Dabei wird diese Substanz zur Diagnose von Erkrankungen, die mit einer gestörten Regulation der Apoptose im Zusammenhang stehen, genutzt. Hierbei kann es sich z. B. um die Erkennung von genetischen Dispositionen für bestimmte Erkrankungen infolge von Mutationen, z. B. am Kationenkanal, handeln. Es kann so unter Umständen eine Anfälligkeit gegenüber bestimmten Erkrankungen diagnostiziert werden, so daß bei entsprechenden Symptomen schnell auf eine gestörte Regulation der Apoptose, mit den damit einhergehenden klinischen Symptomen, geschlossen werden kann. Bei der Substanz, die zum Nachweis der Expression und/oder Funktion von Kationenkanälen eingesetzt wird, kann es sich z. B. um einen Antikörper handeln, der gegen den Kationenkanal gerichtet ist, und in einem dem Fachmann bekannten Nachweisverfahren, wie z. B. ELISA (enzyme linked immuno sorbent assay) eingesetzt wird. Weiterhin kann z. B. mit Hilfe der sogenannten PCR-Technik (polymerase chain reaction) dieser Nachweis erbracht werden. Bei diesem molekulargenetischen Verfahren werden selektiv bestimmte DNA- Abschnitte amplifiziert. Dabei erfolgt eine Neusynthese von DNA- Sequenzen, die von zwei synthetischen Oligonukleotiden eingerahmt werden, mittels DNA-Polymerasen. Durch die exponentielle Anreicherung, ausgehend von geringen Mengen DNA (10-9-10-15 g) können nach mehrmaliger Wiederholung des Vorgangs (20-40 Zyklen) die DNA-Abschnitte nachweisbar gemacht werden. Zum Nachweis von RNA-Abschnitten muß die RNA mittels einer RNA-abhängigen DNA-Polymerase in eine DNA umkopiert werden. Durch Auswahl von geeigneten DNA- Abschnitten am Zielprotein, dem Kationenkanal, kann so der erfindungsgemäße Nachweis erfolgen. Weitere Verfahren, wie ein bekanntes Zielprotein bzw. Domänen eines aus mehreren Untereinheiten zusammengesetzten Zielproteins quantitativ und/oder qualitativ nachgewiesen werden kann, sind dem Fachmann bekannt, und in diesem Zusammenhang wird auf die umfangreiche dementsprechende Fachliteratur verwiesen. In weiteren Experimenten konnte gezeigt werden, daß der Kationenkanal- Hemmstoff Amilorid die osmotisch, oxidativ oder metabolisch induzierte Apoptose unterbinden kann. Amilorid hemmt einen Kationenkanal, der durch osmotischen Schock [Huber et al. 2001] und oxidativen Streß [Duranton et al. 2001] aktiviert wird und Ca2+ in die Zelle einströmen läßt [Duranton et al. 2001]. Die Hemmung der erythrozytären Apoptose durch Amilorid zeigt daher, daß die Apoptose durch Aktivierung dieses Kanals zustande kommt. Befunde in kernhaltigen Zellen zeigen, daß auch diese Zellen durch Aktivierung des Amilorid-sensitiven Kationenkanales getötet werden können. Studies have been conducted to investigate whether osmotic shock or oxidative stress are well-known triggers of apoptotic cell death in nucleated cells [Bortner & Cidlowski 1998, 1999, Lang et al. 1998, Maeno et al. 2000, Michea et al. 2000, Roger et al. 1999, Rosette &, Karin 1996] also drive erythrocytes into apoptosis. It could be shown that both measures and glucose deficiency trigger apoptosis. Surprisingly, apoptosis was prevented by removing the extracellular Ca 2+ . In further experiments it could be shown that the cation channel inhibitor amiloride can suppress the osmotic, oxidative or metabolically induced apoptosis. Amiloride inhibits a cation channel that is caused by osmotic shock [Huber et al. 2001] and oxidative stress [Duranton et al. 2001] is activated and Ca 2+ flows into the cell [Duranton et al. 2001]. The inhibition of erythrocytic apoptosis by amiloride therefore shows that apoptosis is caused by activation of this channel. Findings in nucleated cells show that these cells can also be killed by activating the amiloride-sensitive cation channel.

Um die Bedeutung von Ca2+ für die erythrozytäre Apoptose zu überprüfen, wurde das Calciumionophor Ionomycin eingesetzt. Wie in Abb. 1 B gezeigt wird, führt die Verabreichung von 1 µM Ionomycin binnen 3 Stunden zu massiver, Ca2+-abhängiger Zeltschrumpfung, wie anhand der "Vorwärtsstreuung" (forward scatter FC) der Fluoreszenz-aktivierten Zell Sortierung (FACS) gezeigt werden kann. Die FC nimmt von einem Wert von 428 ± 13 (n = 3) auf einen Wert von 121 ± 2 (n = 4) ab. Darüber hinaus steigert 1 µM Ionomycin die Bindung von Annexin von 4.1 ± 0.4% (n = 3) auf 71.3 ± 3.2% (n = 4) (Fig. 1 D). Ionomycin induzierte Zellschrumpfung und Annexinbindung werden beide in Abwesenheit von extrazellulärem Ca2+ gehemmt. Osmotischer Schock ist als Auslöser von Apoptose in kernhaltigen Zellen bekannt [Bortner & Cidlowski 1998, 1999, Lang et al. 1998, Maeno et al. 2000, Michea et al. 2000, Roger et al. 1999, Rosette &, Karin 1996]. Wie überraschenderweise beobachtet wurde, steigert osmotischer Schock die Annexinbindung auch von kernlosen Erythrozyten (Abb. 2B, E). Werden Erythrozyten einer Osmolarität von 850 mosmol/l ausgesetzt (Zugabe von Sucrose), dann steigt die Zahl Annexin-bindender Zellen binnen 24 Stunden von 3.9 ± 0.3% (n = 4) auf 51.4 ± 3.8% (n = 9). Ein typisches Experiment ist in Abb. 2B dargestellt. Nominale Abwesenheit von Calcium mindert die Zahl Annexin-bindender Zellen nach osmotischem Schock signifikant auf 21.1 ± 7.2% (n = 4, siehe Abb. 2C für ein typisches Experiment und Abb. 2F für arithmetische Mittelwerte ± SEM). Oxidativer Streß, hervorgerufen durch die Verabreichung von 0.66 mM oder 1 mM tert-butyl-hydroperoxide (tBOOH) führte zu massiver Schrumpfung von Erythrozyten, wie durch die Änderung der Vorwärtsstreuung (forward scatter) in der FACS-Analyse von 370 ± 13 (n = 4) auf 314 ± 51 (n = 4) bzw. 167 ± 14 (n = 4) erkennbar wird. In einer ersten Serie von Experimenten konnte beobachtet werden, daß eine Behandlung der Zellen für 15 min mit 0.66 mM tBOOH signifikante Annexin-Bindung auslöst (Fig. 3B). Die Annexinbindung konnte durch Weglassen des extrazellulären Calciums (Abb. 3C) oder durch Verabreichung von 1 mM Amilorid (Abb. 3D) signifikant reduziert werden. Wie in Abb. 3E gezeigt wird, steigern 0.66 mM und 1 mM tBOOH die Zahl Annexin-bindender Zellen von 3.2 ± 0.5% (n = 4) auf 26 ± 7.5% (n = 4) und 68.5 ± 5.3 (n = 4). In der nominalen Abwesenheit extrazellulären Calciums sind sowohl die Oxidationsinduzierte Zellschrumpfung, als auch die Annexin- Bindung beide herabgesetzt. Die Werte für die Vorwärtsstreuung (forward scatter) in Calciumfreiem Incubationsmedium erreichten 339 ± 22 (n = 4) (0.66 mM tBOOH) und 307 ± 15 (n = 4) (1 mM tBOOH), im Vergleich zu 314 ± 51 (n = 4) und 167 ± 14 (n = 4) in der Anwesenheit von Calcium. Die Zahlen für Annexin-positive Zellen erreichten nur 11.0 ± 1.4% (n = 4) (0.66 mM tBOOH) und 34.1 ± 2.6% (n = 4) (1 mM tBOOH) in der Abwesenheit von Calcium (Fig. 3E). Entsprechend hemmte das Weglassen von extrazellulärem Calcium die tBOOH-induzierte Phosphatidylserin-Umlagerung um 66% bzw. 53%. Gleichermaßen minderte die Anwesenheit von 1 mM Amilorid die Wirkung von tBOOH selbst in der Anwesenheit von 1 mM Ca2+. Die jeweiligen Werte für die Vorwärtsstreuung (forward scatter) erreichten 356 ± 24.0 (n = 4) (0.66 mM tBOOH) und 291 ± 27 (n = 4) (1 mM tBOOH). In der Anwesenheit von 1 mM Amilorid wurde die Zahl Annexin-positiver Zellen auf 7.9 ± 3.2% (0.66 mM tBOOH) bzw. 37.7 ± 4.4% (1 mM tBOOH) herabgesetzt, ein Befund, welcher die Hemmung der t-BOOH-induzierten Annexin-Bindung um 80 bzw. 48% reflektiert (Abb. 3E). Es wurde früher gezeigt, daß Glucosemangel die antioxidativen Mechanismen von Erythrozyten schwächt. Daher wurde die Wirkung von Glucosemangel untersucht (Abb. 4A-E). In der Anwesenheit von Glucose banden 3.1 ± 0.6% (n = 4) der Erythrozyten Annexin. Die Entfernung von Glucose aus dem extrazellulären Medium steigerte die Zahl Annexinbindender Zellen auf 11.2 ± 2.2% (n = 4) nach 24 Stunden und auf 49.4 ± 5.7% (n = 4) nach 48 Stunden. Die Zunahme Annexin-bindender Zellen war in der nominalen Abwesenheit von Calcium signifikant herabgesetzt. Die entsprechenden Werte waren 10.4 ± 2.5% (n = 4) nach 24 Stunden und 10.0 ± 1.9% (n = 4) nach 48 Stunden. In gleicher Weise wurde die Wirkung von Glucose-Entzug durch Amilorid (1 mM) abgeschwächt. Die jeweiligen Werte waren 5.7 ± 1.8% (n = 4) nach 24 Stunden und 11.7 ± 1.9% (n = 4) nach 48 Stunden (Abb. 4E). The increase in annexin-binding cells was significantly reduced in the nominal absence of calcium. The corresponding values were 10.4 ± 2.5% (n = 4) after 24 hours and 10.0 ± 1.9% (n = 4) after 48 hours. In the same way, the effect of glucose withdrawal was weakened by amiloride (1 mM). The respective values were 5.7 ± 1.8% (n = 4) after 24 hours and 11.7 ± 1.9% (n = 4) after 48 hours ( Fig. 4E).
Jüngste Befunde zeigten, daß der Ca2+-Ionophor Ionomycin erythrozytäre Zellschrumpfung und Annexinbindung von Erythrozyten auslöst, beides typische Hinweise auf Apoptose in kernhaltigen Zellen [Bratosin et al. 2001, Berg et al. 2001, Daugas et al. 2001]. Wie und unter welchen Bedingungen die Ca2+-Konzentration in Erythrozyten gesteigert wird, blieb bisher unklar. Mit den vorliegenden Ergebnissen ist zum ersten Mal die Bedeutung eines Amilorid-sensitiven, Zellvolumen-regulierten Kationenkanales für die Auslösung von apoptotischem Zelltod gezeigt worden. Die Kanäle wurden früher charakterisiert, ihre Hemmbarken durch Amilorid [Huber et al. 2001, Duranton et al. 2002], sowie ihre Empfindlichkeit gegenüber osmotischem [Huber et al. 2001] und oxidativem [Duranton et al. 2002] Streß gezeigt. Recent findings have shown that the Ca 2+ ionophore ionomycin triggers erythrocytic cell shrinkage and annexin binding of erythrocytes, both typical indications of apoptosis in nucleated cells [Bratosin et al. 2001, Berg et al. 2001, Daugas et al. 2001]. Durch Hemmung der Kationenkanäle wird dieses Absterben verzögert.
Ähnliche Befunde lassen sich auch in kernhaltigen Zellen nachweisen.
