Die Erfindung betrifft ein Verfahren zur Herstellung von N-Alkoxy-jÖ-phenylalkylaminen und deren Additionssalzen mit bronchienerweiternder Aktivität und relativ geringer Toxizität der allgemeinen FormelThe invention relates to a process for the preparation of N-alkoxy-jÖ-phenylalkylamines and their addition salts with bronchodilator activity and relatively low toxicity of the general formula
in der R Wasserstoff oder eine Methylgruppe, R' Wasserstoff, eine Methyl- oder Äthylgruppe und R" eine Methyl- oder Äthylgruppe bedeutet, das darin besteht, daß man in an sich bekannter Weise, a) /S-Phenyläthylhalogenide, -sulfate oder -toluolsulfonate mit niederen Alkoxyurethanen in Gegenwart von Alkali zu N-Alkoxy-ß-phenylalkylurethanen umsetzt und diese zu N-Alkoxy-ß-phenylalkylaminen verseift oder b) Phenylacetoxim am Sauerstoff alkyliert und den erhaltenen Äther katalytisch hydriert und die nach a) oder b) erhaltenen Reaktionsprodukte gegebenenfalls am Stickstoffatom alkyliert und gegebenenfalls die erhaltenen Basen mit anorganischen oder organischen Säuren in die Additionssalze überführt.in which R is hydrogen or a methyl group, R 'is hydrogen, a methyl or ethyl group and R "denotes a methyl or ethyl group, which consists in the fact that in a manner known per se, a) / S-phenylethyl halides, sulfates or toluenesulfonates with lower alkoxy urethanes in the presence of alkali to form N-alkoxy-ß-phenylalkyl urethanes converts and these to N-alkoxy-ß-phenylalkylamines saponified or b) phenylacetoxime alkylated on oxygen and the ether obtained catalytically hydrogenated and the reaction products obtained according to a) or b) optionally am Nitrogen atom alkylated and optionally the bases obtained with inorganic or organic Acids converted into addition salts.
Compounds prepared in this way form addition salts with mineral acids and organic ones Acids.
In general, the N-alkoxyamines and their salts thereof made by the process have nontoxic salts pharmacological properties similar to those of known corresponding phenylethylamines are. They are therefore useful for increasing and maintaining blood pressure, various show Activity levels as sympathomimetic drugs for use as local vasoconstrictors, can relax the muscular system, the bronchi and the intestinal tract, for enlargement the pupils of the eyes and produce reactions similar to those which are obtained by stimulating the adrenergic nerves. But they are important greater value than the corresponding known amines, as they have a lower toxicity and decreased Have side effects.
The alkoxyamines produced by the process according to the invention are relatively weak bases compared to the corresponding known amines in which instead of the alkoxy group Hydrogen is bonded to the nitrogen atom. So you can z. B. not titrated in aqueous solution will. However, they manage to produce N-alkoxy - /? - phenylalkylamines with strong acids

Despite Due to their comparatively weak basicity, the compounds form both slightly acidic addition salts with mineral acids such as hydrochloric acid, sulfuric acid and phosphoric acid, as well as with organic acids, such as tartaric acid, acetic acid, citric acid, maleic acid and succinic acid.
Contain the N-alkoxy-a-methyl '/ S-phenylethylamines an asymmetric carbon atom and are therefore clockwise and counterclockwise as racemates optical isomers obtained in a manner known per se by crystallization of their Salts can be separated with optically active acids.
In the production of N-alkoxy-a-methyl- / 3-phenylethylamines it is advisable to first convert phenylacetone into the phenylacetoxime, the oxime, by reaction with hydroxylamine then with a lower alkyl halide, sulfate or toluenesulfonate in the presence of alkali to implement the lower alkyl esters of the oxime, which is now catalytically reduced with hydrogen will.
If compounds alkylated on the nitrogen atom are used, d. An alkoxyamine can be used instead in a manner known per se with formaldehyde and formic acid to form N-methyl-N-alkoxy-ß-phenylalkylamine methylate.
Es wurde ohne weitere Reinigung wie folgt weiterverarbeitet.1205 g of phenylacetoxime, 1000 g of hexane and a solution of 430 g of potassium hydroxide in 3000 g of water were placed in a flask equipped with a stirrer, dropping funnel and reflux condenser. 1800 g of dimethyl sulfate were then gradually added at about 40 to 45 ° C. over a period of about 7 hours. The hexane layer was then separated from the aqueous layer and washed with 1000 g of water. After the hexane had been separated off, the remaining material was distilled and gave 779 g of a product which distilled at about 80 ° C. and 3 to 5 mm pressure. This product was found to be a 55-45% mixture of phenylacetoxime O-methyl ether and phenylacetone. It was processed further as follows without further purification.
Das erhaltene Destillat ist ein racemisches Gemisch von zwei optisch aktiven Isomeren, die durch bekannte Methoden der fraktionierten Kristallisation ihrer Salze mit einer optisch aktiven Säure getrennt werden73.5 g of the mixture were dissolved in 190 ml ^ normal ethanolic hydrochloric acid, with 1.1 g Platinum oxide added and under a hydrogen atmosphere at an initial pressure of about 4 at stirred. The reaction was complete when a pressure drop of about 1.6 at had occurred. so the catalyst was filtered off and the solution was diluted with 800 g of water. The mixture was extracted with two portions of 170 g of benzene, the aqueous layer separated and with a solution made alkaline by about 15 g of potassium hydroxide in 100 g of water. The released oil was used twice extracted with 170 g of benzene and the combined benzene extracts are distilled. After separating the Benzene, 15 g of a fraction boiling at 72 to 92 ° C about 7 mm was collected. Another distillation yielded 14.4 g of N-methoxy-β-phenylisopropylamine with a boiling point of 3 = 50 to 54 ° C. The amine was more inorganic or organic in aqueous solutions Acids soluble. It is a relatively weak base that cannot be titrated in aqueous solution could, but gave an analytical value of 97.5% by non-aqueous titration method. - phenylisopropylamine were heated with 4.0 g of aqueous 33% formaldehyde and 7.5 g of 90% formic acid at about 100 ° C. in the course of 2 hours. After the evolution of carbon dioxide had ceased, the reaction mixture was made strongly alkaline with 25% strength caustic soda. The hereby liberated oil was extracted with ether, the ether evaporated and distilling the remaining residue as oil, yield 4.7 g of N-methyl-N-methoxy-JOE phenylisopropylamine from Kp.3 = were obtained 84 to 88 0 C.
N-Methyl-N-methoxy-jö-phenylisopropylamin kann auch dadurch hergestellt werden, daß man N-Methoxy-/?-phenylisopropylamin mit Methyljodid in Äthanollösung unter Rückfluß in Gegenwart von Kaliumcarbonat umsetzt.N-methyl-N-methoxy-jö-phenylisopropylamine can can also be prepared by using N-methoxy - /? - phenylisopropylamine with methyl iodide in ethanol solution under reflux in the presence of potassium carbonate implements.

Das aus diesem Amin hergestellte Hydrochlorid hatte einen Schmelzpunkt von 94,8 bis 95,70C.8 g of N-methoxy-ß-phenylisopropylamine, prepared as in Example 1, were refluxed with 10 g of ethyl iodide in ethanolic solution in the presence of potassium carbonate for 2 to 3 days. Then, unreacted starting material was removed by treating the reaction mixture with sodium nitrite solution (NaNCfe). In this way, N-ethyl-N-methoxy-iS-phenylisopropylamine was obtained in a yield of 45 percent by weight, with a purity of 97% and a refractive index nf of 1.4920. The hydrochloride prepared from this amine had a melting point of 94.8 to 95.7 0 C.

Das kristalline Salz derPhosphorsäure schmilzt bei 140 bis 142° C.0.5 mol of N-methoxy urethane in 250 g of ethanol were treated with a solution of 33 g of potassium hydroxide in ethanol and then with 92 g (0.5 mol) of /? - phenylethyl bromide. After refluxing for a further 4 hours, the mixture was cooled to room temperature and the precipitated inorganic salts were filtered off. The alcoholic solution was then distilled at about 100 ° C. in order to remove most of the ethanol, then the residue was diluted with 170 g of benzene and extracted with three portions of 50 g of a 5% strength aqueous sodium hydroxide solution in order to separate off all acidic products. The distilled benzene extract left 76 g of aqueous oil. 175 g of ethanol and 146 g of 45% strength aqueous potassium hydroxide solution were added to this, and the mixture was refluxed for about two and a half hours, then diluted with 300 g of water and extracted three times with 85 g of benzene. The combined benzene extracts were again extracted three times with 100 g of 3η hydrochloric acid. After this acidic solution was made alkaline with aqueous potassium hydroxide, an oil separated from, which was extracted with benzene and distilled to give 17.7 g of N-methoxy-jS-phenylethylamine were obtained from Kp.3 = 68-78 0C. It forms a crystalline hydrochloride, mp = 97.8 to 100 0 C, which precipitates by treatment of a ether solution of the base with gaseous anhydrous hydrogen chloride. The crystalline salt of phosphoric acid melts at 140 to 142 ° C.
The N-methoxy urethane required for this was produced in the following way, for which patent protection is not sought: 52.5 g of N-hydroxy urethane (0.5 mol) were added to a solution of 33 g of potassium hydroxide (0.5 mol) in 250 g of ethanol given. Then 75 grams of methoxy iodide (0.53 mol) was added. The reaction took place with evolution of heat, the temperature of the mixture rose to about 85 0 C. Then the mixture was stirred and heated approximately 75 minutes under reflux. The mixture thus obtained was used as a starting material.
Aus dem rohen Reaktionsprodukt wurde dann ein Gemisch des O-Äthyläthers Following the procedure of Example 1 Phenylacetoxim was reacted with diethyl sulfate in benzene solution at 50 to 70 0 C in the presence of sodium hydroxide. A mixture of O-ethyl ether was then made from the crude reaction product
