Verwendung der Tetracycline bei bakteriellen Infektionskrankheiten ist weit verbreitet.
Wegen der einfachen Verabreichungsmöglichkeitwerden die Präparate vorwiegend peroral
zugeführt. Diese Applikationsform ist aber nicht frei von Nachteilen. Durch die
höhe Konzentration im Intestinaltrakt wird auch die physiologisch dort vorkommende
Darmflora geschädigt mit der Möglichkeit des nachfolgenden pathogenen Pilzwachtstums
(Candida albicans). Außerdem vermögen die im Darm vorhandenen Salze, vorwiegend
Calcium- und Magnesiumionen, die Therapeutika unwirksam zu machen und die Resorption
zu verschlechtern. Überdies ist bei gewissen Erkrankungen die Resorption aus dem
Darm a priori gestört und unzureichend. Durch gleichzeitige Verabreichung der Tetracycline
mit Phosphaten, Vitaminen und Antimykotika hat man diese Nachteile nur zum Teil
auszugleichen versucht. Darüber hinaus erfordert die perorale Verabreichung der
Tetracycline zur notwendigen Konstanz eines Blut- und Gewebespiegels eine über den
Tag verteilte regelmäßige Einnahme der Präparate. Das oft undisziplinierte Verhalten
der Patienten macht eine dauernde Kontrolle der Anordnungen notwendig, die aber
bei der breiten Verwendung der Tetracycline besonders bei ambulanten Patienten undurchführbar
ist. Diese Nachteile können durch parenterale Verabreichung abgeschwächt werden.
Die bisher zur Therapie verwendeten Salze der Tetracycline sind leicht wasserlöslich
und werden daher schnell ausgeschieden, so daß sie für eine rationelle parenterale
Verabreichung nicht geeignet sind.. Es war daher anzustreben, mit einer einzigen

bei niedriger Temperatur im Vakuum zu gewinnen ist. Die so erhaltenen neuen Verbindungen
können in wässeriger Lösung, in Suspension oder in Emulsion zur praktischen Anwendung
gelangen. Außer Wasser kann man auch noch andere Lösungsmittel, wie physiologische
Kochsalzlösung und Polyglykole, gebrauchen. Beispiel 1 0,5 g Tetracyclinhydrochlorid
werden in 5 ccm Wasser gelöst und mit einer wässerigen Lösung (5 ccm) von 0,251
g laurinsaurem Natrium zusammengegeben. Das ausgefallene schwerlösliche Tetracyclinsalz
wird von dem überschüssigen Wasser abgetrennt, mit wenig Wasser gewaschen und im
Vakuum vom letzten Wasserrest befreit. Es stellt eine schwach gelblichgefärbte kristalline
Masse dar, die in Wasser schwer löslich ist. F. = 162 bis 165° C, unscharf, unter

von laurinsaurem Tetracyclin noch nach 24 Stunden ein therapeutisch wirksamer Blutspiegel
gemessen werden. Ein Vergleich der Depotwirksamkeit des nach der französischen Patentschrift
1098 974 hergestellten Tetracyclinpalmitinsäureesters z. B. mit dem Tetracy
clinlaurinsäuresalz zeigt den überlegenen. Depoteffekt der erfindungsgemäß hergestellten
Verbindungen: 10 ing jedes Präparates werden in 10 ccm Phosphatpuffer bei einem
pg-Wert von 7 suspendiert und die nach 3 Stunden an. die Lösung abgegebene Menge
Tetracyclin mittels des Plattenverfahrens biologisch durch Messung der Keimhemmhöfe
bestimmt. Der bessere Depoteffekt der erfindungsgemäß hergestellten Verbindungen
ergibt sich aus der geringeren Antibiotikumabgabe an die Lösung, nämlich 55 yo/o
