Description
As is well known, the blood that accumulates in the slaughterhouses can be reduced by adding citrates, Phosphates, etc. are stabilized, and the plasma is separated from the blood cells in many cases. A more or less yellow-red colored liquid is obtained, which essentially consists of the dissolved proteins, some red blood pigment and the dissolved salts. For pharmaceutical, The plasma is generally not suitable for cosmetic or nutritional purposes in this state use. In addition to considerable impurities, it also contains its fibrin content, which despite the previous stabilization often significantly interferes with further processing.
A gentle removal of the impurities as well as the removal of the gelatinization are desired for further processing. It has surprisingly been found that by the treatment of the plasma with montmorillonites and all unwanted impurities in simple Wise removed from the plasma as well as the protein substances that cause gelatinization can be. For this purpose, it is expedient to bring the plasma with the fuller's earth for such a long time in contact until the proteins mentioned are deposited on it and, at the same time, those in the plasma carry away existing impurities. Because of the changing blood composition, the amount will of the montmorillonite to be used is best determined by a preliminary test; in general one needs about 5% of the plasma weight of montmorillonite. It is also useful to use the Select the montmorillonite to be used on a case-by-case basis by means of samples.
I use untreated, neutral montmorillonites best for this purpose. The plasma is after this treatment a clear, somewhat viscous liquid that no longer tends to gelatinize.
According to a further embodiment of the invention, the cleaning can be improved by a second treatment with activated charcoal follows the pretreatment with montmorillonite described. It is expedient to place montmorillonite in front of this, too, and only then add the activated charcoal to the plasma. Here, acid-digested montmorillonites have proven particularly useful. the The amount of activated carbon and fuller's earth added depends on the degree of cleaning required.

Heat until boiling does not coagulate. This shift! the salt balance can, for. B. by slowly adding acids or alkalis. Is particularly suitable according to one further development of the invention, the shift of the salt equilibrium in the plasma by ion exchangers.
For scientific purposes, blood plasma has already been treated with cation exchangers in the Na + or H + form. In the first case a plasma was obtained which only contained sodium as a cation, in the other case the respective dissolved salts were converted into the free acids. The subsequent treatment with anion exchangers has so far failed because the protein bodies immediately flocculated when the acidic plasma came into contact with anion exchangers. The blood proteins fall upon reaching a p H -value of about 5 and does not dissolve again until reaching a p H -value of 7, obtained in this case positively. It was therefore not possible to achieve desalination of the plasma by successive treatment with cation and anion exchangers. The use of mixed-bed ion exchangers also did not achieve the goal for the same reason.
It has now been found that a particularly pure, almost colorless and ion-free plasma is produced

Wegen der oben geschilderten Schwierigkeiten geht man hierzu erfindungsgemäß folgendermaßen vor:To produce the desalinated plasma, it can first be treated with anion exchangers. This gives a plasma with a strongly alkaline reaction, which is subject to the usual discoloration and then brought into contact with a cation exchanger. The cation exchanger now binds the free cations, creating a plasma that is almost neutral and not at all Shows signs of flocculation. You get a better product if you first remove the cations from the Plasma removed. Because of the difficulties outlined above, this is done according to the invention do the following:
The appropriately pre-cleaned blood plasma is treated with a strongly acidic cation exchanger freed from the dissolved cations (Na, Ca, Li, Fe) in the H + form. Here are formed the equivalent amounts of the free acids concerned, d. i.e., the reaction of the plasma is after Treatment strongly acidic.
Treatment with the cation exchanger means that the plasma is largely insensitive to flocculation become, e.g. B. it can be heated to temperatures of up to 100 ° C without hesitation.
By titration of a small sample of this plasma, it is found now, how much NaOH sufficient to bring this sample of p H 7.0 to the point where the proteins are clearly redissolved. This point is according to experience at p H values from 8.0 to 10.5. If one sets the acidic blood plasma as much of a soluble alkali metal salt or base to that resulting from the salt Alkalihydroxydmenge sufficient to bring the p H of the plasma after removal of the anions to the desired value. It does not matter in which form the alkali salt is added to the acidic plasma, e.g. B. as a pure salt, as a solution, as a non or only partially desalinated plasma, etc. It is essential that the plasma has a certain alkali content before treatment with the anion exchanger. The anion run is now carried out in such a way that the plasma is led into a column of a strongly basic anion exchanger in the OH ~ form and by moving the exchange it is ensured that the initially cheesy protein does not clog the flow. On reaching the above-mentioned p H values then, the flocculated protein released again; Completely. The movement can e.g. B. be effected by a stirrer or by introducing air. The plasma flowing out of the column, which is completely clear, contains only small traces of free alkali and can be used for most purposes. These small amounts of alkali can also be bound to the plasma by adding a cation exchanger without the protein bodies flocculating. A plasma produced in this way does not contain any dissolved salts and is at the same time largely stable to coagulation. It is almost colorless, clear, odorless and tasteless.
For some purposes it is advantageous to have a decolorized plasma. The plasma, e.g. B. decolorized in such a way that, after adding 0.05 to IVoH 2 O 2, it is heated on a water bath until no further lightening of the color can be detected. Mart can also perform the decolorization of the plasma after the described anion run, but it is generally more convenient to perform the decolorization between the cation and anion run.

Das ablaufende Plasma hat einen pH-Wert von 1,75.Through a column of a strongly acidic cation exchanger in the H + form 25 1 of this pre-purified plasma are added with a pH value of 7.9. The column has a diameter of 68 mm and a layer height of 660 mm, the volume of the cation exchanger is 2 liters and the flow rate is 1.38 m / h. The outflowing plasma has a pH value of 1.75.
B. durch Eindampfen konzentriert, durch ein Sprühverfahren getrocknet bzw. durch Tiefkühlung konserviert werden.4 l of this plasma are mixed with 8 g of NaCl and passed through a column of a strongly basic anion exchanger in the OH ~ form of the same dimensions from bottom to top. With the same flow speed, the entire filter filling is moved by a grid stirrer which runs at about 60 rpm. In the lower third of the column, a white precipitate forms immediately, but it disappears more and more towards the top. The outflowing clear plasma is collected and has a pH value of 8.45 and contains 0.03% Na. This plasma is stirred with 21 cm 3 of a strongly acidic cation exchanger in the H + form and separated from it again after 5 minutes. It has a pH value of 8.2. The plasma is sterilized by heating it to 95 ° C. for one hour. The plasma can now be further processed by known methods, e.g. B. concentrated by evaporation, dried by a spray process or preserved by deep freezing.
