Description
Process for the production of vitamin B ", and its stable solutions Vitamin B12,. differs from the cyanocobalamin known as vitamin B12 in that it contains the cobalt in divalent form. Vitamin B12 r is against Oxygen is extremely sensitive and is very easily lost in aqueous solution Formation of aquocobalamin, also called vitamin 1312, is oxidized.
On the other hand is vitamin B12. an important intermediate for the Synthesis of derivatives of vitamin B12.
J. Sci., Band 26 (1952), S. 555, beschrieben. So far there is no known method by which it is possible to obtain vitamin B12 r produce in a durable form. The previously known methods start from vitamin B12, which is dissolved in water and then catalytically hydrogenated. However, that has the disadvantage that the vitamin B12r is obtained in an aqueous solution, in which it, as explained at the beginning, easily merges into aquocobalamin. One is therefore forced to use the further reactions with aqueous vitamin B12 .. solutions in complicated apparatus to be carried out in the absence of oxygen. Such an apparatus is for example in the journal Iowa State Coll. J. Sci., Vol. 26, p. 555 (1952).
No. 2,738,301 mentions that vitamin B12 can also be hydrogenated in alcohol; but the process actually described in this patent only relates to the production of vitamin B12 a. The reactions are carried out in aqueous solution and the only reaction product (vitamin B12 a) is recrystallized from water. This patent specification does not convey the knowledge that the hydrogenation of vitamin B12 in absolute alcoholic solution surprisingly produces vitamin B12r and that this compound is stable in this solution. It has now been found that it is surprisingly possible to produce vitamin B12r and its stable solutions by catalytic hydrogenation of vitamin B12 in alcoholic solution if the hydrogenation is carried out in an absolute monohydric or dihydric alcohol containing up to 10 carbon atoms performs. According to the invention, it is also possible to precipitate the vitamin B12r from the alcoholic solution obtained after the catalytic hydrogenation with an organic solvent in which vitamin B12r is insoluble and to separate it by conventional methods.
As a solvent for the catalytic hydrogenation, besides aliphatic Alcohols such as methanol, ethanol, propanol, butanol, also cycloaliphatic, aromatic or araliphatic alcohols are used, such as phenol, benzyl alcohol, ß-Phenylethyl alcohol or substituted by hydrocarbon radicals derivatives of named alcohols.
Furthermore, divalent charcoal, for example ethylene glycol or Butanediol, can be used as a solvent. Also unsaturated mono- or bivalent All charcoals can be used for the reaction, such as butynediol, acrylic alcohol, Pentinol etc.
The use of n-propyl alcohol or is particularly advantageous Benzyl alcohol as a solvent, as alcohols contain more vitamin B12 in tiles dissolves more than 50% better than in water.
The hydrogenation can take place at room temperature or at slightly higher temperatures Temperatures, e.g. B. at about 50 ° C. It is not necessary, to carry out the hydrogenation under pressure, but the reaction can also be carried out under increased pressure Pressure, for example at 6 atm, take place.
The usual hydrogenation catalysts are suitable, for example Noble metal catalysts such as palladium or platinum, but also Raney nickel. The catalysts can be used on the usual carriers.
The reaction times during the implementation are between a few minutes and about an hour. The reaction can be followed by measuring the UV spectrum will. It is known that the UV maxima of vitamin B121 'at 473, 405, 348 to 355 (shoulder) and 312 mw.
The new method has the advantage that the vitamin B12 r is more stable Solution obtained, with the further reactions without the use of complicated equipment can be carried out.
From the alcoholic products obtained after the catalytic hydrogenation Vitamin B12I solutions can be combined by precipitating with an organic solution Solvent in which vitamin B12. insoluble ] I is, win. Such Examples of solvents are acetone, ether, ethyl acetate, tetrahydrofuran and dioxane etc. This method of obtaining vitamin B12r has the advantage of being essential can be carried out more quickly than the corresponding isolation of vitamin B12. the end aqueous solution. The process is also gentler, as you work at low temperatures can work.
According to the invention it is thus possible for the first time in a simple Reaction Vitamin B, ". In a stable solution suitable for practical purposes to win and to separate them gently from such solutions.
Example 1 In a test tube with gas inlet and outlet tube 20 mg of vitamin B12 are dissolved in 20 ccm of absolute methanol and 70 mg of 10% Pd carbon offset. A slow stream of hydrogen is passed through the suspension pearls. The red-violet solution turns brown. After 15 to 20 minutes, the Catalyst sucked into a glass frit. The solution thus obtained can be used directly for further experiments related to the vitamin B12. but can also with acetone be precipitated as an amorphous precipitate and centrifuged off. The one with this solution measured UV spectra show maxima at 473, 405, 312.5, 348 to 355 m [(shoulder) and thus correspond exactly to the information in the literature.
Example 2 In a test tube with a gas inlet and outlet tube 50 mg of vitamin Bi are dissolved in 10 cc of absolute benzyl alcohol and 100 mg Pd carbon offset. A slow stream of hydrogen is passed through the suspension pearls. The reduction is complete after 10 to 15 minutes. The catalyst is through A glass frit is sucked off and the brown vitamin B "solution can be used for further experiments be used.
Example 3 A solution of 1.5 g of vitamin B12 in 200 cc of absolute methanol is hydrogenated in the presence of 1.5 g of methanol-moist, anhydrous Raney nickel at room temperature and under normal conditions. The calculated amount of hydrogen is absorbed over the course of 16 hours. The catalyst is filtered off. The brown solution of vitamin B12 obtained in this way can be used for further experiments.
