden Erfindung gehören daher auch Salze der Verbindungen
der Formel I mit basischen Antibiotika, wie z. B. Streptom7cin, Dih-ydrostreptom-ycin,
Kanamycin, Spiramycin bzw. basischen Antibiotikaderivaten-wie z#. B. Pyrrolidinomethyltetracyclin
usw. Derartige Salze können..in an sichbekannter Weise z. B. dadurch hergestellt
werden, daß man ein Salz des kiitibiotikumis --mi-t -einer Verbinaung der -Formel,
I" in der R 2 für ein Kation steht, in Gegenwart eines-Lösung.snittels umsetzt.
Man kann aber auch so vorgehen, daß man in einer Lösung der freien Base des Antibiotikums
eine Verbindung
der Formel I, in der R 2 für ein Wasserstoffatom
steht, löst und dann die so erhaltene Lösung als solche zur Therapie verwendet oder
das Produkt z. B. durch Gefriertrocknung isoliert. Beispielsweise können Streptomycin
oder Dihydrostreptomycin mit bis zu drei Mol eIner Verbindung der Formel I Salze
bilden. Für die Herstellung von Salzen, in denen die antibakterielle Wirkung der
Antibiotika erhöht ist, reicht es aber in den meisten Fällen hin, nur eine oder
zw*ei der basischen Gruppen der Antibiotika mit den Verbind * ungen der Formel
I in Salze zu überführen. Gewünschtenfalls können dann die freien basischen Gruppen
der Antibiotika mit anderen Säuren (z. B. Pantothensäure, ichwefelsäure) zu Salzen
verbunden sein. Die erfindungsgemäßen Produkte (Gemische aus Verbindungen der 1,ormeil.
I mit Antibiotika und/oder salzartige Verbindungen dieser Komponenten) sind auf
Grund der überraschenden synergistischen Beeinflussung der Komponenten hinsichtlich
ihrer antibakteriellen Wirksamkeit-den Einzelkomponenten Überlegen. Sie können aber
in gleicher Weise wie die Einzelkomponent-eii zur Therapie eingesetzt werden, d.h.
sie können z. B. oral, rektal, parenteral oder auch lokal angewendet werden. Die
Produkte lassen sich ohne weiteres in die für diese Applikationsarten üblichen pharmazeutischen
Zubereitungen (z. B. Tabletten, Kapseln, Sirupe, Lösungen, Suspensionen,. Suppositorien,
Globuli, Salben, Cremes, Puder usw.), einarbeiten, wobei sich die für eine Stabilisierung
usv. erforderlichen (an sich bekannten-), Maßnahmen im wesentlichen. nach der Natur
des in deia Produkten enthalteneu Antibiotikums# rtchtet auBe da#die Verbindungen-
der- Formel I- stabiler -als. äie Antibiotika ])it :rölgenden,Beispie14 dienen zur
weiiteren. ErläuteTung der Forf indung.
Beispiel 1:
Gleiche Gewichtsmengen von Chloramphenicol und 1-Hydroxy-
2-(2'-chlor-4'-aminophenylsulfonyl)-benzo-2,3,1-diaza-
borin werden innig vermischt. Je 250 mg dieses Gemisches
werden in Gelatinekapseln abgefüllt.
Beispiel 2:
Man verfährt wie in Beispiel 1, verwendet jedoch statt
des Chloramphenicols Tetracyclin-Hydrochlorid.
Beispi 1
Man stellt ein Gemisch aus 5 g Chloramphenicol und

1-Hydroxym-2-(2'-chlor-4'-aminophenylsulfonyl)-benzo-2,3,1-
diazaborin her und trägt dieses dann in 90 g einer ge-
schmolzenen handelsüblichen Suppositorienmasse auf der
Basis von Glycerinfettsäureestern ein. Das Gemisch wird
gut gerührt und dann in jeweils 2,5 g des Gemisches
fassende Suppositorienformen abgefüllt.
Beispiel 4:
Je 17,5 g Propicillin-Kalium und 4-Hydroxy-2-(2'-chlor-
4'-aminophenylsulfonyl)-'benzo-2,3,1-diazaberin,
25 g
mikrokristalline Cellulose, 33 9 sprühgetrockneter Milch-
zucker, 6 g getrocknete Maisstärke und 1 g Magnesiumstea-
rat werden vermischt, zu bikonvexen Drageekernen gepreßt
und in üblicher Weise dragiert.
Beispiel 5.-
7 g Propicillinkalium, 7 g 1-Hydröjr#2-(2'-chlor-4'-
aminophenylsulfony,l)---benzo-2,3,1-diazaborin, 1 g
Natrium-
carboxyiüethylcellulose, 0,2 g Kaliums-orbat,
0,05 g-
Saccharin-Natrium und 20 g Zucker werden innig vermischt.
Beim Auffüllen mit Wasser auf 100 mi erhält man einen Sirup, der
je 350 mg der Wirkstoffe-in 5 ml enthält. Beispiel 6:
Aus
10 ml 0,4n-Natronlauge und 1 g 1-Hydroxy-2-(2'-ählor-4'-aminophenylsulfonyl)-benzo-2,3,1-diazaborin
wird unter Rühren und leichtem Erwärmen eine Lösung hergestellt und diese nach Abkühlen
mit 1,4 g Chloramphenicolhemisuccinatnatrium versetzt. Nach Sterilfiltration
erhält man eine Lösung, die als solche zur Therapie eingesetzt werden kann.
Beispiel '7: Man verfährt wie in den vorhergehenden Beispielen, verwendet
jedoch statt des 1-Hydroxy-2-(2'-chlor-4'-aminophenylsulfonyl)-benzo-2,3,1-diazaborins
jeweils 1-Hydroxy-2-(21-chlor-4'-acetaminophenylsulfonyl)-benzo-2,91,1-diazaborin
oder 1-Hydroxy-2-(2'-chlor-4'-acetaminophenylsulfonyl)-thieno-'!2,3-el-2,3,1-diazaborin
oder 1-Hydroxy-2-(21-me-bhyl-4'-aminophenylsulfonyl)-benzo-2,3,1-diazaborin.
Beispiel 8:
Eine Lösung von 1 g Dihydrostreptomycinbase in 20
ml Wasser wird unter RÜhren bei 30 -,35 0 C mit 1,15 g 1-Hydroxy-2-(21-chlor-4'-aminophenylsulfonyl)-benzo-2,311-diazaborin
versetzt. Unter Bildung des Salzes mit Dihydrostreptomycin geht dabei das Benzo-2,3,1-diazaborinderivat-na-ch
kurzer Zeit in Lösung. In dieser Lösung kann gewünschtenfalls das Salz eines Penicillins
gelöst-werden,
Beis-Piel 9:
Man verfährt wie in Beispiel
8, jedoch unter Verwendung von 1,03 g 1-Hydroxy-2-(p-tolylsulfonyl)-benzo-2,3,1-diazaborin.
Beispiel 10:
Man verfährt wie in Beispiel 8, jedoch unter Verwendung
von 1,23 g 1-Hydroxy-2-(2'-methyl-4'-acetaminophenylsulfonyl)-benzo-2,3,1-diazaborin.
Beispiel 11.
Man verfährt wie in einem der Beispiele 8 - 10, jedoch unter Verwendung einer
Lösung von 1 g Streptomycinbase in 20 ml Wasser.
BeisEi.21 12:
Eine Lösung von 1 g Kanamycinbase in 20 ml Wasser wird auf 30 - 3500
erwärmt. Unter gutem RÜhren gibt man 1,38
g 1-Hydroxy-2-(2'-chlor-4'-aminophenylsulfonyl)-benzo-2,3,1-diazaborin
