Description
The invention relates to a process for the production of 6-aminopenicillanic acid by enzymatic means Cleavage of phenoxymethylpenicillin or one of its salts by means of fusaria or yeast.
A number of processes have already become known for the production of 6-aminopenicillanic acid. So it succeeds after the so-called "precursorless fermentation" using the for the Penicillin formation common fungi in a culture medium of the usual composition 6-aminopenicillanic acid to create. But one can also - and the method according to the invention represents a further one Development of this type of manufacturing process for 6-aminopenicillanic acid represents - from preformed ones Penicillins run out and in these penicillins by enzymatic means a cleavage of the Cause side chain. The cleavage of phenoxymethylpenicillin has so far been carried out in particular by means of penicillin amidases has been made from mushrooms and there have been a number of mushrooms for that enzymatic cleavage of phenoxymethylpenicillin is used. One has already tried by adding of phenylacetic acid or its derivatives in the multiplication of a suitable for the cleavage of penicillin G. E. coli strain shows the yields of 6-aminopenicillanic acid in the cleavage of penicillin G to increase.
When using the known for the cleavage of phenoxymethylpenicillin into 6-aminopenicillanic acid Fungi always have a more or less long lag phase. During this lag phase is in the presence of the penicillin to be cleaved (substrate) no enzyme activity observed; however, it comes with it to an undesired destruction of the penicillin to be cleaved.
Attempts at the cleavage of phenoxymethylpenicillin using actinomycetes and fungi to influence by the fact that one phenoxyacetic acid or 6-aminopenicillanic acid at the same time admits the substrate, showed that an addition of phenoxyacetic acid is ineffective, while a those of 6-aminopenicillanic acid result in a reduction in yield [Proc. of the Royal Soc, Vol. 154, pp. 522 to 531 (1961)]. It was also found that phenoxymethylpenicillin-cleaving fungi, which had been grown in a medium modified by the addition of phenylacetic acid, did not provide an increased yield in the subsequent cleavage process.
Es wurde nun gefunden, daß es bei bestimmte zur Phenoxymethylpenicillinspaltung geeigneten i\ kroorganismen, nämlich Fusarium anguicides, Fui rium lycopersici, Fusarium oxysporum, Fusarii semitectum oder Torulopsis albida gelingt, die bei achtete Lagphase (innerhalb der keine Enzymtätigk zu beobachten ist) dadurch abzukürzen, daß m während einer vorhergehenden Wachstumsphase < ringe Mengen von 6-Aminopenicillansäure oder tIt has now been found that certain at-organisms to Phenoxymethylpenicillinspaltung suitable i \, namely Fusarium anguicides, Fu i Ministry lycopersici, Fusarium oxysporum, semitectum Fusarii or Torulopsis albida succeed, (can be observed within the no Enzymtätigk) at stayed lag phase shorten characterized that m during a previous growth phase <small amounts of 6-aminopenicillanic acid or t
ίο ren Salzen zusetzt. This is what makes it open during growth to adaptive formation of an active enzyme. When using the ^ The measure thus sets the enzymatic splitting activity after the introduction of the enzyme carrier fc The period of cleavage begins immediately after the addition of a substrate, and height is obtained in less time Yields of 6-aminopenicillanic acid.
The process according to the invention is characterized in that Fusarium anguioides, Fus rium lycopersici, Fusarium oxysporum, Fusariu semitectum or Torulopsis albida are allowed to wax under normal cultivation conditions in the presence of 0.001 to 2% 6-an nopenicillanic acid or a salt thereof before adding phenoxymethylpenicillins . In particular, a concentration of 0.2 ! 6-aminopenicillanic acid is used.
In this adaptive formation of the cleavage d for cleavage which follows during the cultivation of the enzyme carrier c Phenoxymethylpenicillins suitable activator by adding 6-aminopenicillanic acid As tests have shown, it is not a general principle that can be used, as with others, Phenox methylpenicillin-splitting microorganisms due to this measure an abbreviation of the lag phase or Shortening and increasing the split cannot be observed.
The adaptation can be advantageous; perform successive passages, in wt Chen the added amount of 6-aminopenicilla acid is increased; but you can also do one adaptation z.
