Description
Method of Making an Antacid The invention relates to on a process for the production of an antacid from carbonate-containing aluminum-maanesium-hydroxide gel.
Stomach deacidifier, the magnesium carbonate and aluminum hydroxide at holding are known. In British Patent 745 493, for. B. Mixtures these two components are described as containing about 30 parts by weight of magnesium carbonate and contain 23 parts by weight of aluminum hydroxide. The magnesium carbonate can be included be wholly or partially eietzt by calcium carbonate.
There are also known antacid absorptive therapeutics, the aluminum silicate and magnesium salts which, calculated from the residual oil, contain about 16 ° / o Al 2 O 3, 260 / o IgO and 57% SiO2 have (German patent script 941 864).
According to the aforementioned British patent, the antacid is used thus produced, that an aqueous solution of a caronate on the one hand and a aqueous solution of aluminum and magnesium and / or calcium salts on the other hand Warming up with each other will be missed, the entire mass after completion of the precipitation cooled, decanted, then slurried and decanted several times with water. Dagewashed moist product is then dried at 50 to 0 ° C so quickly that the The antacid effect of the aluminum hydroxide is not damaged.
These known gastric desiccants containing 17 to 26 parts by weight of Al (OH) 3 are said to be the well-known ones made from the sonats and then mixed with one another in particular, because the buffering effect of the Al (OH) 3 lasts for a relatively long period of time, namely in the p = range of 3.5 to 4, 5, tested according to the in the J @ nal of Pharmacie and Pharmacologie, 5, 1953, @ 686 to 691, described method.
According to the German Paltschrift 941 864 are allowed to flow ice diluted solution mixture of alkali silicon, and alkali hydroxide in a dilute Dissolving salts of aluminum and magnesium containing about 30% water Mixed silicates are produced, the residue on ignition about 16% Al2O3, 2 / o MgO and 5711 / o Contains Si02. The combination of the anhydrous basic substance corresponds to r formula AlMg2Si309.
Der Wirkungsbereich : r Entsäuerung bzw. The range of action: r deacidification or
The buffering effect should be almost complete and longer with these preparations than with Use of the individual components NaHCO3, CaCO3, @CO3 or Mg2Si2O8 in the physiological pH range of interest from 3.5 to 6. From the pH curve, however, is to see that the pH already 25 minutes after the addition of 25 ccm 1/10 n-HCl to over 6 increases and only drops to 4.5 in the course of about 80 minutes. The high silica content this well-known preparation gives this one physiologically not negligible acidic character, so that the antacid effectiveness is not only weakened, but is transformed into the opposite.
In the journal "Drug and Cosmetic Industries", 1949, p. 10, it is stated that the pH range for an antacid should be between 3 and 5. If this is above 5, gastric secretion is undesirably stimulated, so that additional amounts of gastric juice may be secreted.
Magnesiumphosphat enthaltende Antacida zeigen keine bessere Wirkung als Magnesiumcarbonat enthaltende Mittel. Antacids containing magnesium phosphate do not show any better effect as agents containing magnesium carbonate.
This prior art shows that effective antacids have been tried based on magnesium and / or calcium carbonate and aluminum hydroxide or Aluminum silicate on the one hand by mixing the separately produced components and on the other hand to produce them by coprecipitation of the same from aqueous solutions with coordination of the Mg, Al and water content in the end product to a special one good antacid effectiveness in the pH range from 3.5 to 4.5 or from 3 to 6.
The invention is based on the object of producing an antacid, Although this is also particularly effective in the preferred pH range of 3 to 5, the but also maintains the effect considerably longer than the known preparations, as can be demonstrated on the basis of comparative tests.
Those made by the process of the present invention Antacid agents are fundamentally structured differently in terms of their composition, than they 95 to 65 ° lo carbonate aluminum hydroxide gel and only magnesium carbonate in addition to a characteristic water content of about 5 to 250 / o in dried Condition included.
This is the highly absorptive property and the long-lasting Deacidification effect limited.
To obtain these products, the method of the invention is used a carbonate-containing aluminum hydroxide gel with a suspension of magnesium carbonate mixed in water in a ratio corresponding to 1 to 35 parts of MgO to 100 Al2O3 and then the separated mixture, optionally after aging, except for a remainder Dry water content of about 5 to 25 percent by weight.
According to a particularly advantageous embodiment, the formation the magnesium carbonate for the process product done in such a way that one the magnesium carbonate precipitates in a suspension of an aluminum hydroxide gel, washes out the soluble electrolytes and then the mass down to a water content dries from about 5 to 250 / o.
Aluminum hydroxide gels containing carbonate are used to treat ulcers on the digestive organs or other symptoms caused by overacidification of the stomach, suitable. The gels neutralize the excess stomach acid to one pH around 4.0, which makes them mildly astringent and analgesic are. On the one hand, it is important for the success of such a treatment that the deacidifying agent does not temporarily produce a strong alkalinity of the stomach, which disrupts the Processes of the stomach was only aggravated, and on the other hand the depreciation of the Acidity for as long as possible. The deacidifying agent must not be used have high activity, and it should be as long as possible constant Have an effect.
In the case of many known agents, the effectiveness is now enhanced by a strong one Aging decreased during storage, so the products in their application react only slowly and therefore only slowly with the dilute hydrochloric acid of the stomach slowly come into effect. It has been observed that the neutralization of the Gastric acid required time; a neutralization is used as a comparison acid measured with 1 / lo n-hydrochloric acid at 37 ° C, increases up to eight times if the product has been dried to such an extent that the loss on ignition of the product is only was about 42 weight percent.
After that it was to be expected that with further drying the reactivity of the aluminum hydroxide gel is a further slowdown, especially since in the event of prolonged storage.
Surprisingly, they showed according to the method of the invention produced relatively strongly dried products even after prolonged storage as fast reaction time as freshly precipitated aluminum hydroxide gel without simultaneously creating an undesirably high pH in the gel.
The acidity of the stomach is reduced to a pH below 4.5, normally buffered to about 4.0, and this pH value is maintained over a long period of time (several Hours) with simultaneous sufficient reaction speed. Dehydration down to a little below 501o can take place if a Temperature of 100 ° C is not maintained for a long time.
The aluminum hydroxide gel used as the starting material is preferable one that meets the requirements of the pharmacopoeia. With advantage becomes a Gel used, which contains a carbonate dissolved in water, so that a basic Aluminum carbonate with 10 to 40 percent by weight carbonate, calculated as CO and based on Ais0.
Magnesium chloride or magnesium sulfate or nitrate are used as magnesium salts used, but they have no advantage over the chloride.
In the following examples, the amounts are parts by weight, so far nothing else is noted.
B e i s p i e l 1 6, 24 Teile wasserfreie Soda wurden bei Zimmertemperatur in 107, 4 Teiltn Wasser gelöst. Example 1 6, 24 parts of anhydrous soda were used at room temperature dissolved in 107.4 parts of water. 25.3 parts aluminum chloride solution, density approx 1, 2 at 20 ° C, were added with shaking over about 36 minutes, a carbon-containing aluminum hydroxide gel fell through. The pH was initially 10, 85 and after adding the last remainder of the aluminum chloride solution approximately 5, 4. 4 parts of magnesium chloride (MtgCl 6H., O) were dissolved in 36 parts of water Lung was added to the suspension of the aluminum hydroxide glass. A 10% sodium carbonate solution (1 part of NasCO3 in 8.9 parts of water) was added over a period of 8 minutes. The pH changed from 5.4 initially to about 8.1. The precipitation mix was aged for 1 hour and then passed through a plate and frame filter press at etva 3 at filtered, the filter cake washed with water until it is free of soluble salt wax which was then taken out of the filter press4 cake dried at about 40 ° C to a constant weight. The weight of the dry solids was about 100% of the wet cake.
The dried filter press cake was then put in a high speed Grind the hammer mill and pass through a sieve with a mesh size of about 0.87 mm sieved. The sieved product is a finely divided, white, winches powder the following composition and properties: Al20s ... 44.0 ° / o MgO ..................... 4, 31 ouzo carbonates, calculated as CO) 14, 6 ° / o Ci ...................... 0, 08 ° / o Water ................... 19.6% acid consumption ('/ n-HCl / g) 282 ccm reaction rate a) after deer ice. 22 seconds b) according to Mutch .... 54 seconds Apparent density ... 0, 196 pH of 4% aqueous suspension ............. 9, 1 wet separated by 325 mesh .................. 75, 53% In general, the new product improves on '/ xo n-HCl in the Reh @ method in no more than 60 seconds to de Toepfer's reagent end point (pH 2.8 to 4, 2).
The methods for determining the reaction rates according to Reheis and Mutch by means of acids are as follows: According to Reheis, 0.5 g of Algol make 100 ccm 1/10 n-HCl given at 37.5 ° C and with two drops of Toepfer's reagent as an endpoint indicator offset. The whole thing is shaken and kept at a temperature of 37.5 ° C, until the indicator changes color. It becomes what is required for this response Time determined.
The Mutch test is similar to the Reheis test with the difference that 0.1 g Al (OH) and 30 ccm viol-Cl are implemented here.
Example 2 The same components as in Example 1 were used, with the exception that the precipitations are kept independently of one another in separate containers and the washed and filtered products are then placed in a Waring blender were intimately mixed. The mixed material was then, as described in Example 1, dried. The dried gel substance contains 17.50% water.
Aluminum hydroxide gel generally undergoes drying a change that affects its effectiveness as a deacidifier.
The gastric enzyme pepsin sets the antacid effect of the aluminum hydroxide gel down, as from Murphy, Journ. Amer. Pharmaceutical Assoc., Sc. Ed., 41, p. 361, 1952; the dried one made by the process of the invention However, gel does not significantly affect the effectiveness of the pepsin. this can be seen from test table I. This contains comparative data on the antacids Effect of 1. an aluminum hydroxide gel, 2. of dried aluminum hydroxide gel, 3. from a dried gel made from basic aluminum hydroxide and magnesium carbonate according to the invention.
From the table it can be seen that the values for the according to the invention produced product similar in terms of reaction rate to that of dried Are aluminum hydroxide gel. The pH drops with the dried aluminum hydroxide gel however, at first it decreases much more and lies far during, the entire time of the experiment below the desired optimum. The optimum should be between a pH value of 3 and 5 are, as already explained. According to the method according to the invention In contrast, the product produced keeps the desired pH value practically above that entire trial period. In addition, it shows little aging compared to that undried aluminum hydroxide gel.
The test procedure used is a modification of that used by Holbert, Noble and G r o t e method specified for determining the antacid effectiveness (See Joum. American Pharmaceutical Assoc., Vol. 37, p. 292, 1948). Here is apart from the use of an artificial gastric juice with 2 g pepsin per liter Acidification with hydrochloric acid to the pH value 1, 5 the test mixture in certain A part of the mixture is taken at intervals. This is done to reduce the loss of material, as it happens in the stomach when food is delivered to the intestine in practice, to imitate, whereby the addition of artificial gastric juice takes place continuously and a part can also be discharged continuously.

The obtained product (6417 g) became up to a weight of 879 g dried and tested after pulverization.
The preparations produced according to Examples 3 to 5 were compared with a 18.9 g dried aluminum hydroxide gel with 5310 / o A120.- salary and 8.34 g magnesium carbonate (42§ / o MgO) by mixing the components in one about 450 g shake bottle for about 15 minutes produced product, whose MgO-Al2O3 ratio is 35 to 100, which contains practically no water. The test results are shown in Table II.

From the table it can be seen that both the antacid samples with a low (5: 100) as well as those with a high ratio (35: 100) from MgO to A1203 retain their antacid activity considerably longer than those made by the usual simple mixing of the previously dried components became. In addition, the pH is kept at an optimum between 3 and 5, the is a characteristic of a good antacid supplement.
